Regions differing in degree of mineralization were dissected from developing bovine molar teeth and the demineralized matrix proteins subjected to amino acid analysis, gel exclusion chromatography and polyacrylamide gel electrophoresis. The results demonstrate a striking reduction in the proportion of a protein complex of some 30,000 daltons molecular weight and an increasing proportion of smaller polypeptides with higher degree of mineralization within individual teeth. It is proposed that progressive degradation of the larger amelogenins yields increasing proportions of the smaller (J-group) polypeptides.

Adult human normal enamel was extracted with EDTA under dissociative conditions, giving a soluble protein fraction and an insoluble residue. This residue, approximately 10–20% of the original sample mass, contained enamel hydroxyapatite associated with small amounts of matrix protein. The latter only became released after dissolution in lactic acid. Both the EDTA-soluble and the lactic acid-soluble adult enamel proteins were glycoproteins small in size (2,000–3,000 daltons on dissociative gel filtration chromatography) and similar in amino acid composition. These two protein fractions did differ significantly, however, in their organic phosphorus contents, the lactic acid-soluble protein containing higher levels of this substituent. It is postulated that the EDTA dissolution resistance of the hydroxyapatite crystals contained in this insoluble residue resulted from a strong association of the enamel apatite with the phosphorylated adult enamel matrix glycoprotein constitu
