In this paper the in vitro remineralization of human dentine with 0, 0.5, 2 and 10 ppm F in the remineralization solution is presented and analyzed by microradiography. Furthermore, the remineralization data of human and bovine dentine are quantitatively compared. The main results of this paper are: (1) the efficacy of human (and bovine) remineralization is about proportional to the square root of the F level in the rem-ineralizing solution, and (2) the amount of mineral effectively deposited in the dentine is most likely controlled by the diffusion of fluoride into the tissue. This study shows furthermore that although numerical differences exist in the remineralization of human and bovine dentine, the general remineralization behaviour is quite similar. The presence of F–– in the parts per million range is essential for the remineralization efficacy in vitro and also for the ‘overremineralization’ of the outer dentine surface.

The fluorescence intensity of the dye Fluorol® 7GA in in vitro caries lesions was measured through a strip of plastic either 2 or 1.5 mm in thickness. The light scattering in the plastic strips was comparable with light scattering in sound enamel. This method was chosen as a model for fluorescent dye measurement at approximal sites, where the fibreoptic probe cannot be placed directly on the lesion, so that measurements have to be made through a thin layer of sound enamel. It appeared that the fluorescent dye can be measured through a layer of scattering material and that a linear correlation exists between fluorescence intensity and calcium loss as measured by longitudinal microradiography. The correlation coefficient for the 2-mm plastic strip was r = 0.76. With the 1.5-mm plastic strip the fluorescence intensity recordings were about twice as high, and the correlation coefficient was r = 0.81. From these measurements a proposal for an application procedure emerged for the use in a clinical situation. Also the transmission of light at 550 and 650 nm, at which wavelengths the fluorescent dye does not absorb, through plastic strips of varying thickness was measured. It is shown that by measurement of transmission of a non-adsorbing wavelength, the fluorescence intensity measured through a layer of scattering material can be corrected for this layer.

The purpose of this study was to investigate in vivo enamel fluoride uptake of a slow-release boron trifluoride BIS-GMA resin material. Study subjects were orthodontic patients with at least one pair of permanent bicuspid teeth indicated for extraction. The material was applied to the buccal surface of the test tooth 1 month prior to extraction; the contralateral tooth served as the control tooth. Following extraction, the resin was removed by soaking in acetone for 36 h and polishing with a sodium bicarbonate slurry. Prior scanning electron microscopic studies have shown that this cleaning procedure effectively removes all residual resin. Enamel fluoride analyses were completed for 12 pairs of teeth from 9 subjects. The mean differences in enamel fluoride concentrations between the treated and control teeth were significantly different from zero at the 0.01 level. At each successive depth, the absolute mean amount of fluoride uptake by the test teeth was fairly constant; however, the proportional mean amount or percent increase in fluoride concentration resulting from treatment became greater in the deeper enamel layers. The data suggest that this material has the potential to provide an effective means of introducing substantial amounts of permanently bound fluoride into surface enamel, even into the deeper layers.

The effects of hyposalivation on the induction of dental caries were examined in specific pathogen-free Sprague-Dawley rats infected with Streptococcus mutans MT8148R and fed the caries-inducing diet 2000. Hyposalivation was induced by either irradiation or surgical removal of salivary glands in 30-day-old rats. After either procedure, severe dental caries was induced in the rats when examined at 84 days of age, and a statistically significant correlation was found between caries scores and flow rates of saliva. However, the recovery of strain MT8148R from hyposalivated rats was not always high, and it is possible that microorganisms other than the inoculated S. mutans strain MT8148R may have played a role in promoting dental caries.
