SUMMARYOne hundred and six patients with chronic dermatophytosis unresponsive to griseofulvin were studied.Trichophyton rubrumwas the causative organism in 93% of cases. Chronic dermatophyte infections caused by organisms other thanT. rubrumoccurred predominantly, but not exclusively, in patients with underlying diseases. Forty‐nine percent of the patients had a personal or family history of atopy but other abnormalities included disorders of keratinization, collagen vascular disease and systemic steroid therapy. The commonest sites of infection were the palms and soles, as opposed to toe webs or groins in control patients with griseofulvin‐res‐ponsive dermatophyte infections. SUMMARYFive patients with reticular erythematous mucinosis were studied by skin biopsy and phototesting. Light microscopy showed a dermal mononuclear cell infiltrate (composed largely of lymphocytes), separation of collagen bundles and fragmentation of elastic fibres. Histochemical stains showed increased dermal mucin with a profile consistent with hyaluronic acid. In one case, the ultrastructural distribution of mucin was demonstrated using colloidal iron staining, electron microscopy and X‐ray microanalysis. SUMMARYSixteen years ago, an area on the forearm of a freckled subject was painted with a solution containing 500 esu of thorium‐X per ml. Since then, despite much exposure to solar irradiation, there has been a complete absence of freckles from this area, which stands out in marked contrast to the surrounding skin. Biopsy of the skin from the area was examined by the Dopa technique and by electron microscopy. Melanocytes are present in apparently normal numbers, but in a condition of almost complete inactivity. SUMMARYEpidermal sunburn (SB) cells were counted 24 h after exposure of the skin to suprathreshold doses of solar simulated radiation. SUMMARYWe measured free and total 5‐S‐cystcinyldopa excretion in 24‐h urine specimens from nine patients with malignant melanoma and forty‐five controls, and found that the levels of 5‐S‐cysteinyldopa excretion (measured by a fluorometric method) were greatly increased in the melanoma patients. SUMMARYStratum corneum was removed from the surface of pig skin using a wet brushing technique. The material so obtained was fractionated into cell clumps, discrete cells and fine material by differential nitration through nylon gauzes. Ceramide, cholesterol and free fatty acid accounted for about 95% of the lipid present in each fraction: phospholipid and glycolipid were not detected. Material removed by successive brushings showed no detectable differences in lipid composition.

SUMMARYAdult human skin was maintained in organ culture in a chemically defined medium without serum for up to 10 days. Under these conditions cultured epidermis revealed ultrastructural changes suggesting alterations in keratinization. Half of the newly formed stratum corneum became parakeratotic. The number and size of keratohyalin granules were reduced, and only few keratinosomes were present after the 5th day of culture.The effect of hydrocortisone at final concentrations of 10−5–10−9mol/1 was studied by applying stereological techniques for electron microscopic morphometry on systematically sampled tissues. The volume densities of tissue components and cell organelles were assessed at three different strata of epidermis on the 5th day of culture. Changes of keratinization due to the culture conditions were partly reversed by the hormone, i.e. hydrocortisone increased the volume densities of keratohyalin granules, keratinosomes and tonofilaments.
