Rabbit xenoantisera against membranes of mouse spleen cells and cultured human lymphocytes were used to monitor the purification of solubilized mouse H–2 and human HL–A antigens in parallel with routine serological methods employing alloantisera. Antigenic material was subjected to gel filtration, ion exchange chromatography and electrophoresis in polyacrylamide gel, and in all cases the only material which inhibited the lymphocytotoxic action of the xenoantisera was that which inhibited defined alloantisera. Conversely, all alloantigenic material inhibited xenoantisera. Highly purified H–2 and HL–A preparations completely inhibited the lymphocytotoxic action of xenoantisera to crude membrane antigen. The specificity of the anti‐mouse xenoantisera was examined. Antibodies specific for the immunizing mouse strains were revealed only after so‐called species‐specific antibodies were removed by judicious absorption of the serum with cells of a mouse of another H–2 type. The residual strain‐discriminating antibodies reacted with target cells from a large panel of different mouse strains with an intensity directly proportional to the number of reacting H–2 specificities. It was not possible, however, to detect monospecific H–2 antibodies in any absorbed sera It is concluded that the lymphocytotoxic antibodies in xenoantisera predominantly are directed against determinants intimately associated with H–

The plasma samples of 11 out of 16 cytotoxicity‐negative hemodialysis patients were found to suppress 43% of 122 mixed leukocyte culture (MLC) combinations. An almost linear relationship was found between the number of transfusions and the blocking activity of the plasma. Plasma from parous women without cytotoxins suppressed 23% of 241 MLC combinations. With plasma from normal controls, only 2.8% of 140 MLC combinations tested were suppressed. The suppressive activity was found to be specific (though somewhat broad) and was absorbable from plasma by the stimulating cells. Inhibition could not be obtained by coating of the responding cells, although a weak degree of suppression was found by precoating the stimulating cells. If suppression of MLC can be explained by the presence of enhancing antibody, active production of such antibodies can be induced by transfusions. Unresponsiveness to incompatible transplants could then be attributed to preconditioning by transfusion

Forty‐eight patients with carcinoma of the breast and 122 controls from the Boston area were typed for 24 HL–A specificities. Types were assigned objectively by a computer program, and two controls were matched to each of 40 breast cancer patients on the basis of the countries of origin of the patients’ ancestors or race. Our data fail to confirm an association of HL–A7 with breast cancer suggested by the data of Patel and coworkers (1972) who typed 52 patients from the Boston area. The effects of alloantisera on lymphocyte proliferation were studied in 45 multipara. Lymphocytes from mother (M), father (F) and unrelated person (U) were typed for HL–A specificities and cultured in bilateral and unilateral MLC comparatively in mother's and father's sera. Statistical analysis of these paired cultures demonstrated an inhibitory effect of mother's sera in 16 cases. Blocking factors were directed against stimulator as well as against responding lymphocytes. In most cases this effect was observed in the absence of detectable cytotoxic antibodies. PHA response was but slightly affected. The blocking effect was often directed against unrelated lymphocytes, which did not share evident HL–A specificities with the father. A method of isolating and concentrating monospecific antibody from multispecific HL–A antiserum by adsorption with and elution from platelets is described. One unit of platelets containing approximately 1 × 1011platelets is sufficient to absorb one unit (300 ml) of serum of low titer. Antibodies directed against antigens determined at separate loci are readily separated by adsorption with appropriately selected platelets, whereas antibody specificities directed against antigens determined by the same locus are usually not separable.
