Human antigenic determinants present on activated T and B lymphocytes and not on resting B and T cells defined by serological studies are reported. The reactivity of 13 sera against human PBM activated by various agents was tested using microlymphocytotoxicity technique. Results showed that generally similar reactions are present on B and T blasts whatever the technique of activation (PHA, ConA, PWM, TCGF, EBV and allo activation). Absorption elution studies with 5 sera on 10 alloactivated cells clearly demonstrated their independence from DR determinants. The analysis of results obtained by these 15 sera on blasts from 50 donors showed no correlation with HLA‐A, ‐B, ‐C, DR and ‐MB antigens, neither can it be HT (Qa‐like) antigens. Furthermore, 5 clusters of sera are reported. The weak correlation coefficients cannot affirm the existence of a system with an allelic distribution although each cell except one out of a panel of 50 individuals carries at most 2 specificities. Balb/c mice were immunized with a human B‐lymphoblastoid cell line typed HLA‐A3, B7. The splenocytes of the immunized mice were fused with a murine myeloma. Supernatants of the cultures were screened against the immunizing cell line in fluorochromasia. Positive cultures were expanded and cloned. One of the clones, X 15.4, was expanded and brought to ascites in Balb/c mice. Monoclonality of the antibody X 15.4, which belongs to the class IgM and immunoprecipites a molecule of 44000 daltons, was demonstrated by isoelectric focusing. By complement dependent citotoxicity the ascites only reacted with the lymphocytes of all HLA‐A3 individuals from a panel of 146 donors, showing no crossreactions. Twenty‐one Israeli Jewish pemphigus vulgaris (PV) patients were studied for the HLA‐D lymphocyte defined determinants and the serologically defined antigens of the HLA‐A, B, and DR series. HLA‐D typing revealed that Dw10 is significantly associated with PV: 86% of patients vs 18% of controls carried Dw10, and DR4 was present in 86% of patients as compared to 38% in the controls. The most striking observation was that all Dw10 positive patients were also positive for DR4, and no other patient carried DR4 alone. The relative risk for a Dw10‐DR4 carrier to develop PV was estimated at 31.9, higher than that observed for Dw10 alone (RR 26.7) or DR4 alone (RR 9.6).Probably HLA‐Dw10 predisposes for pemphig

The frequencies of the HLA‐A, B, C, D, DR and MB antigens have been determined in a homogeneous Shanghai Chinese population. Comparisons with the HLA‐A and ‐B frequencies in other subsets of the Chinese population revealed some marked differences. No comparisons were possible for the D, DR and MB antigens since there were no previous studies of the antigens in those loci.We suggest that studies of the Chinese population should be confined to clearly defined homogeneous subsets. In this manner, the confounding effect of population heterogeneity may be avoided, and it is this heterogeneity which calls for extensive surveys of the huge Chinese popul

A search for antigens of the HLA system has been carried out in 53 Mataco Indians of Argentina living in a geographically isolated area in the northeast of the country. Samples were mostly collected from adults of both sexes who were not directly related.Lymphocyte typing was performed using the microcytotoxicity technique of NIH. 118 sera specific for 15 antigens of the first HLA locus, 22 antigens of the second and 6 of the third were used.The most frequently found alleles were HLA‐A28, Aw31 and A2 for the first locus; B15 and B40 for the second; and Cw3 and Cw4 for the third.In addition to previously published investigations on South American Indians, our typing work shows a remarkable homogeneous gene pool and a restricted range of polymorphism; therefore, a further set of haplotypes rendered us also restricted. The most frequent haplotypes that showed a significant statistical linkage desequilibrium were: A2‐Cw4, A28‐Bx, A2‐Cw3, Aw31‐Bw16, Aw24‐Cw3, B15‐Cw3, Bw16‐Cw3 and A28‐B5.Some of these haplotypes have also been found in other i

The serological typing of determinants within the guinea pig histocompatibility complex (GPLA‐complex) has, until now, largely been carried out by the51Cr‐release technique. As target cells, a mixture of B and T cells from peripheral blood and lymph nodes were used. However, for the detection of Ia‐determinants, which predominantly are expressed on the surface of B cells, the use of lymphocyte suspensions with a relatively low content of B cells can cause some evaluation problems.We have developed a simple method for obtaining a relatively pure suspension of B cells, making use of the ability of guinea pig T cells to form rosettes with rabbit red blood cells and the possibility of removing these by sedimentation on lymphoflot. A new antigenic determinant defined by a monoclonal antibody (6.7) is described which displays restricted expression being present only on the cells of the immunizing donor and his father and not on a panel of over one hundred unrelated HLA‐typed cells. To evaluate further the feasibility of HLA typing for prenatal diagnosis, we tested human amniotic fluid cells (AFC), known to express HLA‐A, ‐B, and ‐C antigens, for the presence of HLA‐DR antigens using type‐specific antisera in the microcytotoxicity assay and a monoclonal antibody directed against the common HLA‐DR structure (cDR) in indirect immunofluorescence. Prenatal typing of HLA‐DR on AFC in the microcytotoxicity test was possible in only one out of eight families studied. The detected DR2 antigen was confirmed by postnatal typings of cord blood lymphocytes. Thereafter, 23 different AFC cultures were tested with monoclonal antibodies in indirect immunofluorescence. Only six cultures were partially positive (23–35% fluorescent cells) with the monoclonal cDR antibody while all AFC cultures demonstrated strong positive fluorescence (68–100%) with a monoclonal antibody against the common HLA‐A, ‐B, and ‐C structure (cHLA). These data suggest that only a small subpopulation of AFC expresses class II (HLA‐DR) antigens in contrast to the nearly ubiquitous expression of class I (HLA‐A, ‐B, and ‐C) antigens. Furthermore, the heterogeneous expression of cell surface antigens within the various AFC cultures was substantiated with monoclonal antibodies directed towards cell surface antigens of the OKT, OKM, and Lyt series that have been found to be characteristic for subpopulations of lymphoid and hemapoetic cells.
