Synthetic activity of collagen types was examined in cultured arterial smooth muscle cells during modulation from synthetic to contractile phenotype by treatment with dimethyl sulfoxide (DMSO). Smooth muscle cells of rabbit thoracic aorta cultured with a 1% supplement of DMSO for 8 days (DMSO group) predominantly exhibited cellular features of the contractile type with abundant microfilaments and a distinct basement membrane. Cultured cells in the DMSO group or in controls during stationary or subconfluent phase were labeled with [3H]proline for 24 h, and the samples including the cell layer and medium were analyzed. The incorporation of proline into bacterial collagenase digestible fractions was increased slightly in the DMSO group. Type analysis of the collagenous protein by SDS PAGE and subsequent fluorography disclosed a markedly increased ratio of type IV/I collagen and a slightly increased type V/l collagen ratio, as compared with those of controls. A decrease of type III collagen production in DMSO treated cells probably due to their lower cell density was also recognized. From these biochemical and morphological observations, it is suggested that increased synthesis of minor collagen types, particularly type IV collagen, is closely associated with smooth muscle phenotypic expression following DMSO treatment. Similar cellular events may occur in smooth muscle cells migrating into the intima during the process of arteriosclerosisin vivo.

We developed a competitive enzyme immunoassay for serum type IV collagen peptide as a marker of fibrogenesis, and examined the relationship between serum type IV collagen peptide and hepatic disorder in CCI, treated rats. The rats were treated for 8 weeks and signs of liver damage began to appear from about week 2. With the progression of these signs to liver fibrosis, type IV collagen increased in the fibrous septa and especially in the perisinusoidal walls, where the increase was manifested as development of a real basement membrane beneath the sinusoidal endothelial cells. In CCI4‐treated rats, serum type IV collagen peptide significantly increased with the progression of liver fibrosis. When CCI4administration was stopped, the collagen peptide rapidly decreased without any rebound rise. An intimate relationship was found between the production of serum type IV collagen peptide and liver prolyl hydroxylase activity and the amount of collagen deposited in the liver. Lectin binding in diseased murine glomeruli was studied in MRL 1 mice, using seven different fluorescence‐ or peroxidase coupled lectins:Griffonia simplicifoliaI (GS‐I),Ulex europaeusagglutinin I (UEA‐I),Ricinus communisagglutinin I (RCA I), wheat germ agglutinin (WGA), concanavalin A (Con A), peanut agglutinin (PNA), andHelix pomatiaagglutinin (HPA). Lectin binding in diseased glomeruli of MRL 1 mice was different from that in normal glomeruli. Light and fluorescence microscopy showed that: 1. in mesangial proliferative lesions, the binding of RCA I, WGA and Con A increased and that of GS I and PNA appeared in the mesangium; 2. in other glomerular lesions, UEA‐I bindng appeared and RCA I stained the altered membranes irregularly. Electon microscopy showed that: 1. GS‐I stained the endothelial cell coat and the glomerular basement membrane covered by the endothelial cells; 2. GS I strongly stained the dilated subendothelium in regions of mild mesangial interposition; 3. GS‐I stained the cell coat of invasive macrophages; 4. GS‐I and UEA‐I stained the cell membrane‐like material derived from degenerative endothelial cells; 5. RCA I stained the epithelial and endothelial cell coats and the glomerular basement membrane. A cell line (designated SHIN 3) was established from a 56‐year old Japanese woman diagnosed as having serous cystadenocarcinoma of the ovary. SHIN 3 produced the tumor marker, CA125. When samples were subjected to one dimensional gel electrophoresis followed by immunoblotting with anti CA125 antibody, antigen binding appeared in the low‐molecular mass fraction around 50 KDa as well as in the high molecular‐mass fraction (more than 200 KDa). Subsequent resolution of this lower molecular‐mass component by two dimensional gel electrophoresis gave rise to bands of 49 KDa near an isoelectric point of 7.3. Treatment of the CA125 antigen with 10 mM periodic acid resulted in no loss of activity. However, reduction and alkylation in 6 M guanidine HCI or treatment at 100 C for 20 min resulted in complete loss of activity. Treatment with various glycosidases did not inhibit activity, whereas digestion with proteases (except for papain) produced complete loss of activity, suggesting that the CA125 antigenic determinant is composed of con‐formationally dependent

In order to differentiate yolk sac type alpha fetoprotein (AFP) from hepatic‐type AFP, 5 yolk sac tumors (YSTs) and 6 hepatocellular carcinomas (HCCs) were examined immunohistochemically by the peroxidase antiperoxidase (PAP) method for AFP, and paradoxical concanavalin A (P Con A) staining, which has been reported to detect glycoprotein including AFP. In all 5 YSTs, AFP was negative for P Con A staining. On the other hand, AFP was strongly positive for the same staining in all 6 HCCs. A similar staining pattern for AFP was observed in human yolk sac endodermal cells and embryonal hepatocytes. Thus, it was clarified that yolk sac type AFP was unable to bind with Con A, in contrast with hepatic‐type AFP, on tissue sections. It was concluded that the PAP method for AFP and P Con A staining might facilitate the immunohistochemical differentiation of these two types of AFP, and that it would be usetul for clarifying the histogenesis of various AFP secreting tum

Ffteen cases of peripheral T cell lymphoma were studied to evaluate the respective properties of various histologic types using enzyme histochemical and ultrastructural examinations in addition to immunological methods. Eleven cases in an ATLA negative group manifested various histologic patterns such as IBL like, pleomorphic and Lennert's lymphomas in comparison with the relatively monomorphic proliferation of neoplastic lymphoid cells in the 4 ATLA positive cases. The presence of neoplastic clear cells is characteristic of peripheral T‐cell malignancies, and is likely to be found in CD4 lymphomas. There is an occasional reaction of epithelioid histiocytes and plasma cells with eosinophils, the former being designated Lennert's lymphoma and the latter IBL like T‐cell lymphoma. Immunological examination revealed four immunophenotypic patterns: (1) CD2+3+4+8+, (2) CD2+3−4+8−, (3) CD2+3+4−8+, and (4) CD2+3+4+8+, but did not provide information concerning the intimate relationship between histologic types and immuno phenotyes. β‐Glucuronidase reactivity, however, contributed to the distinction between helper and suppressor T cell malignancies, suggesting its usefulness for distinguishing these two cell types and their malignant

A case of mixed medullary follicular carcinoma of the thyroid is reported. Grossly, the tumor was a solid, grayish white, well circumscribed mass without lymph node metastasis. Microscopically, the tumor showed both medullary and follicular areas. The follicular areas occupied discrete portions of the tumor, and were considered to be neoplastic. Tumor cells in the medullary area were polyhedral or spindle‐shaped. There was no amyloid deposition within the tumor. Immunohistochemically, tumor cells in the medullary area were positive for calcitonin and negative for thyroglobulin. Some cells lining the follicles were positive for thyroglobulin. By electron microscopy, two types of tumor cell were observed. One type contained numerous cytoplasmic secretory granules, whereas the other type had few granules and showed a prominent rough endoplasmic reticulum. A 44 year old Japanese woman with leiomyosarcoma of the inferior vena cava is reported. She presented with Budd Chiari syndrome and died of hepatic failure about 3 months after the onset of symptoms. We report an autopsy case of malignant histiocytosis. The clinical course was rapidly progressive and terminated with jaundice and respiratory failure. Histologically, there was diffuse infiltration of large atypical cells in the liver, spleen, lymph nodes and bone marrow. It was of interest that these tumor cells contained a number of bizarre multinucleated cells histologically indistinguishable from Reed Sternberg cells of Hodgkin's disease, and that these atypical cells expressed DAKO Ml (identical to Leu Ml) and Ki‐1 antigens and also showed binding to peanut agglutinin (PNA), representative markers of Reed Sternberg cell. An absence of epithelial membrane antigen and presence of Leu Ml antigen in the tumor cells made a diagnosis of Ki‐1 lymphoma unlikely.
