摘要:
The molecular cloning of carcinoembryonic antigen (CEA) and several cross-reacting antigens reveals a basic domain structure for the whole family, which shows structural similarities to the immunoglobulin superfamily. The CEA family consists of approximately 10 genes which are localized in two clusters on chromosome 19. So far, mRNA species for five of these genes have been identified which show tissue variability in their transcriptional activity. Expression of some of these genes in heterologous systems has been achieved, allowing the localization of some epitopes. The characterization of a CEA gene family in the rat and a comparison with its human counterpart has been utilized in the development of an evolutionary model.

Rats of the Lewis strain were immunized with bovine basic myelin protein to develop experimental allergic encephalomyelitis. Neither human nor rat fetal sources of purified α-fetoprotein were able to significantly ameliorate the development of this dyscrasia. The plasma of pregnant rats at term was without effect. Immunization of pregnant rats was also accompanied by the usual course of development of typical encephalomyelitis

Concentration of fetal hemoglobin (HbF) was measured in the blood of 172 cancer patients with solid tumors. The patients’ age groups were compared with matched age groups of normal controls. Above the age of 50, a significant difference was found between patients and normal controls. In 70% of the patients with active disease, the concentration of HbF was above the normal range. There were indications that by reducing HbF concentrations irradiation of patients interferes with the evaluation of the results. In addition to whole blood, HbF was also measured in the plasma of 35 patients; the average HbF concentration in this group was found to be twice as high as that of the control

A panel of 13 mouse monoclonal antibodies (mAb) and 1 rabbit polyclonal antibody was tested for reactivity with tumor cells in 26 effusions obtained from patients with carcinoma of ovary, breast, or mesothelioma, using an immunoperoxidase staining reaction. Specific staining of tumor cells but not reactive mesothelial cells was demonstrated with some of the mAb in the panel. In 4 of 26 effusions no evidence of malignancy was obtained after routine cytological staining, but this was reversed on the basis of immunoperoxidase staining of tumor cells with the mAb in the panel. Serial effusions were evaluated in 4 patients during the course of chemotherapy, allowing an assessment of effect of therapy on the antigenic characteristics of the tumor cells. In another 4 patients, the results of immunoperoxidase staining of effusions were compared with those obtained by applying the same antibody panel to solid tumor nodules. There was a tendency to develop changes in the pattern of reactivity during therapy, and the pattern of reactivity was more restricted in tumor nodules than in effusions. One of the mAb in our panel (2G3) was consistently shown to produce strong staining of a high proportion of tumor cells in effusions and tumor nodules from patients with ovarian or breast cancer and may be of value in immunocytochemical diagnosis and therapy of epithelial malignancies.

A sensitive procedure involving lectin affinity electrophoresis of α-fetoprotein (AFP) was established. AFPs electrophoresed on lectin-containing gels were blotted on nitrocellulose membrane which was precoated with the specific antibody to AFP and stained with peroxidase-labeled anti-AFP antibody. This method could detect as little as 4 μg/l of purified AFP dissolved in buffer, or 50 μg/l in serum specimens. A number of patients with liver disease have been followed for long periods in Nihon University Hospital, Tokyo. Serum specimens were collected serially and stored frozen. The IgG1 monoclonal antibody, 44–3A6, was raised against the human lung adenocarcinoma cell line, A549. It has been shown to react with a 40,000 MW protein found on the cell surface, which is preserved in formalin-fixed paraffin-embedded tissues. A recent study of pulmonary carcinomas utilizing immunohistochemical methods showed exclusive binding to lung adenocarcinomas, subsets of neuroendocrine tumors, some carcinoids and a subset of large cell carcinomas. Reactivity was not seen in squamous cell carcinomas and small cell neuroendocrine carcinomas. In addition, melanomas, sarcomas and hematologic malignancies do not express this antigen. We now report on the reactivity pattern of 44–3A6 in adenocarcinomas of nonpulmonary primary sites and in normal adult organs. Strong diffuse staining of neoplastic cells in adenocarcinomas of the stomach, colon, pancreas, gallbladder and breast was noted. Adenocarcinomas arising in the endometrium, ovary, kidney, prostate, thyroid and liver were either negative or showed weak and/or focal reactivity. Strong staining patterns were even noted in adenocarcinomas which had an ‘undiffer-entiated’ component; i.e., lacking well-defined glandular elements.Immunoreactivity was noted in epithelial cells in several tissues from which these adenocarcinomas arose including the bronchial tract, stomach, small intestine, pancreas and colon, whereas epithelial cells from the endometrium, kidney, ovary, prostate and thyroid were negative or showed diffuse weak immunoreactivity. Our findings indicate that monoclonal antibody 44–3A6 recognizes an epithelial antigen on subsets of normal as well as transformed glandular epithelia. Alpha-fetoprotein (AFP) and albumin are related proteins, which contain about 585 amino acid residues that are organized in a characteristic structure of 3 homologous domains of about 195 amino acid residues. We have compared the domains of mouse, rat, and human AFP and rat, bovine, and human albumin using computer programs designed to quantify relationships between proteins. The comparisons of corresponding domains of the AFPs (e.g. domain I of rat and human AFP) reveal that each domain is well conserved. Similar results were found for the comparisons of corresponding domains in the albumins. In contrast, there was much less similarity between corresponding domains of albumin and AFP. These comparisons between AFP and albumin revealed that : (1) the amino acid sequences in domain III, which is at their carboxy terminus, are most conserved, and (2) the amino acid sequences in domain I, which is at their amino terminus, are least conserved. This suggests that there are differences in the constraints on amino acid substitutions among the domains of AFP and albumin during the approximately 400 million years since they diverged from a common ancestor. Also, computer studies revealed that there are substantial differences between domains I, II, and III in each AFP and albumin species, which indicates that these domains are significantly different from their ∼ 195 residue ancestral domain. Finally, we find that mouse and rat AFP are more dissimilar to the albumins than is human AFP. Monoclonal antibody (moAb) B72.3 appears to be exceptional among the many mouse moAbs reactive with human tumor cells in its degree of tumor specificity. The antigen recognized was characterized as a mucin-like molecule. We report here that B72.3 reacts strongly with ovine submaxillary mucin and that reactivity is abolished by desialylation, suggesting that the determinant recognized is the disaccharide N-acetylneuraminic acid α(2→6)-N-acetylgalactosamine, linked to serine or threonine, which might be designated sialylated Tn. A variety of human mucin preparations, including human salivary mucins, and other glycoproteins containing O-linked or N-linked carbohydrates were nonreactive. This study concerns the immunohistologic distribution of carcino-embryonic antigen (CEA) in tissues and organs from 86 legal abortions, stillborn fetuses and perinatal deaths and from 5 adults without inflammatory disease or cancer. Monospecific antibodies to CEA of both polyclonal and monoclonal origin were applied to serial sections obtained from formalin-fixed, paraffin-embedded tissue blocks. Starting from the 9th week of gesta-tional age, a positive staining reaction for CEA was found in the surface epithelium of the tongue, the tracheal mucosa and the following locations of the gastro-intestinal tract: the gastro-oesophageal junction, the pyloric antrum, the upper duodenum, throughout the colon and appendix. In the adult, CEA was also found at these sites. All other organs such as the central nervous system, lung, thyroid, thymus, liver, pancreas, gastric corpus, spleen, adrenals, kidney, ureter, bladder, gonads and breast were negative for CEA. Therefore, CEA appears to be a normal antigen in the gastro-intestinal tract at any age from fetal life onwards.
