An in vitro latency system using herpes simplex virus type 2(HSV2)-infected human embryonic lung cells treated with cytosine arabinoside (ara-C) and incubated at 39.5° after drug removal was used to examine the ultrastructure of the infected cells during (i) ara-C treatment, (ii) the latent period after ara-C removal and temperature elevation, and (iii) reactivation after superinfection with human cytomegalovirus. In the presence of ara-C ‘empty’, nonenveloped, herpesvirus-like particles were observed in nuclei with fragmented chromatin. After removal of ara-C and temperature elevation, no virions were found. At no time during latent infection were complete virions seen. After superinfection with human cytomegalovirus, extensive synthesis of proteins was suggested by a marked proliferation of rough endoplasmic reticulum, polysomes, and Golgi vesicles. This was followed by the appearance of an electron-dense viral matrix in both the nucleus and cytoplasm and by formation of normal nucleocapsids in the nucleus. Avian myeloblastosis virus (AMV) genomic RNA was translated in vitro to identify the AMV transforming gene (myb) product(s). Full-length RNA yielded, in addition to the expected Pr76gag, a gag-related product of 92,000 daltons which did not appear to be a gag-oncogene fusion protein. Subgenomic AMV RNA in vitro translation, nonstructural products (specific for AMV) were of 54,000, 49,000 and 34,000 daltons. Based on their sizes and similar peptide maps for the smaller two proteins, it is proposed that they derive from the myb region of AMV.

By analyzing the fine structure of varicella-zoster virus (VZV) DNA, a naturally occurring heterogeneity was found on the right end of VZV DNA, but no evidence of a true terminal repetition was uncovered. We were unable to confirm the report of Straus and co-workers (1981) that there is a relatively high frequency of circular VZV DNA in low-passage virus. On long-term cell passage, extensive heterogeneity appeared concomitant with the accumulation of apparently defective VZV DNA.

摘要:
The antigenic, biological, and chemical properties of 24 selected Changuinola serogroup viruses were examined. The viruses tested were chloroform-resistant, and they were lethal to newborn hamsters after intracerebral inoculation. The prototype Changuinola virus strain (BT-436) replicated in mosquito and sandfly cell cultures. In complement-fixation tests, the viruses were broadly cross-reacting and indistinguishable; but by neutralization test at least 12 distinct serotypes were identified, and by PAGE of double-stranded RNA 22 distinct profiles were found. These data suggest that the Changuinola serogroup may be comprised of a large number of genetically different viruses. A brief review of the natural history of Changuinola serogroup viruses is also given.

A singly enveloped nuclear polyhedrosis virus (SNPV) of the white-marked tussock moth, Orgyia leucostigma SNPV was serially passaged 60 times each in OL-12 and OL-13 cells. Percentage infected cells and PIB production stayed generally high throughout serial passaging in OL-13 cells, but declined sharply after the 41st passage in OL-12 cells. PIBs from the 4th passage of the virus in OL-12 cells were tested and found to be pathogenic to O. leucostigma larva
