Neutropenia can be produced with antimitotic chemicals, but this method lacks specificity. An alternative is to use antibody‐dependent cytotoxicity to produce neutropenia; however, this method has not been completely evaluated with respect to efficacy, specificity, and potential collateral damage, especially to constituents of bone marrow. This study used in vitro and in vivo methods to evaluate specific biological effects of a commercially available rabbit anti‐rat neutrophil (PMN) antiserum in F344/N rats. The viability of rat pulmonary alveolar macrophages (PAMs), PMNs, and lymphocytes in vitro was quantified using a trypan blue dye exclusion test. Amounts of antiserum in vitro that rendered PMNs 100% nonviable did not decrease the viability or phagocytic ability of the PAMs and did not decrease the viability of the lymphocytes. Intraperitoneal (IP) injection of the antiserum into rats resulted in complete depletion of the PMNs and about a 50% depletion of the lymphocytes in circulating blood within 24 hours. The numbers of both cell types remained lowered for 5 days, but returned to control values by Day 6 after the IP injection. The antiserum had no effect on the numbers of PAMs or lymphocytes in the pulmonary alveolar airspaces, as determined by quantifying the numbers of these cell types in bronchoalveolar lavage fluid (BALF). The numbers of PMNs in BALF, however, decreased on Days 3 and 4 after IP injection of antiserum, but were not different from control values by Day 5. The viability of the PAMs in BALF of treated rats was not different from control values at any time point. There were no morphological indications that the injected antiserum damaged lung tissue or stem cells in bone marrow. Results demonstrate that the anti‐rat PMN antiserum administered IP to F344/N rats depletes circulating PMNs and partially depletes lymphocytes for a period of about 6 days without adversely affecting the precursors of red or white blood cells in bone marrow. Serum α‐fetoprotein (AFP) concentrations were determined, by use of an automated microparticle enzyme‐linked immunoassay (MEIA), in 16 control dogs and 48 dogs with previously untreated, histologically confirmed, naturally occurring neoplasia (17 dogs with lymphoma and 31 dogs with nonhematopoietic malignancies; 13 dogs with carcinomas, 18 dogs with sarcomas). Mean serum AFP concentrations for untreated dogs with lymphoma, for dogs with sarcomas, and for dogs with carcinomas were not significantly different from the mean serum AFP concentration for the 16 untreated control dogs. Mean serum AFP concentration for dogs with transitional cell carcinoma (n = 7) was not significantly different from the mean serum AFP concentration for the control dogs. It has been shown previously by others that a mean serum AFP concentration<225 ng/mL is suggestive of a hepatic malignancy (e.g., hepatocellular carcinoma, lymphoma). In our study, all 48 dogs had a normal complete blood count, serum biochemical analysis, and urinalysis. Only one of the 48 dogs was found to have a serum AFP concentration225 ng/mL, with or without evidence of a serum biochemical abnormality, may suggest primary and/or secondary hepatic involvement with a neoplastic disease and may warrant an adjustment in clinical stage and prognosis. A prospective diagnostic and therapeutic evaluation of dogs with naturally occurring tumors having an elevated serum AFP concentration would determine the validity of this conclu

The objective of this study was to compare total carbon dioxide (TCO2) levels measured by three different methods. Two hundred jugular venous blood samples from dogs admitted to the Veterinary Teaching Hospital, Ontario Veterinary College with various clinical disorders were analyzed by the Radiometer blood gas analyzer (BGA) and the Coulter DACOS analyzer. In 70 of these samples, TCO2was also measured by the Kodak Ektachem DTE chemistry analyzer. Comparison of the agreement between methods revealed intraclass correlation coefficients (ICC) of 0.63, 0.79, and 0.82 for the DACOS‐Ektachem, DACOS‐BGA, and Ektachem‐BGA comparisons, respectively. Evaluation of the effect of storage time on TCO2content of stored serum samples measured on the DACOS analyzer revealed a decrease of almost 2 mmol/L and a decrease in the intraclass correlation coefficient values between the DACOS analyzer and other methods after 7 hours storage time. The results of this observational study revealed lower agreement between the three methods in this study than previously reported between other methods of TCO2measurement.
