AbstractIn this study the influence of sodium ursodeoxycholate (UDC) on hepatocyte replicative activity was evaluated using quiescent or primed hepato‐cytes obtained from normal rats or from rats fed with a protein‐free diet, respectively. At physiological concentrations UDC stimulated proliferation in quiescent or primed hepatocytes cultured in minimum essential medium plus insulin, and augmented the replicative activity in hepatocytes already stimulated by low concentration of epidermal growth factor and normal rat serum. However, UDC was not the only bile salt (BS) to show this stimulatory effect on hepatocyte proliferation. The stimulatory activity of BSs was not correlated with their degree of hydrophilia. © 1993 Wiley‐Lis

AbstractEstrogen receptor mRNA was detected by a non‐radioactive in situ hybridization assay in tumor and non‐neoplastic liver tissues. A synthetic oligo‐nucleotide complementary to the human estrogen receptor mRNA was 3′‐labeled with digoxigenin‐deoxyuridine triphosphate (dUTP). Hybrids were revealed by an alkaline phosphatase‐conjugated anti‐digoxigenin antibody. Fourteen primary hepatocellular carcinoma tissues (and one meta‐static) were obtained at surgery from 15 patients. The corresponding non‐neoplastic liver tissues were available in 13 cases. The estrogen receptor mRNA was detected in 11 tumorous and 7 non‐tumorous liver specimens. The staining was cytoplasmic and involved the majority of transformed hepatocytes, whereas a less widespread and weaker signal was found in normal hepatocytes. Within non‐neoplastic tissue, bile duct epithelial cells could also be occasionally stained, whereas other cell types, such as vasal endothelial cells, were negative. Appropriate controls established the specificity of the reaction. Detection of the estrogen receptor protein by immu‐nohistochemistry in these same specimens was invariably negative. AbstractThe assessment of biological markers as potential indicators of disease aggressiveness is still an open problem in hepatocellular carcinoma. Cell proliferation and extracellular matrix (ECM)‐associated antigens and tumor‐cell products can be associated with clinical aggressiveness in this tumor type, as has already been demonstrated for others. Cell proliferation, expressed as the in vitro [3H]thymidine labeling index, and ECM‐associated antigens, such as type IV collagenase and laminin receptors, were assessed on the same paraffin‐embedded samples. A strong association (P =0.0001) was observed between the expression of collagenase and laminin receptors, with a correlation coefficient (rs) of 0.68. No relationship was found between cell proliferation and ECM‐associated antigens. Moreover, the biological markers were generally independent of clinicopathologic features, except for a higher number of collagenase‐ and laminin receptor‐positive cells in large (≥ 5 cm) compared with small (<5 cm) tumors. In the present series of hepatocellular carcinoma patients, the 3‐year clinical outcome was significantly affected by cell proliferation and ECM‐associated antigens. ©

AbstractThe biochemical, regulatory, and physiological aspects of plasmin formation by the urokinase‐type plasminogen activator (uPA) is discussed and its involvement in cancer malignancy analyzed. Cell surface plasminogen activation has been shown to be essential in extracellular matrix degradation, basement membrane dissolution, and cancer invasiveness and metastasis in a variety of model systems. The ability of cells to produce plasmin on their surface depends on the presence of uPA and plasminogen receptors, and is the basis of the regulation of the plasminogen activating system in vivo. © 1993 Wiley‐Liss,
