S.‐F. LU AND P.‐P. CHANG. 1996. A bacterium producing thermostable neutral protease was isolated from an orchard in Taiwan and identified asPseudomonas aeruginosa. Optimal activity of the protease fromPs. aeruginosawas observed at 60°C and pH 7.0. The protease was thermostable, 85% activity being retained after incubation at 60°C for 1 h. When growing cells were incubated in the presence of 100 ppm benzylpenicillin (BP) for 30 min and then harvested for metabolic studies, the initial rate of glucose utilization by resting cells was reduced by 19.6% compared to control cells, whereas the rates of protein, DNA and RNA synthesis were reduced by 40.7%, 55.0% and 87.5% respectively. Most chemically‐defined media for the growth ofBacillus subtilis168 in batch culture are unsuitable for use in nutrient limitation experiments due to either their low cell density yield or their high numbers of added amino‐acids. The authors have developed a medium which gives relatively high cell densities and is highly chemically‐defined, containing only one added amino‐acid. Growth ofB. subtilisin this new medium was observed over time using optical density measurements at 470 nm. The growth curve exhibited entry into exponential phase after only 1–2 h and stationary phase after 8 h. Cell density yields and comparison of growth rates withB. subtilisgrown in Fang and Demain Medium (1989) suggest that this new medium is highly suitable for the growth of this micro‐organism under both normal and nutrient limited

A compact battery‐operated, portable field filtration apparatus was developed for efficient recovery of parasites, includingGiardia lambliacysts, from large volumes of water. In conjunction with modified filter elution and processing procedures, the filtration device was evaluated in field trials. To monitor performance of the procedure, a known number ofGiardia lambliacysts were added during filtrations through a control injection assembly. This allowed calculation of cyst recovery at each test site, thereby allowing comparison of percentage recoveries from many diverse water sources. This apparatus and modified method now provide the tools to investigate further extraction and purification techniques to improve overall yield ofGiardia lamblia, other parasites and potentially other organisms such as bacteria and viruse

Generally pseudomonads degrade phenol through themeta‐pathway, butPseudomonas stutzeristrain SPC2 isolated by flask enrichment of municipal sewage degraded phenol through theortho‐pathway. The strain utilized up to 1200 ppm of phenol as a sole source of carbon and energy. The strain also degraded benzoate and 4‐hydroxy and 3,4‐dihydroxybenzoates via theortho‐pathway. The effect of pH on the heat resistance ofBacillus stearothermophilusspores at 100°C in the presence of 0.11 mol 1‐1lactic acid and 0.2 mol 1‐1sodium phosphate buffer was examined. At pH values of 7.0 and 6.0 spores survived 60 min exposure unharmed but at pH 4.3 and 3.0 they died with decimal reduction times (DRTs) of 27 min and 2.8 min, respectively. Death rates were similar in the presence or absence of hydrated soybean cotyledons. In the presence of phosphate buffer and cotyledons at mean pH 3.6 the DRT was 118 min but in the presence, in addition, of lactic acid it was 11 min. It is suggested that the enhanced death rate was due to toxic effects of undissociated lactic acid.Rhizopus oligosporusNRRL 2710 grew well on cotyledons, having pH values from 7.0 to 3.7, prepared by boiling for 60 min in the presence of 0.11 mol 1‐1lactic acid and 0.2 mol 1‐1phos

The ratio unsaturated/saturated fatty acids decreased following spray drying, indicating the formation of lesions in cellular lipid‐containing structures. The same method was used to analyse membrane lipids ofLact. bulgaricusduring storage. Similarly the ratio of unsaturated/saturated fatty acids in dried cells decreased further during storage in air, presenting evidence of lipid oxidation after prolonged storage.
