The present studies were carried out in order to explore, whether or not senile mice react in the same manner to a single injection of either 50 μg Serratia marcescens endotoxin (ET) or 3 × 109 killed Bordetella pertussis organisms (PO) as young adult mice. This was not the case. The following essential differences were observed. (1) Aged mice produce PO-mediated blood leukocytosis to a distinctly reduced extent, as compared to young adult mice. (2) The injection of either ET or PO into adult mice led to a short-lived, but significant increase in the numbers of cells producing 19S hemolysins to sheep erythrocytes, whilst the response regarding those cells synthesizing 19S antibodies to rabbit erythrocytes was very poor. Thereby, the reactivity of young adult mice was found to be very homogenous. In contrast to untreated young adult mice, the spleens of untreated senile animals contained on the average larger numbers of plaque-forming cells, the values differing strongly from spleen to spleen. Following treatment with either ET or PO the senile mice showed extreme differences regarding their capacity to react with an increase in hemolysin-forming spleen cells, thereby, reaching the maximum response in each case later than young adult mice. (3) There was no close positive relationship between the existence of cells producing 19S hemolysins to sheep erythrocytes and the occurrence of those synthesizing corresponding antibodies to rabbit erythrocytes. Aged (20 months old) NMRI mice, showing a significantly reduced primary immune response to 4 × 108 sheep erythrocytes (SE), reacted to a secondary injection of 4 × 108 SE 1 month after primary immunization with a typical secondary immune response at both the adjuvant cellular and humoral levels. This specific anamnestic response was not found to be diminished, as compared to that of young adult controls (3 months old at the time of primary immunization). When killed cells of Bordetella pertussis were given as an adjuvant together with the primarily injected erythrocyte antigen, the process of priming for the secondary immune response was found to be significantly increased in both the aged and the young adult animals. The impressive restoration of the antibody-forming potential observed following secondary antigenic stimulation was associated with a remarkable restoration of the splenic structure. Splenic amyloidosis did not impair the secondary antibody-forming potential to a significant extent. It is suggested that the suppressed immunological reactivity of aged mice to a primary antigenic stimulus is mainly due to deficiencies in the regulatory factors, such as antigen-processing and antigenic triggering of immunocompetent precursor cell

Synthetic sponge (polyether) was implanted in the subcutis of male albino rats of 3 age groups: 6 weeks old, 9 months old and 20–22 months old. Morphological and biological examinations were made on the granulation tissue which migrated into it. As parameters for the inter-cellular catabolic process, the activity measurements of the β-glucuronidase, β-acetylglucosaminidase, cathepsin D and a collagen catabolic enzyme were conducted. The incorporation rate of 14C-glucosamine and 14C-ρroline served as an indicator for the synthesis mechanisms. In addition to this, the DNA content of the sponge was determined. The measurements of all the above-named parameters were made 14, 21 and 30 days after the implantation, on the sponge implanted in the paravertebral subcutis. The results demonstrate that, depending on the duration of implantation as well as on the age of the animal, distinct alterations of the examined parameters take place. 摘要:
With increasing dosage ACTH was applied intramuscularly to old and young rats. The capacities of stain affinity of hepatic nuclei for acridinorange and phenanthrenquinone were analysed cytophotometrically. There were significant differences between the age groups, which might suggest age-dependent stabilisation of DNA-nucleoprotein complex.

Age-dependent changes of the DNA-nucleoprotein complex in hepatic cells of guinea pigs are analysed by a modified Sakaguchi reaction. There is a statistically significant difference of the capacity of stain affinity between cells of young and old subjects.

Lacrimal gland, glandula orbitalis externa and glandula infraorbitalis were investigated in rats of different ages and both sexes with light microscopy and by chemical methods. Cells of the lacrimal gland displayed no sexually-dependent properties. Only the glandula orbitalis externa and the glandula infraorbitalis showed sexual dependence and age-dependent changes, beginning in 3-month-old rats. While both glands of the female rats displayed only enlargement of the nucleus, the cells of the male rats, beginning at the onset of puberty, displayed marked enlargement, changes in shape, and the appearance of inclusion bodies and cytoplasmic inclusions. With the occurrence of these manifestations in the males, there was also a reduction in the DNA content in these glands. Sexually-dependent manifestations of the glandula orbitalis externa and glandula infraorbitalis in the male rats may be caused by a viral infection.

With autoradiographic and cytophotometric methods, the labelling index and ploidy pattern of the cells of the lacrimal gland, the glandula orbitalis externa and the glandula infraorbitalis of male and female rats of different ages (1,3,6 and 12 months) were investigated. The nuclei of the lacrimal glands showed a predominantly diploid pattern in both sexes. Only the 12-month-old male and female rats showed a small number of tetraploid nuclei In contrast, beginning with 3 months, both male and female rats exhibited a large number of tetraploid nuclei in the glandula orbitalis externa and glandula infraorbitalis. In addition, the female rats displayed round nuclei and the male rats exhibited dumb-bell-shaped nuclei. The dumbbell-shaped nuclei in the males were a result of what appeared to be polyploidisation. With the increase in polyploidisation, there was a simultaneous decrease in the DNA content and an increase in the labelling index. This indicated that these cells had a decreased turnover. The mechanism for the apparent polyploidisation of the male rats, beginning in the 3rd month, is discussed.
