The aim of this investigation was to develop an experimental model for studies on the significance of fibrin deposition for wound healing. Systemic defibrinogenation with Arvin® was used to achieve abnormal fibrin deposition in subcutaneously implanted cellulose sponges. The formation of granulation tissue in the sponges was studied histo-logically. Arvin injections resulted in a rapid decrease in plasma fibrinogen concentration with a concomittant rise in fibrin-degradation products. No change in FXIIIa activity was seen. During defibrinogenation an abnormal fibrin deposition was found in the sponges. The fibrin strands appeared irregular and disrupted. The number of fibroblasts and collagen fibrils was reduced in granulation tissue formed during defibrinogenation. The model used seems to allow controlled studies on the significance of fibrin deposition for wound healing. Defibrinogenation was found to influence granulation tissue formation

A guinea-pig model is presented for the research in burns. Partial and full-thickness burns can dependably be produced. An intra-arterial catheter can be used to draw blood as well as to perfuse the wound with drugs.

Structure and function of isolated liver ribosomes from 10 dogs subjected to 1 h of hepatic artery ligation (HAL) or hemorrhagic shock were studied. A shift of polyribosomes to monoribosomes and a decreased capacity to incorporate amino acids into proteins were seen in both experimental conditions. The effects were more pronounced in hemorrhagic shock. Both structural and functional changes were reversible after HAL. In the shock experiments a slight increase of the ribosomal activity was seen after reinfusion but the initial values were not reached during the 2 h after the shock period. Some of the underlying mechanisms of altered ribosomal structure and function in liver ischemia are discussed.

Intravascular hemolysis and ultrastructure of erythrocytes from liver sinusoids were studied during and after infusion of Escherichia coli endotoxin in Labrador retriever dogs. Endotoxin infusion caused hemoconcentration, and induced disseminated intravascular coagulation (DIC), characterized by a rapid drop of platelet numbers, a gradual consumption of coagulation factors and activation of fibrinolysis. Advanced DIC and circulatory shock gradually developed, and the animals died after 7–15 h. Plasma hemoglobin concentrations did not rise for several hours, but late in the experimental period a significant intravascular hemolysis was constantly found. Circulating adenosine diphosphate (ADP) did not appear. During shock, liver biopsies revealed accumulation of erythrocytes often disintegrated within distended sinusoidal lumina. In advanced shock the fragmented erythrocytes seemed to form occlusive masses within the vessels. A fibrin-like material frequently appeared adjacent to the red cells. However, it did not have the periodicity characteristic for fibrin, and the ultrastructure of the material was very similar to that inside the erythrocytes. None of these changes were induced by saline infusion in control animals. The lack of fibrin formation and the late development of intravascular hemolysis indicate that red cell breakdown was of little importance for the initiation and progress of DI

The objectives of the present work were to study the role (and possible sources) of gastrin and glucagon, as measured by radioimmunoassay techniques, in the acute response to massive bowel resection in dogs. Four acute experimental models were designed for this purpose. Circulating portal and arterial hormone concentrations were estimated under basal conditions and after administration of an intraduodenal fat stimulus. Stimulated portal gastrin levels were significantly higher in animals with bowel resections than in intact animals (p < 0.05 to p < 0.01) but the depletion of N-terminal glucagon-like (GLI) reactivity or total GLI was only transiently significant (p < 0.05). Pancreatectomy, with or without resection, resulted in depletion of circulating N-terminal and C-terminal GLI, although not to zero values, suggesting extra-pancreatic sources of the GLI. Portal-arterial differences were noted for GLI but not for gastrin.

Renal revascularization by splenic artery implantation was performed in 5 dogs using a temporary arteriovenous fistula in the distal end of the implanted artery for the purpose of increasing the blood flow and counteracting the thrombus formation in the artery. Following creation of renal artery stenosis, the revascularization process was evaluated by serial consecutive splenic arteriographies and by estimation of blood flow through the implanted arteries. The experimental animals survived occlusion of the ipsilateral renal artery and contralateral nephrectomy with maintenance of a reasonably good renal function. The present study also indicates a facilitation of the vascularization from the implanted artery when the initial thrombus formation within the artery was prevented by creation of such a fistula.

The role of pancreatic fragment size prior to collagenase treatment, pH, K+ concentration and length of intraportal infusion was studied in islet cell transplantation of streptozotocin-induced diabetic adult rats. This study demonstrates that pancreatic fragments of 1 mm prior to collagenase digestion, pH of 7.4, extracellular potassium concentration and 10 min of intraportal infusion are important factors in successful islet cell transplantation of adult rats receiving six donor pancreases for amelioration of chemical diabetes. These factors should be taken into consideration when consistent islet cell transplantation results are to be expected.
