The acute thrombogenicity of a new polyurethane urea graft has been compared with polytetrafluoroethylene (PTFE) grafts using labelled homologous platelets in the carotid arteries of sheep in a low-flow model. There was no difference in patency. Thrombus weight and thrombus free surface were the same in both types of graft after 4 h. The accumulation of homologous 32P platelets was measured at the 2 anastomoses and at a midgraft position. At no point was there any statistically significant difference between the 2 grafts, but the activity was always somewhat higher in the polyurethane grafts. Although more compliant, in this study polyurethane urea grafts behaved much like PTFE ones.

Some biochemical characteristics of collagen extracted from granulation tissue were studied and compared with those of normal skin and scar. By using electrophoretic techniques the type III collagen content was confirmed to be significantly greater in granulation tissue and lower in scar with respect to normal skin. The chromatographic determination of hydroxylysine (Hyl) glycosides in collagen extracted from granulation tissue showed a significant increase in both the degree of Hyl glycosylation and in the di-/monoglycoside ratio, while both parameters turned out to be lower in scar. These data suggest that the degree of Hyl glycosylation and the di-/monoglycoside ratio could represent an index of the degree of collagen fiber maturation.

Measurement of reticulo-endothelial system (RES) phagocytic function by clearance of intravenous micro-aggregated human albumin (HMAA) showed prolonged clearance in both patients (p < 0.001) and rats (p < 0.001) with extrahepatic biliary tract obstruction compared with non-jaundiced controls. After the administration of the immune stimulator, N-acetyl-L-α-aminobutyryl-D-isoglutamine, the mean HMAA clearance rate in jaundiced animals was similar to that of non-jaundiced controls. The implications of modifying RES phagocytic function to prevent overspill of endotoxins from the portal to systemic circulation in obstructive jaundice are discussed

Beta-blocking agents are increasingly used as preoperative treatment of hyper-thyroid patients. Relatively little is known about the effects of these drugs on metabolic alterations in hyperthyroidism. The aim of this investigation was to study the effects of two different beta-blocking agents on the urinary excretion of 3-methylhistidine (3-MH) during experimental hyperthyroidism in rats. Experimental hyperthyroidism was induced by daily intraperitoneal injections of triiodothyronine (T3; 100 µg/l00 g body weight) for 3 days. Control animals were injected with corresponding volumes of solvent. Groups of rats received food enriched with metoprolol (8.8 mmol/kg of diet) or propranolol (3.3 mmol/kg of diet) or food without additions. Urinary 3-MH excretion was increased by about 40% during experimental hyperthyroidism. A similar increase of 3-MH excretion was found in animals receiving T3 + metoprolol, whereas the excretion of 3-MH was reduced to control level in hyper-thyroid rats receiving propranolol. No effects of metoprolol or propranolol on 3-MH excretion were found in control animals. Although the source of 3-MH cannot be exactly defined, the present results indicate that increased proteolysis in skeletal muscle and/or other tissues during experimental hyperthyroidism was reduced by propranolol

Subtotal colectomy was performed in 17 rats. After 2 or 4 weeks the entire pancreas was excised and isolated pancreatic lobules were incubated for 60 min in Krebs-Ringer-Hepes solution either with or without addition of ceruletide (5 × 10–10 g/ml). Amylase was determined every 15 min in the supernatant medium. Amylase, trypsin, DNA and total protein were measured in homogenized pancreatic tissue. Morphology was performed via light-microscopy analysis. Two weeks after subtotal colectomy we found an increase in pancreatic wet weight, enzyme and protein content, cellular size and secretion capacity in vitro, whereas DNA concentration in relation to pancreatic tissue weight in grams decreased. These changes pointed to pancreatic hypertrophy. Four weeks after surgery, pancreatic wet weight, enzyme content and DNA content as well as in vitro amylase discharge increased, whereas cellular size remained unchanged compared to controls and sham-operated rats. The long-term effect of total parenteral nutrition (TPN) was studied in rats. A failure to thrive developed and all animals died within 40 days of TPN. Autopsy invariably demonstrated the increased weight and volume of the liver and spleen. Multiple fat emboli were found in the lungs of all animals, and multiple epithelioid cell granulomas in the liver of all and in the spleen of most animals. Chronical low grade infection could not be demonstrated. Microorganisms were not found in blood or tissue specimens and endotoxin could not be demonstrated in plasma samples.

The study deals with the immunological capacity and perfusion rate of splenic grafts in pigs after autologous transplantation into the omentum majus. 12 months after the operation, the weight and relative perfusion of the regenerated tissue was clearly reduced. Independent of this fact, the splenic grafts could balance out an IgM-defíciency, observed due to splenectomy, 4 months after the transplantation. Furthermore, the number of antibody forming cells in the grafts was only slightly decreased compared to the normal spleen. These results show that regenerated splenic tissue is able to carry out specific immunological functions. One of the limiting factors in preventing postsplenectomy sequelae (i.e., sepsis) seems to be the poor regeneration weight and low perfusion of the grafts
