During protein separation using the toroidal coil centrifuge, the partition efficiency was found to vary according to the direction of the elution through the coil. In most cases, eluting either the lighter phase toward the direction of rotation or the heavier phase in the opposite direction produced higher efficiencies. The cause of this unusual phenomenon may be attributed to the Coriolis force acting on the mobile phase in the rotating coil. Using a flow-through glass vial and colored mobile phases, the effects of the Coriolis force on moving droplets were observed under stroboscopic illumination. As expected, the path of the descending droplets (heavier phase) shifted against the direction of rotation whereas that of the ascending droplets (lighter phase) shifted toward the direction of rotation. The effect of this Coriolis flow on the partition efficiency was studied with a set of toroidal coils with various core diameters.

摘要:
The present work is an attempt to correlate the composition and some physico-chemical properties of two-phase systems with the stationary phase retention in a coil planet centrifuge of J-type. Complex liquid systems containing an organic solvent (n-decane or chloroform), an extracting reagent (di-2-ethylhexylphosphoric acid, D2EHPA), water and a mineral salt (ammonium sulphate) are considered.

The influence of interfacial tension, density difference and viscosities of immiscible liquid phases on the stationary phase retention in a coil planet centrifuge has been studied. Two-phase liquid systems containing organic solvent (n-decane), extracting reagent (di-2-ethylhexylphosphoric acid), water, surfactant (sodium salt of dodecylbenzenesulphonic acid) and mineral salt (ammonium sulphate) have been considered. Varying concentrations of the system constituents allows selectively changing the interfacial tension, density difference or viscosity, all other conditions remaining the same. It has been shown that these parameters, especially interfacial tension, strongly affect the stationary phase retention. The data obtained are in good agreement with proposed theoretical models.

Studies of the separation of benzyl, phenylethyl and phenylpropyl alcohols by flash chromatography (FC) on 1.0 × 30 or 1.9 × 15 cm silica gel columns are compared with separation by countercurrent chromatography (CCC) on a 1.68 mm × 24 m, 56 mL, PTFE multilayer coil. Neither adjacent pair of alcohols could be separated by FC and separation of benzyl from phenylpropyl alcohol was poor. The small coil separation by CCC provided a resolution of 0.91 to 1.01 for benzyl and phenylethyl alcohols and better than baseline resolution of phenylpropyl alcohol from either benzyl or phenylethyl alcohol. Speed, sample capacity and selectivity of the two separation techniques are similar but resolution by CCC is enhanced by the characteristic symmetry of CCC peaks compared with the severe tailing observed in FC.

摘要:
In this paper, we describe the technique of analytical highspeed countercurrent chromatography coupled with electrospray ionization mass spectrometry (HSCCC/ESIMS) for the first time, and evaluate its performance in terms of chromatography and mass spectrometry. This technique demonstrates the capability and resolving power of HSCCC in the separation of a few natural product mixtures and utilizes ESIMS as a detector with continuous detection and high sensitivity. Application of this HSCCC/ESIMS system is illustrated by the separation and detection of biologically important compounds.

KSI (3-oxo-Δ5-steroid isomerase) was purified from crudeE. colilysate by countercurrent chromatography using a polymer phase system composed of polyethylene glycol 3350 and potassium phosphate (pH 7), each at 12.5%(w/w) in distilled water. Using the cross-axis coil planet centrifuge, a preparative scale separation was performed on 25mLE. colilysate containing ca 50 mg of KSI. About 40 mg of KSI was recovered at 98% purity. A small scale separation was performed onca. 3mg of15N-labeled KSI using the toroidal coil centrifuge. The present method eliminates sample loss and denaturation caused by the solid support and yields pure proteins in both preparative and analytical separations.

Countercurrent Chromatography (CCC) has the potential to play a major role in food analysis because it permits analysis of crude and complex samples. CCC was evaluated for its ability to separateS. aureusenterotoxin A (SEA), which is a common cause of food poisoning. Mushrooms containing native- or heat-denatured SEA were separated by Toroidal Coil CCC and the fractions containing the toxin were analyzed by Western immunoblotting.

摘要:
High-speed countercurrent chromatography (HSCCC), a liquid-liquid partition system, was used for the final purification of less polar alkali-labile glycolipids (ALGLs) of human brain. It has been reported that vertebrate brain contains ALGLs consisting of ester cerebroside and monoglucosyldiacylglycerol. ALGLs have alkali-labile ester bonds and are shown to be less polar than cerebroside.

Proteins were separated using a cross-axis coil planet centrifuge equipped with two different types of coiled columns, i.e., toroidal and eccentric coil assemblies. An aqueous-aqueous polymer phase system composed of 12.5% (w/w) polyethylene glycol 1000 and 12.5% (w/w) dibasic potassium phosphate was used for the separation of cytochrome C, myoglobin and ovalbumin. The toroidal coil assemblies with 1 mm ID tubing yielded the partition efficiency of 170 theoretical plates at the myoglobin peak. A slightly higher efficiency was obtained from the eccentric coil assemblies with the same diameter. The highest partition efficiency of 450 theoretical plates was attained from the eccentric coil assemblies with 0.85 mm ID and 64 mL capacity.

Aqueous-aqueous polymer phase systems composed of polyethylene glycol 1000-potassium phosphate were used for the countercurrent chromatographic separation of proteins using the type XL cross-axis coil planet centrifuge. It was found that the peak resolution of proteins is highly dependant on the pH of the solvent system. The best separation of cytochrome c, myoglobin and human serum albumin was achieved with 16.0% PEG 1000-12.5% potassium phosphate buffer at pH 9.2. The resolution and separation time were improved by pH-peak-focusing countercurrent chromatography.
