Chlorpromazine (CPZ) inhibits synchronized cell division in Tetrahymena pyriformis with 100% inhibition occurring at 8 µg/ml. CPZ markedly inhibited the incorporation of precursors into DNA and RNA, but had little effect on the incorporation of acetate into lipids. Amino acid incorporation into protein was also blocked, however, uptake of amino acid by the cell was inhibited to a similar extent. This indicates that CPZ had no direct inhibitory effect on protein synthesis. The data presented here indicate that CPZ may have other sites of action with nucleic acid syntheses appearing to be potential sites of primary inhibition in T. pyriformis

The effect of phenylbutazone in daily doses of 100 mg/kg on collagen metabolism in rats was studied after an injection of 14C-proline, by determining specificand total 14C-hydroxyproline activity in the skin collagen fractions and urine. It was found that a 21-day administration of phenylbutazone influenced the earlyphases of collagen metabolism by inhibiting collagen biosynthesis. The later phases of collagen turnover (maturation) were not affected by phenylbutazone treatment. In the given dose, phenylbutazone did not stimulate the catabolism of either newly-formed, soluble collagen, or mature, insoluble collagen.

Tritium labelled digitoxin and digoxin were administered intragastrically to rats provided with thoracic lymph duct fistulas. Approximately 4% of the label was recovered in the lymph collected during 48 h. It was concluded that lymph duct drainage is ineffective in the elimination of orally administered digitoxin and digoxin.

Using the isolated spontaneously pulsating portal vein of the mouse, it was shown that benzoctamine differs from chlorpromazine and imipramine with regard to its effect on pulsatile frequency and excursions (amplitude).

Using various models of increased vascular permeability (turpentine-induced pleurisy, kaolin-, glass and carrageenin-elicited paw oedema) it was shown that benzoctamine is capable of antagonizing the permeability disturbances occurring in the early phase of inflammatory reactions. The compound does not exert an anti-arthritic action.
