Summary: The cytology of multinucleated spermatocytes and spermatid giant cells in the seminiferous tubules of men with oligozoospermia and of men older than 65 years has been investigated electron microscopically. Two different processes which are responsible for the origin of multinucleated germ cells have been analysed:1. Defects of the intercellular bridges (IB): they move apart and thus allow the confluence of one clone's cells to symplasms.2. The confluence of membranes: within one clone the membranes of neighbouring germ cells are dissolved and thus intercellular bridges are found in the cytoplasm of the resulting giant cells.The spermatid giant cells reveal a new organization of the cell nuclei and the cell or‐ganelles. Yet they disintegrate in the lumen of the seminiferous tubules. The appearance of giant cells therefore is an expression of the germ cell degeneration.Vielkemige Spermatozyten und Spermatiden in menschlichen SamenkanälchenZusammenfassung: Die Zytologie von mehrkemigen Spermatozyten und Spermatiden‐riesenzellen in Samenkanälchen von Männem mit Oligozoospermie und bei Männern, die älter als 65 Jahre sind, wird elektronenmikroskopisch untersucht. Für das Zustandekom‐men der mehrkemigen Keimzellen lassen sich zwei Vorgänge erkennen:1. Summary: Incubation of human spermatozoa of normozoospermic origin with the ga‐lactose specific lectin peanut agglutinin (PNA) resulted in agglutination of the majority of sperm. This finding enabled separation of sperm into two subpopulations. The major one, accounting for 87.0 ± 12.7 (SD)% consisted of agglutinating sperm (PNA+), while the minor subpopulation (PNA‐) did not bind the lectin. The correlation between the percentages of PNA‐sperm and acrosome‐damaged sperm, as detected by light microscopy prior to separation, was analysed and an overlapping has been found. Light and electron microscopy examination revealed that the PNA‐cells were either acrosomeless or possessed severely damaged acrosomes while the PNA+sperm were mostly free of such pathologies. Following treatment of sperm with fluorescin‐conjugated PNA the single cells lacked any labeling while the agglutinates exhibited fluorescence. However, the binding of the lectin to sperm was non‐uniform and differences in fluorescence intensity at various parts of the head have been observed.The sialic acid content in PNA+sperm was found to be significantly higher (7.5 ± 2.2 μg/108sperm) as compared to PNA‐sperm (2.1 ± 0.6 μg/108sperm). It was suggested that sialic acid content could be a biochemical indicator of acrosomal integrity.Separation of sperm by selective PNA agglutination might be valuable for elimination of sperm affected by acrosomal pathologies from the ejaculate.Zusammenfassung: Inkubieren von Spermatozoen des Menschen bei Normozoospermie mit dem Galaktose‐spezifischen Lektin Erdnuß‐Agglutinin (PNA) bewirkt bei der Mehrzahl der Spermatozoen eine Agglutination. Dieses ermöglicht die Trennung der Spermatozoen in zwei Subpopulationen. Die größere Gruppe (87.0 ± 12.7%) enthält die agglu‐tinierten Spermatozoen (PNA+), während die kleinere Gruppe kein Lektin bindet (PNA‐). Die Korrelation zwischen PNA‐Spermatozoen und Spermatozoen mit Akrosomdefekten wurde im Lichtmikroskop analysiert und erne Überlappung festgestellt. Licht‐ und Elek‐tronenmikroskopuntersuchungen ergaben, daß die PNA‐Zellen entweder kein Akrosom oder schwere Akrosomdefekte zeigten, während die PNA+Spermatozoen in der Regel keinen pathologischen Befund aufwiesen. Nach Behandlung mit Fluoreszin‐konjugiertem PNA zeigten die einzelnen Zellen keine Markierung, jedoch wiesen die Agglutinate Fluo‐reszenz auf. Die Bindung der Spermatozoen an Lektin war nicht einheitlich und es konn‐ten Unterschiede in der Fluoreszenzintensität in den Spermatozoenköpfen beobachtet werden. Der Gehalt der im Speichel vorkommenden Säuren war in PNA+Spermatozoen signifikant höher (7.5 ± 2.2 mcg/108Sperm.) als in PNA‐Spermatozoen (2, 1 + 0,6 meg/ 108Sperm.). Der Gehalt an Säuren des Speichels könnte als Indikator für die Akrosomin‐tegrität gelten. Die Trennung der Spermatozoen durch PNA‐Agglutination könnte für die Elimini

Summary: The topographical distribution pattern of the stages of the murine seminiferous epithelium cycle was investigated.PAS‐hematoxylin stained testicular sections from adult mice representative of the apical, equatorial and caudal region of both testes, were used. The relative frequencies (RF) of the stages of the seminiferous epithelium cycle was estimated on the basis of more than 10,000 cross‐sectioned seminiferous tubules identified according to the criteria of Leblond and Clermont (1952). It was found that in the testicular sections the stages of adjacent seminiferous tubules are not distributed randomly. The comparison of the RF of the stages (calculated over all the testicular sections) with the RF of the stages that are adjacent to a given seminiferous tubule stage suggests a clustered occurrence of numerically identical stages. These comparisons very often show statistically significant differences. The finding of such associations among adjacent segments of seminiferous tubules (stages) suggest the existence of an ordered distribution of the seminiferous tubules inside the testis possibly controlled by substances with local control capacity of spermatogenesis.On the basis of the findings here presented, it is suggested an interpretation of the phenomenon of modulations of the waves of the seminiferous epithelium.Die unterschiedlichen Lokalisationen der verschiedenen Spermatogenesestadien im Maushoden:Morphologische Beweise ihrer nicht‐zufälligen VerteilungZusammenfassung: In der vorliegenden Arbeit wurde das topographische Verteilungs‐muster der verschiedenen Stadien des Keimepithelzyklus untersucht. Hierzu wurden PAS‐Hämatoxylin‐gefärbte Hodengewebsschnitte von erwachsenen Mäusen aus der apikalen, äquatorialen und caudalen Region von beiden Hoden untersucht. Die relative Häufigkeit der verschiedenen Stadien der Spermatogenese wurde an mehr als 10.000 Samenkanäl‐chen (cross‐sections) entsprechend den Kriterien von Leblond und Clermont (1952) be‐stimmt.Es konnte nachgewiesen werden, daß in den Gewebeschnitten die Stadien benachbar‐ter Samenkanälchen nicht zufällig verteilt sind. Bei Vergleich der relativen Häufigkeit der verschiedenen Stadien, bezogen auf alle Gewebeschnitte des gesamten Hodens mit der relativen Häufigkeit der Stadien in den angrenzenden Kanälchen ergibt sich ein gehäuftes Vorkommen numerisch identischer Stadien.Dieser Befund ist oftmals statistisch signifikant. Der Nachweis solcher Beziehungen zwischen benachbarten Segmenten läßt eine geordnete Verteilung der Spermatogenese‐Stadien innerhalb des Hodens vermuten, die möglicherweise durch Substanzen mit lokaler Kontrollkapazität auf die Spermatogenese gesteuert werden.Die Befunde legen eine Deutung für das Phänomen des we

Summary: Addition of an optimum level of caffeine (7 mM) during preservation (5° C) of buffalo semen in EYC medium had a stimulatory effect on sperm motility. It increased the preservation time of buffalo semen by delaying the loss of sperm motility. Immotile buffalo sperm due to cold shock could also be mobilised by addition of caffeine.Zusammenfassung: Das Zugeben von Coffein (7 mMol) während der Konservierung von Büffelspermatozoen bei 5° C in EYC Medium (Ei‐Dotter‐Citrat) bewirkt eine Stimulation der Spermatozoenmotilität. Summary: A simple method for collection and measurement of the progressive motility of spermatozoa was developed. The procedure consists of the spectrophotometric measurement of the increase in optical absorbance produced by the upward migration of spermatozoa in a high density medium of Ficoll 400. The suspension containing migrated spermatozoa may be used as a purified population of uniformly motile and undamaged spermatozoa.Zusammenfassung: In einer experimentellen Untersuchung wird eine einfache nephelometrische Methode zur Motilitätsbestimmung der progressiv‐beweglichen Spermatozoen und zur Sammlung der gut beweglichen Spermatozoen entwickelt. Das Verfahren besteht in einer spektrophotometrischen Messung des Anstiegs der optischen Absorption, die durch die Aufwärtswanderung der Spermatozoen in einem Ficoll‐Medium 400 erfolgt. Summary: This investigation was undertaken to study the potassium and sodium concentrations in rete testis fluid and cauda epididymal plasma and spermatozoa following gossypol treatment. Rats were treated orally with gossypol acetic acid at a dose of IS mg/ kg/day for 3 and 6 weeks respectively. There were not abnormal findings in the ionic concentrations of cauda epididymal plasma following 3 or 6‐week‐treatment. After 6 but not 3 weeks the spermatozoa in the cauda epididymidis were rendered almost completely im‐motile and malformed with a significant reduction in their potassium concentration and rise in their sodium concentration. No abnormal findings in the morphological picture of the germinal epithelium were observed under such circumstances and fluid and sperm from the rete testis were normal. Therefore, a direct action of gossypol may be exerted locally on the spermatozoa in the epididymis.Zusammenfassung: Die Untersuchung wird zwecks Kalium‐ und Natriumkonzentration in der Rete‐Testis‐Flüssigkeit, im Plasma des Nebenhodenkopfes und in den Spermatozoen unter Gossypol‐Therapie vorgenommen. Hierzu wurden Ratten mit Gossypolessig‐säure (15 mg/kg/die für 3 bzw. 6 Wochen) gefüttert. Es ließen sich keine von der Norm abweichenden Ergebnisse bei den Elektrolyten im Nebenhodenkopf nachweisen. Ledig‐lich nach 6 Wochen Therapie ergab sich eine totale Unbeweglichkeit der Spermatozoen im Nebenhodenkopf bei signifikanter Reduzierung der Kaliumkonzentration und gleich‐zeitigem Anstieg des Natriums. Unter diesen Bedingungen ergab sich weiterhin kein von der Norm abweichendes Resultat in der Morphologie des Keimepithels; Flüssigkeit und Spermatozoen aus dem Rete testis waren normal. Es dürfte sich deshalb bei der Wirkung von Gossypol um einen direkten Einfluß lokaler Art auf die Sperma

Summary: Seminal plasma β‐human chorionic gonadotropin (β‐HCG) levels were determined in 254 semen samples collected from fertile and suspected subfertile men, and their relationships with seminal characteristics and spermatozoal fertilizing capacity were evaluated. Radioimmunoassayable β‐HCG, as defined by>5 mlU/ml concentration, was demonstrated in only 44.5% of all the samples studied. β‐HCG concentrations were similar in normospermic, oligospermic and azoospermic samples. There were no significant differences in β‐HCG concentration between high‐motility and low‐motility samples, between normal‐morphology and abnormal‐morphology samples, and between high‐fertilizing capacity and low‐fertilizing capacity samples. No significant correlations between the concentrations of β‐HCG and individual parameters of semen analysis and spermatozoal fertilizing capacity among the various groups of samples were observed. The physiological role of seminal plasma β‐HCG is presently uncertain and the relationship of its determination to male fertility requires further investigation.Zusammenfassung: In 254 Ejakulaten fertiler und subfertile Männer wurden im Seminalplasma die Konzentrationen von humanem Choriongonadotropin(4bT‐HCG) gemessen und bezüglich der Rate befruchtungsfähiger Spermatozoen ausgewertet. In nur 44,5% al‐ler Ejakulate wurden durch Radioimmunassaybestimmung β‐HCG Werte von über 5 mlU/ ml gefunden. Bei Normozoospermie, Oligozoospermie und Azoospermie wurden vergleich‐bare β‐HCG Konzentrationen festgestellt. Es fanden sich keine signifikanten Unterschiede der β‐HCG Konzentration zwischen Ejakulaten mit hoher und geringer Spermatozoenmo‐tihtät, normaler und pathologischer Motilität und hohem und geringem Fertilisierungs‐grad der Spermatozoen. In den einzelnen Gruppen konnten kerne signifikanten Korrela‐tionen zwischen β‐HCG Werten, individuellen Spermiogrammparametern und Fertilität beobachtet werden.
