SummaryThe bovine seminal plasma is formed mainly by secretions of epididymis and the glandular epithelia in ampulla, seminal vesicles, prostate and Cowper's glands. The contribution of each organ to the hydrolytic enzyme activities (glycosidases, exopeptidases, phospholipases) of the bull seminal plasma has been analyzed and is reviewed in this paper with special emphasis on the role of the accessory glands. Seminal vesicles seem to have a major role in the secretion of seminal plasma acid α‐glucosidase, acid α‐mannosidase and β‐N‐acetylhexosaminidase, aminopeptidase A, dipeptidyl peptidase II and IV and γ‐glutamyl transpeptidase as well as Ca2+‐dependent and Ca2+‐independent phospholipases A2with distinct substrate specificities, a choline‐specific phospholipase C and a Co2+(Mn2+)‐activated sphingomyelinase. The enzyme pattern in the ampulla closely resembled that of the seminal vesicles and obviously contributes to the seminal plasma level of these hydrolases. The bull prostate and Cowper's glands contained a strong Ca2+‐dependent phospholipase A2activity. However, these glands may not contribute to the seminal plasma PLA2activity. At ejaculation the epididymal spermatozoa are exposed to these enzymes. They may have a specific affinity to sugar, peptide or phospholipid residues at distinct sites of the sperm surface. These enzymes may also participate in the digestion of various other semen components to create a suitable milieu for the emitted spermatozoa.ZusammenfassungDas Seminalplasma des Bullen wird hauptsächlich von den Sekreten des Nebenhodens und der Drüsenepithelien der Ampulle, Bläschendrüse, Prostata und Cowperschen Drüse gebildet. Der Beitrag eines jeden Organs zu den hydrolytischen Enzymaktivitäten (Glycosidasen, Exopeptidasen, Phospholipasen) des Bullen‐Seminalplasmas wurde analysiert und in diesem Übersichtsartikel besonders im Hinblick auf die Rolle der akzessorischen Geschlechtsdrüsen dargestellt. Die Bläschendrüsen haben offensichtlich eine größere Bedeutung für das Seminalplasma bei der Sekretion der α‐Glucosidase, sauren α‐Mannosidase und β‐N‐Acetylhexosaminidase, Aminopeptidase A, Dipeptidylpeptidase II und IV und der γ‐Glutamyltranspeptidase sosie Ca2+‐abhängigen und Ca2+‐unabhängigen Phospholipasen A2mit verschiedenen Substratspezifizitäten einer Cholin‐spezifischen Phospholipase C und einer Co2+(Mn2+)‐aktivierten Sphingomyelinase. Das Enzymmuster der Ampulle ähnelt dem der Bläschendrüse sehr stark und trägt offensichtlich zum Seminalplasmaspiegel dieser Hydrolasen bei. Die Bullenprostata und Cowpersche Drüse enthalten eine starke Ca2+‐abhängige Phospholipase A2‐Aktivität. Diese Drüsen scheinen jedoch nichts zur PLA2‐Aktivität des Seminalplasmas beizutragen. Bei der Ejakulation werden die Nebenhodenspermien diesen Enzymen ausgesetzt. Sie dürften eine spezifische Affinität für Zucker, Peptide oder Phospholipidreste an be‐stimmten Domänen der Spermienoberfläche besitzen. SummaryThe secretions of rat seminal vesicles from the so‐called copulatory plug when getting in contact with the secretions of the coagulating gland. Depending on the sexual activity of the respective animal and the extraction conditions, the protein pattern of the seminal vesicle secretion varies to some extent. We have studied the biochemical properties of the proteins SVS I, SVS II, sulfhydryl oxidase and SVS III‐VIII. The most interesting protein is SVS II which is the main substrate of coagulating gland transglutaminase and serves as the most important monomer during semen coagulation. It is highly soluble in acidic solutions. The monomeric polypeptide has a molecular mass of 49 kDa, is glycosylated with fucose, glucose, mannose and N‐acetylated sugars and has a highly basic pI of 10.5. Particularly interesting are its functional and structural relationships with actin. It is the first described protein with actin modulating properties that is secreted in an androgen‐dependent manner.ZusammenfassungDas Sekret der Rattenbläschendrüse bildet den sogenannten Kopulationspfropf, wenn es mit dem Sekret der Koagulationsdrüse in Kontakt kommt. Je nach sexueller Aktivität des entsprechenden Tiers und den Extraktionsbedingungen variiert das Proteinmuster des Bläschendrüsensekrets in gewissen Bereichen. Wir haben die biochemischen Eigenschaften der Proteine SVS I, II, Sulfhydryloxidase und SVS III‐VIII untersucht. Das interessanteste Protein ist SVS II, welches das wesentliche Substrat der Transglutaminase der Koagulationsdrüse bildet und das den bedeutendsten Monomeranteil bei der Samenkoagulation darstellt. In saurem Milieu ist es sehr gut löslich. Das monomere Polypeptid hat ein Molekulargewicht von 49 kDa, ist mit Fukose, Glukose, Mannose und N‐acetylierten Zuckern glykosyliert und hat einen pI von 10, 5. SummaryIn bull seminal plasma 5′‐nucleotidase is present in heterogeneous forms. The heterogeneity is abolished by treatment of bull seminal plasma with the detergent sodium cholate. The purified enzyme, which is a glycoprotein, shows an apparent molecular mass of 160 kDa on gel filtration in the presence of 50 mmol sodium cholate and an apparent molecular mass of 72 kDa upon SDS/polyacrylamide‐gel electrophoresis. The 5′‐nucleotidase of bull seminal plasma is a metalloprotein containing 2 zinc ions per molecule of dimeric protein. The removal of the two zinc ions from the protein results in a completely inactive apoenzyme. The substitution of the endogenous zinc with Co(II) or Cu(II) produces a holoenzyme which is slightly activated in the case of Co(II), whereas, in the case of Cu(II) only 65% of the initial activity is recovered. The enzyme has a covalently attached glycosyl‐phosphatidylinositol moiety which can be removed by treatment with phosphatidylinositol‐specific phospholipase C. ESR studies have indicated a radius of 35 Å for the protein and that Cu(II) binds to the metal‐free enzyme to a site in which sulphur donors can be excluded.ZusammenfassungIm Seminalplasma des Bullen kommt die 5′‐Nucleotidase in heterogener Form vor. Diese Heterogenität verschwindet, wenn das Seminalplasma mit dem Detergens Natriumcholat behandelt wird. Das gereinigte Enzym, ein Glykoprotein, zeigt nach Gelfiltration in Gegenwart von 50 mmol Natriumcholat ein apparentes Molekulargewicht von 160 kDa und von 72 kDa nach SDS‐Polyacrylamid‐Gelelektrophorese. Die 5′‐Nukleotidase des Bullenseminalplasmas ist ein Metalloprotein, das 2 Zinkionen pro Molekül des dimeren Proteins enthält. Die Entfernung der beiden Zinkionen von dem Protein führt zu einem vollständig inaktiven Apoenzym. SummarySomatic cell hybrids were made from mouse myeloma cells and spleen cells derived from BALB/c mice immunized with homogenized epithelial fractions of BPH. The screening by immunoperoxidase staining on human prostate and non‐prostate tissue resulted in one monoclonal antibody identifying a prostate specific antigen. Upon SDS‐PAGE and Western blot this antigen exhibited a single band at the position of 34 kDa molecular weight. The immunoreactivity of the prostate antigen was found to be localized excluxsively in the epithelial lining of ducts and secretions of normal prostate, BPH and prostate cancer. Anti‐p34 antibody reacted with an antigenic determinant on the PSA molecule and inhibited the binding of Anti‐PSA antibody to PSA by about 80 to 90% in the RIA.ZusammenfassungDie Lymphozyten‐Hybridisierung erfolgte zwischen Myelomzellen und Milzzellen immunisierter BALB/c‐Mäuse. Zur Immunisierung wurde eine homogenisierte Epithelfraktion aus BPH‐Geweben verwandt. Das Screening‐Verfahren erfolgte immunhistologisch von Prostata und nicht‐prostatischen Organen. Mit dieser Technik konnte ein monoklonaler Antikörper identifiziert werden, der ein Prostata‐spezifisches Antigen erkennt. SummaryThe synthetic androgen mibolerone elicits a set of distinct changes in the behaviour of an androgen responsive human prostatic carcinoma cell line (LNCaP). Inhibition of cell proliferation, induction of morphological change and of a prostate specific mRNA, and inhibition of colony formation in soft agar are induced by very low concentrations of mibolerone. The natural androgen dihydrotestosterone is much less effective. The changes in growth characteristics and morphology are reverted by excess antiandrogen, i.e. cyproterone acetate or hydroxyflutamide. Cell lines lacking androgen receptors (PC‐3, DU 145 and MRC‐5) are completely unresponsive to mibolerone. Taken together, our results indicate androgen receptor mediated suppression of the transformed phenotype in LNCaP cells.ZusammenfassungDas synthetische Androgen Miboleron löst eine Reihe bestimmter Veränderungen im Verhalten einer Androgen‐responsiven menschlichen Prostata‐Karzinom‐Zellinie (LNCaP) aus. Hemmung der Proliferation, Induktion morphologischer Veränderungen und einer Prostata‐spezifischen mRNA, sowie die Hemmung der Kolonie‐Bildung im Weichagar werden von niedrigen Konzentrationen von Miboleron hervorgerufen. Das natürliche Androgen Dihydrotestosteron ist viel weniger effektiv. Die Veränderungen im Wachstumsverhalten und der Morphologie werden durch zusätzliches Antiandrogen, z.B. Cyproteronazetat oder Hydroxyflutamid, rückgängig gemacht. Zellinien ohne Androgenrezeptor (PC‐3, DU 145 und MRC‐5) sprechen auf Miboleron überhaupt nicht an. SummaryThe rat ventral prostate synthetizes and secretes a number of relatively abundant proteins, such as Prostatic Binding Protein (PBP), the Proline‐Rich Polypeptides (PRP), the 20 or 22‐kDa protein and a prostatic protease, which respectively constitute approx. 50, 5, 10 and 2% of the secreted protein. The synthesis of these proteins is androgen‐regulated and this is also the case for the concentration of the corresponding mRNAs. The existence of a putative androgen response element in the first intron of the PBP‐C3 gene supports the possibility of a direct control of its transcription by androgen receptor complexes, but there is also evidence for the requirement of protein synthesis for mRNA stimulation. Finally, changes of tissue composition and cell‐cell interactions between epithelial and stromal cells most probably play an important role in the global effect of androgens on the rat ventral prostate.ZusammenfassungDie Rattenprostata synthetisiert und sezerniert eine Reihe von relativ vorherrschenden Proteinen wie Prostatic Binding Protein (PBP), Proline‐Rich Polypeptides (PRP), das 20 oder 22 kDa Protein und eine Prostata‐Protease, die je ungefähr 50, 5, 10 und 2% des sezernierten Proteins ausmachen. Die Synthese dieser Proteine ist Androgen‐reguliert und dies trifft auch für die Konzentration der korrespondierenden mRNAs zu. SummarySeminal vesicle secretion contributes significantly to the proteins of bovine seminal plasma. The following proteins from bull seminal vesicle were isolated and characterized: major protein (PDC 109), the basic proteins BUSI II, RNAse BS1, protein P6 and seminal antimicrobial protein (SAP). Using antibodies against the proteins BUSI II, RNAse BS1, SAP and major protein, the seminal vesicle epithelium was identified as the source of the respective antigens. The biosynthesis of bovine seminal vesicle secretory proteins was studied by cell free translation of poly (A)‐RNA from seminal vesicles and the respective mRNAs were characterized by cDNA cloning. Recombinant clones (103) of a cDNA library of bull seminal vesicle poly (A) + RNA were screened by colony hybridisation using radioactively labelled synthetic probes. The respective clone containing the longest cDNA insert was sequenced. In case of major protein the Mr of the 134 amino acid residue precursor polypeptide was 15 480 as deduced from direct mRNA sequencing. The precursor sequence of 25 amino acid residues has a hydrophobic character and very likely constitutes a signal peptide, directing the protein towards the secretory pathway. The deduced amino acid sequence contained no consensus sequence indicative of N‐glycosylation.ZusammenfassungDas Bläschendrüsensekret des Bullen trägt wesentlich zum Proteinmuster des Seminalplasmas bei. Die folgenden Bläschendrüsenproteine wurden isoliert und characterisiert: Major protein (PDC 109), die basischen Proteine BUSI II, RNAse BS1, Protein P6 und das seminal antimicrobial protein (SAP). Mit Antikörpern gegen BUSI II, RNAse BS1, SAP und major protein wurde das Bläschendrüsenepithel als der Syntheseort dieser Proteine identifiziert. Die Biosynthese des Bläschendrüsenprotein des Bullen wurde durch zellfreie Translation von Poly(A)+RNA aus Rinderbläschendrüse untersucht und die entsprechenden mRNAs durch cDNA‐Klonierung charakterisiert. Rekombinante Klone (103) einer cDNA‐Bibliothek aus Rinderbläschendrüsen Poly(A)+RNA wurden durch Kolonie‐Hybridisierung mit radioaktiv markierten synthetischen Sonden gescreent. Der Klon mit dem längsten cDNA‐Insert wurde sequenziert. Im Falle von major protein betrug die molekulare Masse des 134 Aminosäurereste großen Vorläuferpolypeptids 15 480 nach Ausweis der direkten mRNA‐Sequenzierung. Die Prekursor‐Sequenz von 25 Aminoacylresten ist hydrophob und stellt höchstwahrscheinlich das Signalpeptid dar, das das Protein in den Protein in den Sekretionsweg lenkt. SummaryPolyamines (putrescine, spermidine and spermine), secreted by the prostate gland, occur in high concentrations in the seminal fluid of many species, including man and rat. The physiological significance of seminal polyamines is still obscure, however. It has been postulated that polyamines may serve as amine donor substrates for transglutaminases (TGases), enzymes catalyzing protein crosslinking by the formation of γ‐glutamyllysine or bis(γ‐glutamyl)polyamine cross‐bridges. We have analyzed TGase‐activities and polyamine content of the various rat prostate lobes and rat seminal vesicles. Highest TGase activities were observed in the coagulating gland (anterior prostate) and the dorsolateral prostate, whereas very little TGase activity was present in the ventral prostate gland. In contrast, polyamine concentrations were highest in the ventral prostate but low in coagulating glands. Seminal vesicles, and in particular seminal vesicle secretions, contained low polyamine levels and intermediate TGase activity. Levels of protein‐bound polyamines, not extractable by perchlorid acid, did not correlate with TGase‐activities in ventral prostate and coagulating glands, suggesting an extracellular rather than intracellular function of prostatic TGase and polyamines. The observation that Km‐values of rat prostate TGase for all three polyamines (57–120 μM, using N,N‐dimethylated casein as protein substrate) were well below seminal polyamine concentrations is compatible with a regulatory role of polyamines in the process of seminal clot formation.ZusammenfassungPolyamine (Putrescin, Spermidin und Spermin), von der Prostata sezerniert, werden in hohen Konzentrationen in der Samenflüssigkeit vieler Spezies, Mensch und Ratte eingeschlossen, angetroffen. Die physiologische Bedeutung ihrer Anwesenheit im Samen ist aber unklar. Man hat postuliert, daß Polyamine als Amingruppen‐Donoren für das Enzym Transglutaminase (TGase) auftreten können, welches seinerseits cross‐linking von Proteinen mit Hilfe von γ‐Glutamyl‐Lysin oder bis(γ‐Glutamyl)‐Polyamin‐Brücken katalysiert. Wir haben die Aktivitäten von TGase sowie die Polyamin‐Gehalte in den verschiedenen Lappen der Rattenprostata und in den Samenblasen bestimmt. Die höchsten TGase‐Aktivitäten wurden in der Koagulationsdrüse und im dorsolateralen Prostatalappen aufgefunden, während die ventralen Prostatalappen nur wenig Aktivität aufwiesen. Im Gegensatz dazu wurden die höchsten Polyamin‐Spiegel in der ventralen Prostata und die niedrigsten in der Koagulationsdrüse gefunden. Die Bläschendrüsen, und insbesondere ihre Sekrete, enthielten niedrige Polyamin‐Konzentrationen und eine mäßige TGase‐Aktivität. Der Gehalt an proteingebundenem Polyamin (nicht extrahierbar mit Perchlorsäure) korrelierte nicht mit den TGase‐Aktivitäten der ventralen Prostatalappen und Koagulationsdrüsen. Dieser Befund läßt vermuten, daß Prostata‐TGase und ‐Polyamine ihre Funktion überwiegend extrazellulär ausüben.
