AbstractThe effect of mercuric chloride on akinete germination and sporulation with respect to time lapse and extent was studied inPithophora oedogonia(Mont.) Wittrock (Cladophorales, Chlorophyceae). At HgCl2concentrations from 1.25 to 45 mg/l akinetes showed progressive delay in the initiation and reduction in the extent of germination. HgCl2concentrations of 60 mg/l and above proved to be completely lethal to the spores as they failed to germinate. The time necessary for the initiation of akinete formation and the percentage sporulation also decreased with an increase in the concentration of HgCl2from 1.25 to 45 mg/l where spore germination was encountered. Inactivation of enzymes and metabolic inhibition seemed to be the cause of spore lethality.

AbstractThe localization of hydrolytic enzymes, phosphatase, esterase, lipase and palmitoyl‐CoA hydrolase was analysed in the cytosol, cytoplasmic membrane, periplasmic fraction, outer membrane and culture supernatant in dependence on the growth rate of the bacteria.The unspecific phosphatase was found to be a cytosolic enzyme.A lipase was the only extracellular enzyme detected. The results pointed to a secretion of the lipase into the culture medium via cytoplasmic and outer membrane.The palmitoyl‐CoA hydrolase was found to be attached to the outer membrane, but activities were also detected in the periplasmic fraction.Unspecific esterolytic activities were mainly measured in the cytosol and in the cytoplasmic membr

AbstractTransferable productive lysis in stable protoplast type L‐form cells ofBacillus subtiliswas producted by 6 phages out of 14 strains virulent for the parentB. subtilis170 and 1997. Most of these Phages lytie for L‐forms show the ϕ 29 morphology characteristic for the smallestB. subtilisPhages containing double‐stranded DNA. Among 31 actinophages, 23 of which were virulent forStreptomyces hygroscopicus, only SLE 109 and ϕc 31 gave productive infection of the stable protoplast type L‐form ofS. hygroscopicusNG 33–354.Electron microscopic investigation and treatment by DNAse demonstrated that infection of L‐forms cells is an adsorption‐injection process, and that it is not caused by transfection of free phage DNA or endocytotic uptake of phage particles. Because in both stable L‐forms cell wall biosyntheesis blocked irreversibly the results allow the conclusion that specific receptors must be localized in the cyroplasmic membrane for those phages producing transferable lysis in protoplast type L‐forms. AbstractIn order to determine the localization of DNA‐synthesis inStreptomyces granaticolorandStreptomyces hygroscopicus, mycelia (growing either on agar or in liquid medium) were pulse‐labelled with3H‐thymidine and prepared for autoradiography. The distribution of silver grains showed no regions of preferential incorporation of3H‐thymidine in mycelia up 300 μm in length. Since mycelia grow by apical elongation of hyphae, the frequency of silver grains was quantitatively analysed along individual main hyphase. No significant difference of labelling was found within zones of different age up to a distance of 80 μm from the hyphal tip. AbstractGenetic studies of several events of the life cycle ofY. lipolyticademonstrated that diploid strains were unstable and produced mitotic segregants by haploidization.
