摘要:
The incidence of positive ethanol gelation test (EGT) after addition of brinase (a proteolytic enzyme preparation from Aspergillus oryzae) to anticoagulated and non-anticoagulated human plasma was studied. In vitro addition of brinase to plasma causes positive EGT, and the incidence is dose-dependent. In plasma from warfarin-treated patients and/or after addition of heparin to plasma, prior to the addition of brinase, a significantly reduced incidence of positive EGT is observed. The incidence is lowest after heparin. Gels formed in the presence of heparin are easier susceptible to enzymatic degradation than those formed in the absence of heparin.

Relationships between Normotest, prothrombin time, albumin, γ-globulin, GPT, and the hepatic fractional clearance of 198Au-colloids as a measure of effective hepatic blood flow were studied by correlation analysis, in 50 patients with liver diseases, including chronic hepatitis or cirrhosis. Simple correlation coefficients were highly significant between Normotest, prothrombin time, albumin, and the heaptic fractional clearance; however, not between GPT or γ-globulin and the other parameters. Further correlation analysis revealed a highly significant partial correlation coefficient between Normotest and albumin and the hepatic clearance; however, not between Normotest and albumin. As effective hepatic blood flow is considered to be proportional to the liver parenchymal volume, the above results indicate that vitamin-K-dependent clotting factors, as well as albumin, reflect the liver cell mas

Antithrombin III activity was determined by three techniques in 58 patients with maturity onset diabetes mellitus: a spectrophotometric assay, using the chromogenic substrate Chromozym TH; evaluation of antifactor Xa activity and an immunochemical technique. Although the biological antithrombin III activity was clearly reduced in individual subjects, there was only a slight nonsignificant reduction in the mean antithrombin III activity values as compared to normal subjects. No difference was observed between diabetics receiving insulin and subjects taking oral hypoglycemic agents or between diabetics without vascular complications and diabetics with vascular lesions.

Human blood platelets were found to carry the complete pathway of de novo purine nucleotide synthesis. The rate of purine synthesis was gauged by the rate of incorporation of precursor (14C)formate into purines. The effect on formate incorporation of several compounds known to inhibit purine synthesis de novo was studied. Adenine, orotic acid and azaserine inhibited purine synthesis, but hypoxanthine and allopurinol did not. Platelet content of phosphoribosylpyrophosphate (PRPP) and of ribose-5-phosphate (R-5-P) was found to be similar to that in human blood leukocytes and erythrocytes. Incubation of intact platelets with high inorganic phosphate concentrations caused an increase in platelet PRPP content but did not affect R-5-P content or the rate of purine synthesis de novo.

The quantity of myosin, actin, and actin-binding protein (ABP) in platelets and platelet fractions of 5 patients with Glanzmann’s thrombasthenia were compared to those of normal individuals. No significant differences were observed between the average amounts of these contractile proteins in unfractionated platelets or platelet membrane obtained after osmotic shock. 摘要:
The functional exclusion of the liver, obtained with end-to-side anastomosis of the porta into the cava vein results in impairment of platelet activity. In fact the maximal amplitude and velocity of platelet aggregation in vitro are reduced in animals which undergo the operation and are examined 1 month later. Adding fibrinogen to the platelet-rich plasma after portacaval anastomosis restores a normal type of aggregation indicating that the lack of this factor is involved in the phenomenon, even if alterations occurring to platelets may be implied, too.

Platelet aggregation induced by ADP and collagen was studied in 174 diabetic patients before entering a controlled clinical trial. The clinical characteristics have been precisely defined. The degree of retinal abnormality was assessed by fundus angiofluorog-raphy in every patient. Most patients had background retinopathy; the remainder were normal. All results were computer analysed. The only significant relationship observed was between platelet sensitivity to ADP which increases regularly in relation to age in both insulin- and non-insulin-treated diabetic patients with or without background retinopathy. Spontaneous platelet aggregation was observed in 13 subjects and was not correlated to the existence of a neuropathy or any other clinical characteristics.

摘要:
Histochemically studied, the heart, spleen, liver, kidney cortex, lung, sciatic nerve, gastrocnemius muscle, skin and tongue showed a higher fibrinolytic activity (plasminogen activator activity) in the newborn than in the adult rat. In the medulla of the kidney the activity was slightly lower in the newborn than in the adult. The plasmin inhibition was lower in the newborn than in the adult rat in all organs studied.
