AbstractThe effects of ageing on the numbers of alveolar pores of Kohn and on the cytoplasmic components of alveolar type II cells were studied in monkey lungs by scanning and transmission electron microscopy. Lung tissue from 26 female and three male pigtail macaques whose ages ranged from 1 month to 31 years (life span is 35 years) was analysed. From the age of 1 month to 10 years there was a significant increase in the number of alveolar pores (r= 0·85,p<0·001); however, between the ages of 14 years to 31 years there was no significant change. In seven animals ranging in age from 1 month to 4 years (mean 2·4 years) the number of pores was 5·8 ± 3·9 (mean · S.D.), whereas in 10 animals aged 16 to 31 years (mean 20·3 years) the number of pores was 32·7 ± 17·5 (mean ± S.D.) per alveolar profile, a significant difference (p<0·002). In older animals (15–20 years) there was a significant decrease, both in the number of lamellar bodies per alveolar type II cell (p<0·01) and in the volume density of lamellar bodies to cytoplasmic volume (p<0·05) compared with young animals (1 month to 4·8 years). In older animals, there was also a significant increase in the volume density of a vacuole‐like dilatation of the endoplasmic reticulum in alveolar type II cells (p<0·05) compared with young animals. These findings suggest impaired pulmonary surfactant production with aging. AbstractImmunohistochemical staining of formalin fixed paraffin sections with the mouse monoclonal antibodies E29 and CAM 5·2 effectively distinguish epithelium (antigen positive) from stroma (antigen negative) in normal endometrium. These antibodies were used as epithelial markers in the study of eight malignant mixed Müllerian tumours (MMT) of endometrium and gave normal reactions with well differentiated neoplastic glands; in contrast, negative or very weak staining was observed in poorly differentiated epithelial cells, present in large numbers in three cases. Abnormal antigen‐containing cells were observed in the stroma of seven MMT. AbstractUsing the well‐differentiated human squamous carcinoma cell line LICR‐LON‐HN‐5 as an immunogen we have produced a monoclonal antibody (32a) that reacts with the keratohyalin granular layer of the normal epidermis. We present here results showing the distribution of the epitope recognized by this antibody in human tissuesin vivoandin vitro, as demonstrated using immuno cytochemical staining techniques at the light and ultrastructural levels. Expression of the determinant first appears at 18 weeks of fetal development, localized in cells associated with the hair germ. In hyperplastic epidermis the staining pattern is altered, apparently linked with a switch from orthokeratotic to parakeratotic keratinization. In primary squamous cell carcinomas and in xenografts formed by the squamous carcinoma cell line LICR‐LON‐HN‐5 the keratinized elements are stained. Poorly differentiated tumours are not stained, indicating that antibody may be useful as a marker of terminal differentiationin vivo. When grown on collagen gels both human epidermal keratinocytes and the squamous carcinoma cell line show staining of the more differentiated cells which appears to be associated with the keratinohyalin granules, indicating that this antibody may be of value in studies aimed at the control of squamous di

AbstractThirty‐eight infiltrating ductal carcinomas, nine infiltrating lobular carcinomas, two tubular carcinomas and one papillary carcinoma were studied by light microscopy, immunocytochemistry and electron microscopy. Seventeen cases showed immunoreactivity for NSE. Immunostaining for different peptide‐hormones was observed in 12 of these 17 cases and in none of the 10 NSE‐negative cases used for controls. Scattered cells were positive for gastrin in five cases, pancreatic polypeptide in five, leu‐enkephalin in three, sub‐P in two, ACTH in one, bombesin in one and β‐endorphin in one case. Four cases revealed immunoreactivity for more than one peptide‐hormone. Typical neuroendocrine granules were seen in five cases (all positively stained for NSE). Small, electron dense granules of possible neuroendocrine nature were not found in any of the 33 NSE‐negative tumours. Our results confirm that immunoreactivity for NSE is present in a high proportion of breast carcinomas, but that neuroendocrine differentiation cannot be proved to be present in

AbstractJoint tissues from patients with rheumatoid arthritis (3), osteoarthritis (3) and normal healthy individuals (2) were examined in the transmission electron microscope and the morphological features of capillaries within 2 mm of the synovial membrane were quantified. Weibel‐Palade bodies (WPB) were significantly increased in capillary endothelial cells in rheumatoid and osteoarthritis compared with normal controls, when quantified by three different methods. WPB were positively correlated with both projections of the luminal plasmalemma and pinocytic vesicles in normal joint and some patients with rheumatoid and osteoarthritis. AbstractEpithelial membrane antigen was detected in normal glomeruli by a polyclonal antiserum to the antigen and by the monoclonal antibodies Ca 1, DAKO‐EMA and HMFG 2, but not HMFG 1, using an indirect immunoperoxidase method. The antigen was in the form of a thin ring or collar at the junction of glomerulus and tubule. In a series of 47 renal biopsies from patients with proteinuria, the antigen could still be seen in glomeruli, provided that there were adequate numbers of glomeruli in the sections. The main object of study was the glomerular tip lesion, in which tip adhesions were seen to be just adjacent to the patch of epithelial membrane antigen. This suggested that the antigen may be important in pathogenesis of the lesion. Normal proximal tubules did not express epithelial membrane antigen but it was detected on the luminal border of acutely damaged proximal tubules.
