AbstractAnalysis of serum immunoglobulins from patients suffering from autoimmune disease has shown that pathogenically relevant autoantibodies directed at organ specific antigens are light chain restricted, i.e., they are entirely lambda or kappa type in a given patient.1–3Furthermore, plasma cells involved at tissue level in the production of such antibodies, for example in Graves' disease, have also been shown to express a marked light chain bias as judged immunohistologically.4On the basis of these findings, a study was conducted to determine the light chain status of tissue plasma cell infiltrates associated with Sjögren's disease, a known autoimmune disease. Of the six cases examined, all six showed a marked lambda light chain bias, with two patients exhibiting a monotypic plasma cell infiltrate of IgA, lambda isotype. The significance of the overall observations is discussed in the context of other examples of light chain restricted B‐cell responses and the generally increased incidence of benign and malignant B‐cell neoplasia in autoimmune d

AbstractA quantitative study of glomerular basement membrane (GBM) changes in 16 cases of IgA nephropathy was carried out on two whole glomeruli per case using a wide‐field electron microscope. We found that GBM changes are observed more frequently than previously reported, although some changes are small and uneven in distribution. Changes include (a) attenuation, (b) lytic attenuation, (c) garland‐shaped widening of the GBM, (d) dome‐shaped widening of the GBM, and (e) disruption of the GBM. There was some correlation between the severity of light microscopic findings and the percentage of GBM affected by complicated changes ((b)–(e)). The percentage of complicated GBM changes correlated with the amount of proteinuria at biopsy. AbstractThe innervation of the pulmonary trunk and pulmonary arterial bed was studied in 17 pigs from birth to 6 months of age. After birth, the pulmonary trunk and extra‐and intra‐pulmonary arteries contained neurofilament and protein gene‐product‐immunoreactive nerve fibres in both the adventitia and media. The density of nerve fibres increased from birth to 2 months, this being most marked during the first 2 weeks of life. Most of the fibres in the media were presumed to be sympathetic in origin as they contained both neuropeptide tyrosine and tyrosine hydroxylase immunoreactivity. Fibres were associated with the vasa‐vasorum and vascular smooth muscle running around the vessel, between the elastic laminae and smooth muscle cells, in the outer two‐thirds of the media. Vasoactive intestinal polypeptide and calcitonin gene‐related peptide‐immunoreactive nerve fibres were found to be associated with the pulmonary trunk and extra‐pulmonary artery, but generally not with the intra‐pulmonary arteries. Tyrosine hydroxylase immunoreactivity was detected in the glomus cells at birth, but peptide immunoreactivity (enkephalin) was not demonstrated in paraganglia until 14 days of age. Adaptation to extra‐uterine life is associated with rapid development changes in the innervation of the pig pulmonary trunk, extra‐and intra‐pulmonary arteries and in the expression of peptide immunoreactivity in both nerve fibres and glomus cells.
