ABSTRACT—In order to determine if iron load was able to stimulate specifically ferritin synthesis and secetion in adult human hepatocytes, primary cultures were submitted to increasing concentrations of ferric iron. The following results were obtained: 1. iron incorporation within the hepatocytes increased as a function of culture time and reached a high level after 48 h of treatment; 2. a dose‐dependent significant stimulation of ferritin synthesis and secretion was observed when the medium iron concentration increased from 1 to 40 μmol · l‐1; 3. transferrin and albumin secretions were unaffected. ABSTRACT—The histological outcome in liver biopsies following 9 months of interferon alpha‐2b treatment was assessed in detail in 19 patients with chronic posttransfusion non‐A, non‐B hepatitis (PTH‐NANB) and compared with 12 untreated PTH‐NANB patients. Fourteen (74%) treated and 7 (58%) control patients were reactive for antibodies against hepatitis C virus (anti‐HCV). Liver biopsies taken before and after the 9‐month period were scored numerically for portal inflammation, piecemeal necrosis (PMN) and fibrosis, without knowledge of whether the specimens came from control or treated patients. There were no score differences in the initial biopsies between the treated and control group. In the follow‐up biopsies the treated group showed significantly less portal inflammation, PMN and fibrosis than the control group (p<0.05–0.01). When paired samples from the treated group were compared, significantly regressed portal inflammation, PMN and fibrosis were noted in the follow‐up biopsies (p<0.05–0.001). The presence or not of anti‐HCV antibodies in serum had no impact on the histological response to interferon treatment. ABSTRACT—When mononuclear phagocytes, including Kupffer cells, are activated by various agents, they synthesize and release cytokines such as interleukin 1 (IL‐1) and tumor necrosis factor (TNF). In this study, we examined the effect ofin vitroKupffer cell activation by recombinant murine interferon γ (IFNγ) on IL‐1 and TNF secretion. IFNγ enhanced TNF production in the presence or absence of lipopolysaccharide (LPS), but suppressed IL‐1 production by Kupffer cells. Because IFNγ also stimulated prostaglandin E2(PGE2) production, the effect of indomethacin, which is an inhibitor of cyclooxygenase and which inhibits PGE2biosynthesis, on IL‐1 and TNF production by Kupffer cells was examined. As a result, indomethacin enhanced TNF production by Kupffer cells, but had no effect on IL‐1 synthesis. ABSTRACT—Sera from 111 patients with chronic viral hepatitis and from 55 cases with other liver disorders were assayed for serum inhibitory factor. The prevalence of this immunosuppressive factor was very similar between chronic hepatitis B (61%), chronic hepatitis delta (57%) and chronic hepatitis C (68%). At the same time, serum inhibitory factor was never detected in the other disorders studied. The presence of this inhibitory factor was detected in a significantly higher percentage (p<0.05) of HBeAg, HBV‐DNA positive cases (75%) than in anti‐HBe positive, HBV‐DNA negative cases (44.4%). In chronic hepatitis delta, this immunosuppressive factor was also related to HDV‐RNA positivity. The detection of this serum immunosuppressive factor in chronic viral hepatitis and its association with a high viral replication level implies a possible role of this factor in the immune pathogenic mechanism in infectious viral

ABSTRACT—One hundred and seventeen ducklings, 42 inoculated with duck hepatitis B virus (DHBV) 2 days after hatching and 55 connatally infected, were studied over a 6‐month period in parallel with 20 ducklings without DHBV infection. Using immunohistochemical,in situand blot hybridization analyses, the natural course of hepatic and extrahepatic infection was examined. DHBV infection started in the liver 2–4 days post‐inoculation. There, DHBV was found not only in hepatocytes, but also in bile duct epithelial cells. Further, DHBV infection occurred in exocrine and endocrine pancreas (beginning 6–10 days and 20 days post‐inoculation, respectively) and in germinal centers of the spleen (beginning 8 weeks post‐inoculation). Occasionally viral DNA was also found in kidney glomeruli. Using strand‐specific RNA probes, viral DNA in pancreas and spleen was clearly demonstrated to be replicating intermediates. Hepatic and extrahepatic infection with DHBV was not associated with histologic inflammation or pathologic changes in these tissues or the liver. These data indicate that DHBV can infect cells other than hepatocytes.
