The discoloration of white rat hair by acid preparations has been employed as a model reflecting local tissue destruction by intravital fixation as contrasted to tissue dissolution. Nitrate reduction products in equilibrium with nitrite ion in moderately strong acid result in much more rapid tissue fixation than is accomplished by acid preparations alone. Parallel differences were found in the color reaction of normal skin to different acid preparations. The rapid fixing capacity demonstrated by these models may be useful for sharply localized destruction of topical lesions clinically.

The effect of nitric acid preparations on five different solid experimental tumors in mice was studied. None of these highly malignant tumors was eradicated by any of the acid preparations studied, although differences in reactivity were noted between preparations in some models.

Histologic and electron microscopic studies of Solcoderm-treated normal skin and human lesions were performed to help elucidate the mode of action of this new agent. Its fixative properties were readily demonstrated and generally proved to be adequate for histological diagnosis from the post-treatment scab. The degree of preservation of fine intra-cellular structures was particularly remarkable, since similar treatment with equivalent strengths of nitric acid alone destroys these substructures. Dermal lipids and collagen were found to be natural barriers against the penetration of Solcoderm applied to the skin surface. Mild mechanical disruption and the disturbed cellular continuity of pathologic tissue allows the solution to penetrate treated lesions more freely and enhances the likelihood of their complete destruction when an appropriate quantity of solution is properly delivered by an experienced therapist.

We describe a 62-year-old woman who presented with a recurrent neurofibroma, two ‘café-au-lait’ spots and a possible cerebral tumor. This Von Recklinghausen neurofibromatosis was associated with an asymptomatic systemic mastocytosis with bone marrow involvement. Double-blind clinical studies of pairs of lesions were initiated to evaluate several components of Solcoderm treatment. The superiority of Solcoderm over plain nitric acid was as evident with plastic as with wooden applicators. A mixture of oxalic and nitric acid had effects similar to Solcoderm, while acetic and nitric acid was similar in effect to plain nitric acid.
