SUMMARYThe relationship between immunoglobulin allotypes and leprosy was studied in 91 unrelated patients and 100 healthy controls from Vietnam. Twenty Vietnamese patients with tuberculosis were also typed for the Gm, A2m and Km allotypes. The results were compared with those from the healthy controls. No significant association was found for the allotypes G1m(z,a,x,f) G2m(n), G3m(g,b), A2m(1,2) and Km(1,3) between the two groups of patients and the controls. Heterogeneity in the distribution of G2m(n), G3m(b), A2m(2) and Km(3) was found when 60 polar‐lepromatous (LL) patients and 27 borderlinetuberculoid (BT) patients were separated out of the 91 leprosy patients. In the LL patients there appeared to be a significantly higher frequency of G2m(n), G3m(b) and A2m(2) in coḿparison with the BT patients (P<0.05). A significantly lower frequency of Km(3) was found in the LL patients in comparison with the healthy control group (P<0.05). The frequencies of the Gm‐A2m haplotypes and of the occurrence of the Km(1) and Km(3) in the Vietnamese population were calculated on the basis of the results in the 100 samples of healthy controls. SUMMARYGenes associated with immunoglobulin (Ig) allotype determinants are important in regulation of immune responses to bacterial polysaccharides. Furthermore, loci associated with Ig allotypes have been reported to interact with those associated with the major histocompatibility complex and affect susceptibility to certain diseases. In the present study we determined the frequencies of certain Gm phenotypes in patients withHaemophilusmeningitis or epiglottitis and in controls. HLA‐A, ‐B and ‐DR specificities had previously been determined in the majority of these subjects. Although no Ig phenotype was associated with increased or decreased relative risk of disease, the frequencies of several combinations of HLA specificities and Ig phenotypes were significantly different from those of controls. Thus, for subjects with the Gm phenotype (1, 3, 17; 23; 5, 13, 21), the risk ofHaemophilusmeningitis or epiglottitis was lower in individuals with HLA‐B5 than in those without this specificity (odds ratio<0.1, P<0.004). In contrast, for subjects with the closely related Gm phenotype differing only by the absence of Gm(23), (1, 3, 17; 5, 13, 21), the risk of disease was higher in those with HLA‐DR3 than in individuals who lacked DR3 (odds ratio = 11.0, P = 0.02). SUMMARYNormal C57BL/6 (B6) mouse serum was tested in the direct cytotoxicity assay for specific reactivity against lipopolysaccharide (LPS)‐stimulated mouse spleen cells. Selective reactivity was found in weanling and adult serum against lymphoblasts from mice that express an antigen encoded by theH‐2Kkregion of the major histocompatibility complex (MHC). Other strains, congenic with B6 at the MHC, did not exhibit the same alloreactivity. Serum from mice of a congenic strain being derived in our laboratory, which differs from B6 at two unlinked loci,Tlaandnu, exhibited similar reactivity against the B6‐H‐2kLPS‐stimulated lymphoblasts, implying that a competent T cell compartment is not necessary for generation of this reactivity. SUMMARYWe studied structural and functional characteristics of lymphocytes from adult and fetal baboons (Papio cynocephalus). Flow cytometry with monoclonal antibodies to human lymphocyte antigens and plant lectins was used to define expression of surface antigens on lymphocytes from adult and 140 day fetal baboons (term = 180 days). Major T cell antigenic determinants on adult and fetal baboon lymphocytes were the Tp50, Tp32‐45, and p45 glycoproteins detected by monoclonal reagents T11, OKT8, and OKT10 respectively. Baboon T lymphocytes did not react with the OKT3/anti‐Leu4 or OKT4/ anti‐Leu3a reagents which detect, respectively, Tp19‐29 and Tp55, major surface glycoproteins on human T lymphocytes. OKT6, which identifies the human TL antigen equivalent on thymocytes, did not react with baboon thymocytes. These data demonstrate major evolutionary divergence between human and baboon T lymphocytes. By contrast, baboon lymphocytes resembled human peripheral lymphocytes in reactivities with several non‐T cell reagents. Lectin binding studies revealed substantially fewer peanut agglutinin‐and wheat germ agglutinin‐binding cells in suspensions of baboon fetal splenocytes and adult peripheral lymphocytes compared with fetal thymocytes. Thereffore, maturation of baboon T lymphocytes is associated with loss of surface carbohydrate structures that bind these lectins. Adult and fetal baboon lymphocytes resembled human and murine lymphocytes in their capabilities to respond to mitogens and to produce interleukin‐2. As in oter species, adult, but not fetal baboon lymphocytes, mediated NK activity against a variety of nucleated target cells. SUMMARYA monoclonal antibody designated ‘antibody 390’ (Ab 390) with anti‐human Thy‐1 reactivity was prepared by the hybridoma technique from the splenocytes of BALB/c mice immunized with human fetal brain. This antibody was shown to have anti‐human Thy‐1 reactivity because (1) it precipitated a molecule with a molecular weight of about 24,000 daltons, (2) it had a pattern of reactivity similar to that of previously described anti‐human Thy‐1 antibodies and (3) purified human Thy‐1 antigen specifically inhibited binding of Ab 390 to a known antigen‐positive cell line. It was the intent of this study to investigate the distribution of Thy‐1 on normal and malignant haematopoietic cells in humans and non‐human primates. We show here that Ab 390 did not react with human peripheral blood leucocytes, bone marrow cells or splenocytes by immunofluorescence but did react with subcapsular and cortical fetal thymocytes by peroxidase‐antiperoxidase immunohistology. A section of fetal spleen demonstrated staining of connective tissue and blood vessels and rare reactive lymphocytes. Adult spleen contained Thy‐1‐positive cells surrounding the white pulp and in the marginal zone, but single‐cell suspensions of splenocytes did not react with Ab 390. Ab 390 was tested against a variety of fresh human leukaemia cells and human cell lines and was shown to react with only the acute lymphoblastic leukaemia T cell line RPMI 8402 and HPB‐MLT. Non‐human primate studies revealed reactivity with a number of T cell lines from New World primates (cotton‐topped and red‐bellied marmosets) and peripheral blood granulocytes (owl monkey). Our studies support previous findings that suggest that human Thy‐1 may be a marker for early T lymphocytes in man, and its distribution on non‐human primate T cell lines suggests the same for certain species of non‐human primates. SUMMARYIn previous studies from this laboratory the antigenic sites of lysozyme were found to be composed of spatially adjacent surface residues that are mostly distant in sequence (i.e. discontinuous sites). For synthetic mimicking of the sites, we introduced the concept of ‘surface‐simulation’ synthesis by which the binding site residues are linked directly via peptide bonds with appropriate spacing and directionality into a single peptide which does not exist in the protein but mimics a surface region of it. In the present report T cell recognition of the surface‐simulation synthetic antigenic sites has been explored in a mouse strain, B10.BR, that is a high responder to lysozyme. The discontinuous antigenic sites of lysozyme also had the capacity to stimulate proliferation of T cells driven by native lysozyme in long‐term cultures. Thus, in addition to the four continuous T sites that we have recently reported, T cell recognition of lysozyme also involves discontinuous sites. SUMMARYAll mice responding to the terpolymer GLA40make GL, GA and GLA specific antibodies irrespective of their response to GL or GA alone. The mice displayed positive T cell proliferative responses against the homologous terpolymer, but no T cell responses were obtained with GL, which is non‐immunogenic in mice. T cells from GLA immune mice, which are also responders to GA, such as mice ofH‐2haplotypesa, b, d, kandr, could be stimulated by GA.
