By using an in vitro quantitative clonal culture technique for bone marrow granulocyte/macrophage progenitor cells (GM-CFC), we studied the effects of cimetidine alone and cimetidine plus azathioprine on bone marrow toxicity in C57BL/6 mice. Femoral bone marrow showed no effect of cimetidine in doses from 31.25 to 500 mg/kg on either marrow cellularity or the number of GM-CFC. Cimetidine pretreatment with single doses of 62.5 mg/kg or 250 mg/kg had no effect on the bone marrow suppression of azathioprine at 100 mg/kg. Chronic cimetidine pretreatment at 62.5 mg/kg daily for 7 days also had no effects on the single-dose azathioprine toxicity. Cimetidine given either before or after azathioprine had no effect on the rate or final level of recovery of GM-CFC in the marrow after depletion by azathioprine. Cimetidine in vitro at doses of 3.1 to 200 μg/ ml caused no alteration in the proliferative response of the GM-CFC. Analysis of the serum colony-stimulating activity 1 to 24 hr following doses of cimetidine of either 12.5 mg/kg or 31 mg/kg caused no change in the serum colony-stimulating activity. We could find no evidence that at clinically relevant doses, cimetidine increased the hematopoietic toxicity of the azathioprine or altered the rate of bone marrow recovery after azathioprine depletion.

Isolated adult rat hepatocytes were cryopreserved by cooling, in the presence of dimethyl sulfoxide, to low subzero temperatures and they were stored for 14 days at – 196 C. The cells were then recovered and transplanted into the splenic pulp of the original donor animal. A similar transplant procedure was performed with hepatocytes which had not been cryopreserved. Ultrastructural assessment was made 1 month after transplantation. In general fewer hepatocytes survived ectopic transplantation after cryopreservation, but the surviving cells were ultrastructurally similar to ectopically transplanted cells not subjected to cryopreservation. Thus, the reduced metabolic activity measured previously in transplants of cryopreserved hepatocytes is likely to result from a smaller number of competent cells rather than from hepatocytes which have survived in an extensively damaged or abnormal state.

Radiation chimeras in mice were induced by intrasplenic injection of allogeneic bone marrow cells instead of intravenous injection. Interestingly, the survival time in X-irradiated BALB/c mice inoculated intrasplenically (i.s.) with bone marrow cells from C3H/He mice was markedly prolonged as compared with that in X-irradiated BALB/c mice inoculated i.v. with bone marrow cells from C3H/He mice. However, when C57BL/6 mice were used as donors, a significant difference between i.s. injection and i.v. injection has not been found in survival time at 60 days after X irradiation. On the contrary, when bone marrow cells from BALB/c or C57BL/6 mice were injected into X-irradiated C3H/He mice, i.s. injection gave longer survival days to recipients than did i.v. injection. Based on testing their chimerism, it was suggested that lymphoid cells of donor origin were predominantly identified in almost all BALB/c or C3H/He recipients which were inoculated i.s. with bone marrow cells from C5BL/6 mice. However, somewhat incomplete chimerism was observed when the C3H/He to BALB/c donor-recipient combination was used and vice versa.

A 38-year-old male with a 5-year history of dialysisdependent renal failure secondary to membranoproliferative glomerulonephritis, type I, received a cadaveric renal allograft from a 51-year-old male dying from acute trauma. A pretransplant biopsy of the donor kidney revealed no glomerular disease. One month after transplantation, the patient was discharged with a stable creatinine of less than 2.0 mg/dl, but subsequently developed positive urine cultures forStaphylococcus aureusandEscherichia coliand eventually underwent transplant nephrectomy. Blood cultures were positive postoperatively forS. aureus, and examination of the transplant nephrectomy showed ultrastructural, histological, and immunofluorescent findings characteristic of postinfectious glomerulonephritis. While numerous types of glomerular disease have been reported in kidney transplants, this report represents the first case suggestive of bacterial postinfectious type glomerulonephritis we have seen occurring de novo in a renal allograft.

摘要:
Although opportunistic infections after allogeneic bone marrow transplantation are common, cerebral toxoplasmosis has only been reported once. We report a case of lethal cerebral toxoplasmosis in a patient with acute leukemia who had been submitted to intensive cytotoxic therapy and allogeneic marrow rescue. The clinical course was characterized by an unusually sudden neurological onset. It was remarkable that, in this case, toxoplasma serology remained repeatedly negative. The autopsy revealed that the infection also involved the myocardium. Serological data and pathology findings are presented.

HLA-DR-identical North American individuals of Caucasian, black, and Asian Indian origin were tested in checkerboard mixed lymphocyte cultures (MLCs) and the distribution of negative (weak), intermediate, and positive (strong) MLC reactions was determined using the linear clustering analysis program. Overall, the data show that the frequency of weak MLC reactions is significantly higher when the HLA-DR-identical responderstimulator pair derives from the same ethnic group than when they are of different ethnic origin. Caucasians responded stronger to blacks than to Asian Indians; blacks responded stronger to Asian Indians than to Caucasians, while Asian Indians responded stronger to blacks than to Caucasians.The fact that the frequency of MLC identity is greater among HLA-DR-identical individuals of similar as opposed to different ethnic origin suggets that the predictability of successful transplants by HLA-DR matching may be also influenced by the ethnic background of the donor.

Monoclonal antibodies H9–15.4 and H39–459 were derived from an A.TH anti-A.TL immunization. Antibodies H9–15.4 and H39–459 were found to be directed against the I-Ekmolecule with positive reactions against B10.A (AkEk) and B10.S(9R) (A*Ek) but not B10.A(4R) (AkEb). The monoclonal antibodies were also found to react with thebandqhaplotypes, which do not express an I-E molecule. Sequential precipitation analysis showed that in thebhaplotype, H9–15.4 and H39–459 react with the I-Abmolecule. These results showed that H9–15.4 and H39–459 recognize determinants shared by I-Ekand I-Abmolecules, suggesting that I-A and I-E molecules may have a common evolutionary origin, possibly through gene duplication.Sequential immunoprecipitation analysis of I-Abmolecules precipitated by H9–15.4 and H39–459 also suggested that these monoclonal antibodies recognize subsets of I-Abmolecules. Pretreatment with the 17–227 monoclonal anti-I-Abantibody (Ia.15) had no effect on I-Abmolecules precipitated by H9–15.4 and H39–459. Also, pretreatment with H9–15.4 and H39–459 had no effect on I-Abmolecules immunoprecipitated by 17–227. Also H9–15.4 and H39–459 only partially cleared Ia molecules immunoprecipitated by each other. These results suggest that 17–227, H9–15.4, and H39–459 detect a minimum of four subsets of I-Abmolecules. To account for these observations, it is proposed that during evolution of the mouse H-2 complex, in addition to gene duplication of ancestral gene(s) to yield the genes encoding EαAα, Aβ, and Eβpolypetide chains, further gene duplication occurred, forming multile copies of genes encoding each Ia polypeptide chain.
