A histochemical study of diphosphopyridine nucleotide diaphorase activity in second-set skin homografts in rats during the first 3 days after transplantation demonstrated: 1. an ingrowth of new host capillaries into the deep layer of the dermis of both typical second-set grafts and white grafts during the first 24 hours after grafting; 2. the cessation of vascular ingrowth in white grafts between 24 and 48 hr; 3. an atypical homograft reaction of second-set grafts transplanted 5 days after first-set grafting; and 4. the variability of type of homograft reaction occurring in second-set grafts transplanted 6 and 7 days after-set grafting.

摘要:
A new and rapidly fatal form of allogeneic disease, called acute allogeneic disease (AAD) in mice, is described. The disease is produced by the i.p. injection of immune peritoneal cells. It is characterized by extensive necrosis of the pancreas, generalized peritonitis, and circulatory shock. The disease apparently results from the immunological activities of immune peritoneal macrophages while in close association with tissues and organs of the peritoneum.

Histochemical studies of diphosphopyridine nucleotide diaphorase activity, using neotetrazolium salts as reagents, were undertaken in split-thickness skin autografts in rats, removed with a dermatome, and replaced on the donor dermal bed. In contrast to an early and rapid loss of diaphorase activity in the pre-existing blood vessels of full-thickness grafts placed on a suprapannicular bed, the delayed and slower decline of activity in such vessels in the split-thickness grafts suggests that the onset and rate of degenerative changes in these vessels vary with the thickness of the graft; in thin grafts nutrient fluids during the phase of plasmatic circulation have a shorter distance to travel and the thin graft has fewer cellular elements requiring nourishment. Histochemical and stereomicroscopic studies of the split-thickness grafts show that the vascular ingrowth originates from the distended blood vessels lying in the deep host dermal layer immediately superjacent to the panniculus carnosus, not from the vessels at the surface of the host dermal bed in contact with the graft. The revascularization of the split-thickness graft on a dermal host bed occurs at the same rate as that of a full-thickness graft on a suprapannicular bed.

The recovery of the primary immune response in CBA mice which have been given a potentially lethal dose of irradiation and syngeneic bone marrow therapy is very slow and incomplete if the hosts have previously been thymectomized. Restoration of the immune response can be obtained by grafting these mice with a single neonatal CBA/T6T6 or C3H thymus lobe. Mice given C57BL thymus grafts did not show such a marked restoration of immunological function. Histological investigation revealed that the CBA/T6T6 and the C3H grafts were accepted by the host but that the C57BL grafts failed to persist. Mice which had received C57BL thymus grafts were shown to be immune to C57BL skin grafts.

The results of skin graft exchanges between members of a partially inbred line of White Leghorns provide evidence that theCblood group locus is also a histocompatibility locus or is closely linked to such a locus. The effects of twoCalleles were investigated. One allele showed marked influence on graft rejection while the effects of the second allele were not differentiated from the effects of other histocompatibility loci segregating in the population. Previous studies have shown thatCantisera have lymphocyte agglutination properties.

Duration of survival and histopathological appearance of renal homotransplants in 3− to 5 1/2-month-old puppies thymectomized at birth were comparable to that noted previously in intact members. Similarly, neonatal thymectomy failed to inhibit lymphoid development in lymph nodes, spleen, and ileum. These findings fail to provide evidence that the neonatal thymus plays a role in transplantation immunity in this species.

Allogeneic dissociated spleen cells incorporated in Freund's complete adjuvant and injected s.c. induced a homograft response as radio-resistant as that produced by a skin graft. In two of three strains of mice studied, allogeneic dissociated spleen cells alone, when injected s.c., induced a homograft sensitivity only slightly less radioresistant than that induced by skin grafts. Anatomical separation of the injection sites of allogeneic dissociated cells and Freund's complete adjuvant produced a homograft response more radiosensitive than that induced by spleen cells alone.

Temporary lymphatic discontinuity was established in rabbit ears bearing skin autografts and homografts, to determine the effect on lymph node response and graft rejection. Blood vascular continuity was not interrupted, and healing and vascularization of the grafts were not impaired. Rejection of homografts was delayed by the time required for alternative lymph drainage channels to develop. The regional lymph node did not react to the homograft until lymphatic pathways were reestablished; graft rejection then ensued in standard fashion. The reaction of lymph nodes remote from the graft site was also delayed until lymph continuity was restored. These observations support the view that during the stage of establishment of an allogeneic skin graft, the major route of transfer of antigen to the host is along lymphatic pathways.

C57BL/6 (H-2b) mice were grafted with skin from DBA/1 (H-2q) mice and the resultant hemagglutinin titers were observed both in series undergoing graft rejection and in series undergoing surgical excision of the graft just prior to the onset of rejection. Excision of both first- and second-set grafts produced an accelerated rise in the hemagglutinin titer. Elevated hemagglutinin titers were then produced in other C57BL/6 mice by two or more DBA/1 skin grafts and the effects of applications of further DBA/1 grafts were observed. These grafts were found to produce an early transient fall in pre-existent hemagglutinin titers prior to their anticipated rise in response to the challenging graft. The implications of these observations are discussed.
