SUMMARYNewborn rats were inoculated via the umbilical vein with suspensions of tissue-cultured cell lines of human cancer (HEp 2 and J-111). Fluorescent antibody studies demonstrated that many of these cells were deposited in the liver, but they seldom grew to produce tumor nodules in the liver. When they did grow in liver (1/11 rats with HEp 2 and 4/16 with J-111), implants also grew in the lungs. This is in contrast to results of transplantation of these cells into newborn rats via the tail vein, which produced progressively growing tumors in the lungs and adrenals of a great majority of the recipients, and never produced implants in the livers. The observations are interpreted as indicating a role of local tissue factors in determining receptivity to the growth of tumor cell implants. The observations provide experimental evidence in support of the concept that distribution of tumor metastases may be due in part to differences in the susceptibility of various tissues for the growth of randomly distributed cancer cells.

SUMMARYAntibody response of A.CA, A.SW, and C57B1 mice on immunization with A/Sn tissue was studied. Gel filtration on Sephadex G200 revealed two antibody classes: an earlier 19S (peak titers day 4-5) and a later 7S (peak titers day 12-15). On repeated immunization A.CA and A.SW responded with 7S antibody, C57BL with 19S and 7S. Some strain differences in antibody pattern were found after hyperimmunization with sheep erythrocytes, 19S and 7S activities were studied in vitro. In cytotoxic and hemolytic assays orders of reaction close to 1 were found for 19S and close to 2 for 7S. 7S gave positive staining in indirect fluorescent tests whereas 19S was not detectable. The findings are discussed in relation to efficiency per molecule in the antibody classes. Parallellism between strain differences in antibody pattern and homograft reactivity is pointed out.

SUMMARYThe renal homograft response was studied in 23 goats following four procedures—I. Sham operation, II. thymectomy, III. thoracic duct drainage, and IV. thymectomy and thoracic duct drainage. The technique adopted for thoracic duct drainage allowed diversion of lymph flow to be maintained for 5 days before transplantation, and for the lymphatic flow to be restored to normal thereafter. The homografts in groups II and III failed to show prolongation of survival. Those in group IV showed moderate increases in survival, though the results were not statistically significant. The failure of thoracic duct drainage to prolong survival of renal homografts differs markedly from its effect on skin homografts and there is no evidence that it is of use as a therapeutic measure in clinical renal homotransplantation at the present time. The results following thymectomy and thoracic duct drainage suggest that it may be worthwhile investigating the combination of the former operation with a more potent method of immunosuppression for the control of renal homograft rejection.

SUMMARYMouse H-2 isoantigens have been solubilized from lymphoid cell-derived lipoprotein by autolysis. The soluble material was purified on P300 Bio-Gel and DEAE-Sephadex and activity then shown to be associated with the single component revealed on polyacrylamide gel electrophoresis. The product had about 500 times the specific activity of the starting material. Evidence is provided to show that several H-2 specificities are present on the same molecule and that in material prepared from F1hybrid mice at least some hybrid molecules exist, carrying specificities derived from both parental strains. The product did not react in any of several non-H-2 test systems.

SUMMARYMice undergoing transplantation (runt) disease produced by injections of spleen cells across the H-2 locus in various strain combinations show a dramatic elevation of renal lysozyme activity. This enzyme would seem to be a good biochemical marker of the graft-versushost reaction and related phenomena under controlled experimental conditions. An increased renal lysozyme activity also occurs in “normal” mice with the onset of advanced age. This age-related increase in activity of the enzyme can be interpreted as supportive evidence for the immunologic theory of aging.

SUMMARYRabbits bearing skin allografts were treated with methyl-prednisolone to delay graft rejection. The recipients were studied serologically for histocompatibility antibodies by the antiglobulin consumption test, mixed agglutination, lymphoagglutination, and hemagglutination. No serum antibodies were detected in the drug treated host by the latter three methods. Data obtained from antiglobulin consumption show that the usual lag phase between rejection of a graft and the appearance of serum antibodies which is so characteristic of unmodified rejection is greatly reduced if not eliminated in rejection delayed by cortisone. This raises the possibility of a serologic method of diagnosing rejection.

SUMMARYAttempts to characterize the time course of parental-versus-F1hybrid immunological reactions in sublethally irradiated mice have resulted in the following observations. (1) It was possible to reverse the lethal effect of the reaction by means of a second dose of X-rays delivered up to 4 days after injection of the parental lymphoid cells. In contrast, mice irradiated 5 days or later succumbed to the disease. (2) Nonirradiated host-type colony-forming units in the spleens of mice undergoing the parental-F1hybrid disease were depleted at 5 days, and in the marrow by 7 days. (3) It was not possible to reverse the lethal course of the disease with large doses of host type hemopoietic cells injected 3-6 days after injection of the parental cells.

SUMMARYInteractions of humoral antibodies (C57 anti-BP8) and syngeneic immune lymphocytes have been studied in vivo. The effect of each of these agents alone, or in combination, on the growth of the ascites sarcoma BP8 (C8H tumor) in C57 mice was determined. Small amounts of antiserum alone regularly produced enhancement of tumor growth, whereas C57 lymphocytes obtained early after immunization, or large volumes of antiserum alone, were inhibitory to tumor growth. Sensitized C57 lymphocytes obtained at a later time after immunization frequently produced tumor enhancement under the same experimental conditions. Synergistic action was demonstrated between immune serum and such older antibody producing lymphocytes, whereas antagonism was noted between immune serum and more recently sensitized lymphocytes. The importance of these observations in relation to the mechanism of allograft rejection is discussed.
