SUMMARYRats thymectomized neonatally received allogeneic bone marrow implants from randomly bred normal donors and were treated on alternate days with antilymphocyte serum (ALS). Fifty-four percent of implants left in situ for 15 days or more developed into ossicles containing hemopoietic marrow. Allogeneic marrow implanted in rats which were either normal, neonatally thymectomized and not treated with ALS, or treated with ALS alone, with few exceptions failed to develop into marrow-containing ossicles. Initially their development was normal, with mesenchymal cell proliferation and early cancellous bone formation. Rejection of the implant then became evident with loss of osteoblasts, bone resorption, and failure of sinus development. As a result of this lack of microvascular regeneration, hemopoiesis did not occur.

SUMMARYF1hybrid renal grafts survive for markedly prolonged periods in parental rat recipients treated by active or passive enhancement. Splenectomy of the renal recipient, when done 7 days before or up to 6 days after grafting, abolishes the immunological enhancement. Yet, splenectomy has no effect when performed 10 or more days later. Transferable enhancing activity is present as early as 2 days after a single i.v. immunization with allogeneic cells, preceding by 2–3 days both hemagglutinating and cytotoxic antibodies. Splenectomy prevents formation of enhancing activity but not hemagglutinating or cytotoxic antibodies.

SUMMARYThe survival time of grafts exchanged between BALB/c and C57BL/6 mice was significantly prolonged nearly twice that of untreated controls by topical application of triamcinolone acetonide prior to transplantation. A study of the mechanism of this prolongation, using mice and rabbits, suggests a dualistic action of topically applied corticosteroid. Evidence presented shows that local steroid both retards the devlopment of immunity toward skin allografts and suppresses the local inflammatory response in the immunized animal.

SUMMARYFour murine leukaemias, induced by 7,12-dimethyl-benz(a)anthracene (DMBA) in (DBA/2 × C57BL/6)F1hybrid mice have been studied for their H-2 antigenic constitution. Different oligospecific isoantisera have been absorbed with various quantities of each type of leukaemic cell and with normal cells from spleen, thymus, and lymph node for comparison. One of these leukaemias shows a general diminution of all the H-2 specificities without detectable qualitative loss. The loss of some H-2 specificities has been demonstrated on the three others, but those specificities lost varied from one leukaemia to another. In one case, an increase of one or more specificities has been observed. It seems that the affinity between the H-2 specificities and the antibodies of one given antiserum can differ with different leukaemias. No relation between H-2 expression and the number of leukaemic cells giving 50% mortality in the syngeneic host has been found.

SUMMARYThe ♂ antigen in the rat is described. The median survival times (MSTs) of intra-strain ♂ → ♀ skin grafts vary between 50 and 280 days. No association is apparent between ♀ reactivity to the ♂ antigen and theAg-B (Rt-H1)genotype. The ♂ antigen is identical in 6 of 7 strains so far investigated. The MSTs of successive ♂ → ♀ skin grafts become progressively shorter for the first four grafts, but thereafter tend to fluctuate in duration; considerable individual fluctuation is evident over a series of grafts. At no stage does rejection proceed with the maximum intensity of which the host is capable. The MSTs of parental strain ♂ skin grafts on F1hybrid ♀ hosts are intermediate between the MSTs recorded for ♂ → ♀ skin grafts in the two parental strains. Skin from ♂ donors shows a progressive increase in immunogenicity between birth and old age. This is probably an attribute of antigenic weakness rather than a specific characteristic of the ♂ antigen. Neonatal ♂ skin grafts effect greatly prolonged survival of simultaneously grafted aged ♂ skin from the same strain, the effect being immunologically specific.

SUMMARYThe addition of a goat antihuman γ-globulin reagent to the two-stage cytotoxicity test has produced a cell typing method which appears to be more sensitive than other widely-used variants of the lymphocytotoxicity technique. Twenty-six percent of 788 reactions negative with four conventional variants of cytotoxicity testing were positive with the antiglobulin method. The presence of antigen-antibody activity was confirmed by platelet absorption and elution studies for a number of these cytotoxicity negative-antiglobulin positive reaction combinations. The absorbed serum showed no cytotoxic activity against lymphocytes and the eluate recovered from the platelets gave positive tests by the antiglobulin method.

SUMMARYCross reactivity in the HL-A system has been evaluated quantitatively by utilizing human cultured lymphoid cells and soluble HL-A antigens in cross absorption and cross inhibition assays. Most of the cross reactions among different HL-A specificities determined with platelets or peripheral lymphocytes were confirmed in this study. Some cultured lymphoid cells or soluble HL-A antigens possessing a cross reacting specificity cross absorbed a certain anti-HL-A antiserum, whereas others with the same specificity did not. The number of cells required to absorb cytotoxic HL-A alloantisera directed against cross reacting specificities is significantly greater than that necessary to absorb alloantisera directed against HL-A determinants present on the cell surface. This observation suggests that relatively small numbers of available antigenic determinants react with the cross reacting antibodies. The differential absorbing capacity of different cultured cell lines for the same cross reacting HL-A alloantibodies suggests a variability in the cell surface expression of cross reacting HL-A determinants.

SUMMARYAntisera prepared to mouse liver lysosomes were found to prolong the survival of allografts in mice. Antisera to the soluble enzymes contained within lysosomes inhibited the rejection of allografts to a greater extent than did the antisera to lysosomal membranes. Lysosomal antisera may enhance graft survival by inhibition of the processing of antigen by macrophages.

SUMMARYCells proliferating in a local graft-versus-host reaction (GVHR) in the rat kidney were selectively labeled by local injection with radioactive precursors of deoxyribonucleic acid (DNA) while the blood vessels of the renal pedicle were temporarily occluded. The rats were killed sequentially and the amount of label in the local GVHR and other organs of the host was measured. When the label was given on day 5 the level within the local GVHR was sustained for at least 48 hr, whereas when given on day 7 it declined within 24 hr. There was no evidence that intact labeled cells migrated to lymphoid tissues of the host with the possible exception of the wall of the intestine, to which a minority of labeled cells migrated prior to their death. We thus conclude that most of the cells which proliferate in the local GVHR and their progeny died in situ about the 8th day under the conditions of these experiments.

SUMMARYAn alloantiserum (AAS) prepared in adult Brown Norway (BN) rats against Lewis (L) thoracic duct lymphocytes (TDL) exerted anti-L lymphocytotoxins and lymphoagglutinins in vitro and was capable of aborting an already developed graft-versus-host (GVH) reaction caused by L-TDL in neonatal BN rats in vivo. A blastogenic activity was not provoked by the antiserum in L lymphocytes either in vitro or in vivo. However, the AAS did promote the disappearance of L lymphocytes transfused into adult and neonatal BN animals with a higher efficiency in the adult. These observations demonstrated that the AAS did not act solely on circulating lymphocytes but also on those L cells resident in BN lymphoid tissue, because they were preferentially removed in BN animals containing “homed” radioactively labeled L lymphoid cells by lymph node reticular cells. In contrast, BN animals given antiserum prior to being transfused with L cells, and hence possessing circulating antibodies specific for the labeled allogeneic L inoculum, removed the lymphocytes very rapidly (within 2 hr or less), chiefly by action of phagocytic blood sinusoidal lining cells. Therefore, specific removal seemed to occur in animals given antiserum by whatever BN phagocytic cell the L lymphocyte had first contacted. Moreover, removal of circulating and homed cells within 24 hr in the adult and at least 36 hr in the neonatal BN animal, suggested that the antiserum penetrated into lymphoid tissue. This is in contrast to heterologous antilymphocyte serum (ALS) which had been reported not to penetrate into tissues. As transfused radioactively labeled L lymphocytes contained small cells of both newly formed and longer lived varieties, and because all labeled cells were removed by the antiserum, the AAS did not appear to affect one population of small lymphocytes in preference to another.
