Thirty-nine (LEW × BN)F1kidneys were transplanted to LEW rats. Twenty-four untreated recipients survived for a mean time of 16.1 ± 1.7 days (group 1). Fifteen recipients received 4 ml of antilymphocytic serum per rat (group 3). In the last group 10 recipients survived for more than 4 months. The spleen cells of these permanently surviving 10 rats were obtained by splenectomy and used in a graft-versus-host assay, and this assay showed that the reactivity of these cells was normal. Following splenectomy the animals were given an (LEW × BN)F1skin allograft, followed 18 days later by a second. After another 18 days (LEW × Buf)F1“third party” skin allografts were transplanted to the same animals. Animals of group 2 rejected their first grafts with a mean survival time of 12.2 ± 1.2 days, whereas the second grafts were rejected normally as were the third party grafts. Attempts were made to detect lymphocytotoxic antibodies and haemagglutinins before and after the transplantation of skin grafts and none could be found up to day 53. The sera of group 2 inhibited allorosette formation by 38%. This serum-blocking factor was donor specific. It is probable that the survival of the kidney transplants following antilymphocytic serum treatment was brought about by the development of blocking antibodies.

The immunogenic properties of HLA-A9 antigens isolated from serum have been evaluated. A9 antigens at various stages of purification can elicit the formation of cytotoxic antibodies which become operationally specific to A9 either after absorption of the xenoantisera with cultured human lymphoid cells or human red blood cells, or after dilution of xenoantisera with human serum. A9 xenoantisera do not affect mixed lymphocyte reactions between allogeneic lymphocytes carrying A9, suggesting that coating of antigens of the A locus does not impair the functional activity of lymphocytes in the mixed lymphocyte reaction.

Human peripheral blood lymphocytes were digested briefly with protease prior to application of indirect immunofluorescence techniques for detecting alloantibodies in sera of patients with chronic renal failure on maintenance hemodialysis. Background staining of intrinsic surface IgM and cytophilic IgG bound to Fc receptors was eliminated or greatly reduced, enabling detection of B cell specific antibodies, including cold-reactive types not demonstrable by conventional immunofluorescence or complement-dependent lymphocytotoxicity. The antigenicity of HLA and other surface membrane determinants was not decreased by protease, although reactivity with certain sera was enhanced. In experiments comparing indirect immunofluorescence using protease-treated cells with complement-dependent lymphocytotoxicity and antibody-dependent, lymphocyte-mediated cytotoxicity assays, indirect immunofluorescence was more sensitive and comprehensive, but not less specific, in defining alloantibodies of a variety of types.

Phagocytic glass-adherent mononuclear cells in peripheral human blood (morphologically monocytes) have been shown to play an essential role in lymphocyte transformational response to phytomitogens. Lymphocytes from peripheral blood have been exhaustively depleted of monocytes by successive treatments, first with opsonized iron particles with eventual magnetic elimination of phagocytic cells, followed by filtration of the same population through packed glass-sand columns. This subpopulation, enriched in T lymphocytes and containing around 4% B lymphocytes, does not transform in response to concanavalin A, phytohemagglutinin, or pokeweed mitogen unless viable monocytes are added to the culture. Supernatant fluids from monocyte-enriched human blood leukocyte cultures can substitute for monocytes in restoring response. Interaction of the phytomitogen with lymphocyte, while essential, is not sufficient to trigger transformation, requiring a second signal provided by the monocyte probably via release of soluble mediators.

The immunosuppressive properties of niridazole, an antihelminthic drug, have been investigated in rats. When given orally in a dose of 50 mg/kg, it extended the median survival of cardiac allografts from 7 to 20 days. The immunosuppressive effect was not increased by giving either azathioprine or prednisolone concurrently but when all three drugs were combined, immunosuppression was profound and only two of eight grafts were rejected. Drug combinations incorporating niridazole at a lower dosage or for a shorter period were less effective, and azathioprine and prednisolone on their own or used together prolonged graft survival only marginally in this model.It is concluded that niridazole is a powerful immunosuppressive drug in this species and a synergistic effect can be obtained by using it in combination with azathioprine and prednisolone.

A satisfactory and reproducible technique of cryopreservation of canine lymphocytes for use in mixed leukocyte culture (MLC) and cell-mediated lympholysis tests has been developed. Dimethyl sulfoxide was used as the cryopreservation agent. Cells were frozen to −50 C at a controlled rate (-1 C/min) and stored at −169 C. The best preservation was obtained with a concentration of 10 × 106lymphocytes/ml in Way-mouth's MB-752/1 medium supplemented with 30% dog serum (MB-30). Before use in MLC or cell-mediated lympholysis tests, lymphocytes were rapidly thawed at 40 C and then rapidly diluted with MB-30 at room temperature. For best responses in MLC, one fresh component (either stimulating or stimulated cells or serum) was needed. The magnitudes of responses of frozen lymphocytes to phytohemagglutinin or in MLC were similar to those seen with fresh cells, but peak responses occurred usually 24 hr after those seen with fresh cells.

T cell function of “lethargic” mutant mice which exhibit spontaneous thymic involution was evaluated by skin transplantation and graft-versus-host reaction tests. At 15 to 30 days of age, the mutant mice did not reject skin allografts, but by 45 days of age their response was normal. Spleen cells from 24-day-old mutant donors demonstrated delayed appearance of graft-versus-host-induced splenomegaly in F1recipients. Implications of these results in understanding the immunological status of lethargic mice were considered.

Acetone-dried powders were prepared from the spleens of DLA-serotyped dogs and assayed for their specific absorptive properties for anti-DLA-A and B antisera. The specific activity of two different antisera (anti-DLA-A9 and anti-DLA-B13) was readily absorbed with acetone-dried powders prepared from the spleens of dogs of the corresponding DLA types. This specific activity was recovered by the elution of the powder used for the serum absorption. The same method was used to narrow the specificity of a highly polyspecific antiserum. The comparison of the absorption properties of acetone-dried spleen powder versus fresh spleen cells shows that the treatment with acetone does not modify qualitatively and quantitatively the DLA specificities.

Cell-free medium from stimulated murine leukocyte cultures induced spleen enlargement in vitro. Splenomegaly was measured by [3H]leucine incorporation of spleen fragments in tissue culture. Stimulatory cell-free medium were induced in leukocyte cultures with allogeneic or phytohemagglutinin stimulation. Supernatant activity has lymphokine properties, i.e., cell-free soluble factor(s), generated during specific lymphocyte activation, but which are expressed without reference to H-2 specificity.

A method of dissociation of the rat heart cell for the mixed lymphocyte culture purpose is described. The treatment of the young rat heart with 0.1% collagenase-hyaluronidase solution yielded satisfactory heart cell suspension. The heart cells, thus obtained after mitomycin C treatment but not irradiation, stimulated allogeneic lymphocytes in the presence of 2-mercaptoethanol. In the Fischer to Lewis combination compatible at the Ag-B locus, strong reactions of Lewis lymphocytes to the dissociated heart and skin cells of the Fischer rat were related to the acute rejection of heart and skin grafts, while negative reactions by the kidney and spleen cells reflected a prolonged survival of the kidney graft. The role of skin- and heart-specific antigens in the rejection phenomenon is discussed.
