Abstract.95 Dutch haemophiliacs were tested for antibodies to membrane antigens on cells infected with human T‐cell leukemia virus type I (HTLV‐I‐MA) by indirect immunofluorescence and to purified HTLV‐I by enzyme‐linked immunosorbent assay. Abstract.The content of microaggregates was determined in leukocyte‐poor red blood cells (RBC) after 5 weeks of storage in citrate‐phosphate‐dextrose (CPD)‐adenine plasma and in saline‐adenine‐glucose‐mannitol (SAGM) medium. The size range was 3,600‐200,000 fl, corresponding to a particle diameter of 19–73 μm, presuming a spherical particle shape. In comparison with CPD‐adenine whole blood (100%), the total number of particles per unit of RBC was found to be 41.2% (CPD‐adenine anticoagulant) and 25.4% (SAGM medium). The corresponding data for total particle volumes were 43.3 and 30.3%. The significant differences between the two categories of RBC could not be explained on the basis of differences concerning pH or cell composition. A majority of particles (50–62%) was observed in the lowest size range (19–29 μm), corresponding to only 18–26% of the total particle volume. The accumulation of di‐2‐ethylhexylphthalate (DEHP) was determined in two variations of RBC stored in SAGM medium and in RBC stored in CPD‐adenine plasma. Abstract.Commercial reagents of non‐human origin were used to provide a room temperature, microtray chromogenic assay for factor VIII. Abstract.The intravenous immunoglobulin (IV. IgG) preparation used in this study is manufactured by the Scottish National Blood Transfusion Service (SNBTS) by the pH 4/mild pepsin method. Recent reports suggest that non‐A, non‐B hepatitis may be transmitted by certain intravenous immunoglobulin preparations. Serum ALT levels were therefore measured prospectively in 16 patients with primary hypogammaglobulinaemia who received an intravenous immunoglobulin replacement therapy (SNBTS IV IgG) over a period ranging from 6 to 25 months. Retrospective analysis of serum ALT levels was also carried out in 8 patients with primary hypogammaglobulinaemia who received fresh frozen plasma (FFP) for periods ranging from 8 months to 13 years. Abstract.Thrombin generation, as evidenced by radioimmunologic fibrinopeptide A (FPA) determination, was studied upon drawing and storage of blood. Blood from 5 healthy subjects was drawn into Fenwal bap (PL 146, 16‐gauge needle, 92‐cm collecting tube), and blood samples for FPA analysis were taken from the proximal and distal part of the collecting tubes. The FPA concentrations were considerably higher in the distal part of the tubes both at zero time and after 5 and 10 min, implying substantial thrombin generation to have occurred upon blood passing through the tubes. In 20 healthy blood donors (age 19–52 years) subjected to a standard collection procedure (Fenwal bags PL 146, CPDA formula‐1 anticoagulant solution, 450 ml blood), higher FPA values were observed in the bags immediately upon collection, as compared to control venipuncture values (p<0.02). Furthermore, in 6 women using oral contraceptives, even larger increases were observed, indicating thrombin generation to be more pronounced in such donors (p<0.01). No statistically significant increase in FPA concentrations was found upon storage of CPDA‐blood at 4°C for 24 h. Abstract.A procedure for preparing single units of freeze‐dried cryoprecipitate (cryo) in the original bags is described in detail. Batches of frozen plasma bags were rapidly thawed by rocking in a water bath, centrifuged and after complete removal of supernate the compact cryo pellets (2–5 g) were lyophilized with the aid of a holding tray with a filtered air chamber designed for this purpose. Adding synthetic amino acids has greatly improved stability of the dried concentrate during storage and has increased its resistance to heat denaturation (e.g. less than 10% loss of potency after 20 h at 70 °C). Abstract.Performance characteristics of pooled rabbit IgG polyclonal anti‐C3d are compared with one mouse IgM and three mouse IgG monoclonal anti‐C3d antibodies (MAs). IgG MA,s employed singly or in combination, failed to precipitate C3d; by contrast, IgM MA and polyclonal anti‐C3d precipitated C3d. Measurements of polyclonal anti‐C3d concentration by chemical means and by125I‐C3d radioimmunoassay (RIA) agreed closely. RIA values were 50% of chemical measurement values for three of the four MAs. Use of sucrose density gradient ultracentrifugation to assess MA C3d/anti‐C3d molar combining ratios for soluble anti‐C3d/C3d was not possible because fast‐sedimenting multimeric C3d/anti‐C3d complexes did not form. Dissociation and competitive binding studies indicate that (1) two MAs haad substantially lower affinities than the other anti‐C3d antibodies, and (2) polyclonal anti‐C3d recognizes more C3d epitopes than are recognized by individual MAs.
