Abstract.The protein composition of 12 different prothrombin complex concentrates, including 2 purified factor IX concentrates and 2 activated fractions, was evaluated. There is a clear difference between ion exchanger‐prepared fractions and those obtained by adsorption onto calcium phosphate. The former contain high molecular weight kininogen and complement components, the latter only trace amounts of these proteins but relatively high quantities of the pro‐ and even partially activated enzymes of the contact phase. 摘要:
Abstract.A modification of the glycine precipitation method for the production of factor VIII concentrate is reported. In this procedure, cryoprecipitate was prepared, washed and dissolved in buffer. Contaminating fibrinogen was precipitated by the addition of 2.8Mglycine buffer at 30°C. The fibrinogen precipitate was removed by filtration using a layer of glass beads. Factor VIII was then precipitated from the filtrate by adding solid NaCl. After centrifugation, the precipitate was resuspended in buffer, desalted, sterilized by filtration, lyophilized and subjected to dry‐heat treatment. The procedure gave a good recovery of factor VIII (280IU/1) and seems applicable to large‐scale production. Abstract.The multimeric structure of von Willebrand factor (vWF) and its ristocetin‐induced binding to platelets, using a simple and very sensitive radiomonoclonal antibody‐labeled vWF method, was compared in normal plasma, single‐donor cryoprecipitate (CP) and five different antihemophilic factor (AHF) concentrates. All the AHF showed a lack of larger vWF multimers, an abnormal ‘triplet’ pattern, and much lower vWF binding to platelets than that of plasma or CP, vWF being the lowest for those with a lesser proportion of larger vWF multimers. Abstract.Red cells collected in CPD, suspended in SAGM solution and stored for 42 days at +4°C, were tested before and after incubation for 1 h at 37°C with respect to autologous 24‐hour posttransfusion red cell survival, morphology, fluidity at low shear rate and adenylate content. Abstract.The bioassay of platelet‐derived growth factor (PDGF) in platelets was established using the rat fibroblast cell line 3Y1. 3Y1 cells (5 times 103/well) were cultured in 24‐well microculture plates with RPMI 1640 medium using 1% of PDGF‐free serum and 5% of platelet extracts for 4 days. Cell proliferation was measured by the increase of DNA content. This assay made it possible to measure the biological PDGF activity. Abstract.Thrombin generation, as evidenced by plasma fibrinopeptide A (FPA) concentrations, was studied during blood collection from donors taking oral contraceptives (OC). 450 ml blood were drawn into Fenwal PVC bags from 26 OC users and 28 nonusers. Blood samples for determination of FPA, beta‐thromboglobulin (BTG), thrombotest (TT), prekallikrein (PKK), antithrombin‐III (AT‐III) and factor VIII procoagulant activity (FVIII:C) were drawn from the bags immediately after ending blood donation and following storage for 24 h at 4°C. The FPA concentrations following donation were significantly higher in the OC than in the control group (p<0.05). The levels of PKK were also higher in blood obtained from OC users (p<0.001), as was the FVIII:C level, the latter difference, however, was not significant (p = 0.06). No cold‐promoted activation of factor VII, as evidenced from TT, was detected following storage at 4°C, neither was any change observed in the FPA, PKK and AT‐III levels. The BTG concentrations increased significantly during storage, most pronounced in the control group (p<0.05). The decay of FVIII:C was similar in the two groups, averaging 24.7%. No correlation was observed between the FPA levels and the other parameters determined. We conclude that thrombin generation is more pronounced during routine blood collection from donor

Abstract.Erythocytes from 3 Gerbich‐negative siblings readily converted to echinocytes when subjected to changing concentrations of sodium or lithium chloride. When erythrocyte membrane proteins were assayed by immunoblotting with human anti‐Ge 1,2, normal membranes were found to contain a reactive component of an approximate molecular weight of 33,000 daltons which was absent from the Gerbich‐negative individuals. Abstract.The human T cell lymphotropic virus type I (HTLV‐I) is transmitted by blood transfusions that contain cell components as one of the virus transmission modes. We carried out mass screening of sera from seropositive donors by a gelatin particle agglutination test to prevent the spread of HTLV‐I by transfusion. Seroconversion rates were compared before and after screening. The results show a remarkable decrease in the rate of infection from 53.6 to 0.9%. Abstract.Density gradient centrifugation has been used in animal systems to purify stem cells and eliminate T lymphocytes prior to allogeneic transplantation. There is substantial disagreement whether the same approach can be used to purify hematopoietic stem cells obtained from human peripheral blood. The purpose of the present study was to resolve that issue by determining the density distribution of 4 classes of human leucocytes: total mononuclear cells, T lymphocytes, CFUc, and BFUe. To ensure a representative sampling, a large number of randomly selected donors were analyzed. The results show that most T lymphocytes band between 1.068 and 1.071 g/ml, with relatively little variation from individual to individual. In contrast, the density distributions of both CFUc and BFUe fluctuated markedly from donor to donor. As a consequence, there was significant variability in the degree of progenitor‐T cell separation.
