SummaryThe beneficial effect of adenine on red cells of blood stored in A.C.D. at 4°C, has been confirmed by post‐transfusion survival studies in rabbits and by biochemical studies of human and rabbit blood. Nevertheless probably this effect of adenine is too unreliable for routine use in the storage of blood for transfusion.In human red cells adenine has a greater effect on intracellular A.T.P. levels than on overall glycolytic rates. This suggests that loss of A.T.P. is not the only factor leading to the failure of red‐cell metabolism during storage.RésuméL'effet bénéfique de l'adénine sur les érythrocytes du sang conservé A.C.D. à 4° a été confirmé par l'étude de la survie des érythrocytes transfusées chez le lapin et par des études biochimiques du sang humain et du sang de lapin. SummaryA special theory of hemagglutination is developed to estimate antigen strength and antibody concentration under antigen excess in an automated system, using either hemolysis or counting of free cells. Antigen strength and antibody concentration are both expressed in terms of A, the number of bivalent antibody molecules in a given volume which under antigen excess are combined with sites on 2 cells. The theory is illustrated by ABO titrations with a variety of phenotypes and reagents.RésuméUne nouvelle théorie de l'hémagglutination est décrite pour estimer la force de l'antigène et la concentration de l'anticorps lorsqu'il y a excès d'antigène dans un système automatique utilisant soit l'hémolyse ou le comptage des érythrocytes non agglutinés. SummaryA new technique for the quantitation of haemagglutination is described. Having obtained equilibrium between antibody, non‐agglutinated and agglutinated red cells in the experimental suspension, a separation of agglutinated and free red cells is performed by a specially elaborated centrifugation method. When separation is completed, the agglutination percentage can be determined after haemolysis of red cells and photometric estimation of the resulting amounts of haemoglobin. The results which can be attained with this simple technique seem to compare with those obtained by using an electronic particle counter, or the Auto‐Analyzer for quantitation of haemagglutination. The method is relatively uncomplicated, and requires no other equipment than that which already exists in most blood‐group laboratories.RésuméUne nouvelle technique pour le dosage quantitatif de l'hémagglutination est décrite. Après avoir obtenu un équilibre entre l'anticorps, les érythrocytes non agglutinés et les érythrocytes agglutinés dans la suspension expérimentale, on procède à une séparation des érythrocytes agglutinés et non agglutinés par une méthode de centrifugation spécialement mise au point. Quand la séparation est obtenue, le pourcentage d'érythrocytes agglutinés peut être déterminé en procédant à l'hémolyse des érythrocytes et au dosage photométrique de la quantité d'hémoglobine libérée. Les résultats obtenus avec cette simple technique peuvent se comparer à ceux obtenus avec des compteurs électroniques des particulcs ou avec l'Auto‐Analyzer pour le dosage quantitatif de l'hémagglutination. La méthode est relativement peu compliquée et n'exige pas d'équipement autre que ceux qui existent toujours dans la plupart des laboratoires de déterminations de groupes sanguins.ZusammenfassungEs wird eine neue Technik zur quantitativen Erfassung der Hämagglutination beschrieben. Sobald sich in der Testsuspension das Gleichgewicht zwischen Antikörper, agglutinierten und nicht agglutinierten Erythrozyten eingestellt hat, werden die agglutinierten Zellen mittels einer speziellen Zentrifugiertechnik von den unagglutinierten Zellen getrennt. Nach erfolgter Trennung wird der Prozentsatz der agglutinierten Zellen an Hand der photometrischen Messung der aus den hämolysierten Zellen freigesetzten Hämoglobinmenge ermittelt. Die Ergebnisse, die mit dieser einfachen Technik erhalten werden, lassen sich durchaus mit jenen vergleichen, die mit elektronischen Teilchenzahl‐Meßgeräten oder mit dem Auto‐Analyzer bei der quantitativen Messung der Hämagglutination erhalten werden. SummaryThe antibody titers of 94 type 0 persons of college age were determined from blood taken during a single three‐day period and the A or O blood types of the mothers of these donors were determined. The titers of the offspring of the type O mothers were almost exactly the same as that of the offspring of the type A mothers. It must be concluded from the data obtained, that the self‐marker concept does not explain the variations which are characteristic of the titers of the naturally occurring antibodies in the ABO blood group system.RésuméLe titre d'anticorps de 94 personnes de groupe O âgées entre 18 et 24 ans a été déterminé à partir de sang prélevé dans une période de 3 jours; les sangs de groupe A ou O provenant des mères de ces donneurs ont également été déterminés. Les titres des descendants des mères de type O étaient, pour la plupart, exactement les mêmes que celui des descendants des mères de type A. SummaryGm(c) was found in 5 of 250 (2%) unrelated Dutch individuals not known to have Negro ancestry. In three cases Gm(c) was determined by a Gmb(1,2,3,4)callele and in one by aGmb(1,2)callele. The genotype of the fifth individual could not be determined from his phenotype. Gm(c) was determined by aGmb(1,2,3,4)callele in eight members of a Dutch family and by aGmb(1,2)callele in six members of a previously reported English family. The alleles which determine Gm(c) in Caucasoids appear to be Gm(b2+), while the allele which determines Gm(c) in Negroids is usually Gm(b2‐).RésuméGm(c) a été trouvé chez 5 sur 250 (2%) Hollandais pris au hasard connus pour n'avoir pas une ascendance de race noire. Summary1Twenty We (γ2b) myeloma proteins from Negroes were examined to determine their Gm types; none of these proteins was Gm(a + b +); all were Gm(a+). Two Vi (γ2c) myeloma proteins were Gm(b+).2This finding, taken in conjunction with the molecular localization of the Gm(a) and Gm(b) factors, suggests that these factors are determined by two closely linked loci, as appears to be the case in Caucasians. Further, the peptide maps of Gra(a+) Negro myeloma proteins were similar to those of Gm(a+) myeloma proteins of Caucasians, both containing a distinctive “Gm(a)” peptide and lacking the “Gm(b, f)” peptide; the peptide maps of Negro and Caucasian Gm(b+) myeloma proteins were also similar.3It appears unnecessary to postulate the existence of aGmaballele in the Negro to account for the fact that 100% of African Negroes are Gm(a + b +).Résumé120 protéines de myélome We (gamma 2 b) provenant de personnes de race noire ont été examinees de façon à déterminer leur type Gm; aucune de ces protéines n'était Gm(a+b+); toutes étaient Gm(a +). SummaryIn the Norwegian population, the chance of exonerating falsely alleged fathers by means of the Gc system is 15.7%. In 1338 paternity cases 138 men out of 1589 were excluded by the Gc serum types. There was a good agreement between the observed and the expected numbers of exclusions within the different kinds of Gc type constellations, and within the different classes of paternity cases.The paternity indices of the Gc system were calculated, and the relationship between the Gc paternity index in one‐man cases and the frequency of paternity exclusion by 4 other serological systems was studied. It was found that the higher the Gc index, the less was the frequency of paternity exclusion by these systems.The confidence of exclusions by the Gc types is of the magnitude of 99–99.9%.RésuméDans la population norvégienne, les chances d'exclusions de pères présumés accusés à faux au moyen du système Gc est de 15,7%. Dans 1338 cas de paternités, 138 hommes sur 1589 ont été exclus par le facteur sérique Gc. Ceci est un bon rapport entre le nombre d'exclusions observées et attendues entre les différentes espèces de la constellation Gc et dans les différentes classes de cas de paternités.Les index de paternités du système Gc ont été calculés et le rapport entre l'index de paternité du système Gc dans les cas d'un seul homme et la fréquence d'exclusion de paternité par quatre autres systèmes sérologiques a été étudiée. SummaryThe immuno‐electrophoretic technique of Gc type determination has been studied, and a standard technique has been developed which provides reproducible Gc patterns. The antisera should preferably give distinct Gc precipitates but few other lines in the alpha2‐globulin area.The reliability of the technique has been tested by duplicate examinations of serum samples, and no discrepancies as to the Gc types have been observed.Observations by earlier authors and in the present study show that the Gc type of a child is developed at birth, and that a change of type in later life by disease or other influence is highly improbable. The Gc characters are not easily destroyed by exposure of blood samples to unfavourable storage conditions within practical limits.It may be concluded that the confidence of Gc type determinations is satisfactory and justifies the application of the Gc system in cases of disputed paternity.RésuméL'analyse immunoélectrophorétique pour la détermination des types Gc a été étudiée, et une technique standard a été mise au point qui permet d'obtenir des lignes Gc reproductibles. Les anti‐sérums doivent, si possible, donner des lignes de précipitation Gc distinctes mais peu d'autres lignes de précipitation au niveau des alpha2‐globulines.La fidélité de la technique a été éprouvée en pratiquant des examens à double avec les échantillons de sérums, et il n'a pas été constaté de discordance dans les résultats des types Gc obtenus.En accord avec les observations d'autres auteurs, la présente étude démontre que le type Gc de l'enfant est développé à la naissance et qu'une modification de ce type au courant de la vie soit par des maladies soit par d'autres phénomènes est hautement improbable. Les caractères Gc ne se détruisent pas facilement lorsque le sang est conservé dans des conditions défavorables.On peut conclure que la sécurité des déterminations des types Gc est satisfaisante et que leur utilisation dans les cas de paternité contestée se justifie.ZusammenfassungDie immunoelektrophoretische Bestimmungstechnik der Gc‐Typenbestimmung wurde untersucht. Es wurde eine Standardtechnik entwickelt, die reproduzierbare Ergebnisse ergibt. Die hiezu verwendeten Antiseren sollten deutliche Gc‐Präzipitationslinien, aber auch einige zusätzliche Linien im α2‐Globulinbereich erscheinen lassen.Die Zuverlässigkeit der Technik wurde durch Doppelbestimmung zahlreicher Serumproben überprüft. Unterschiedliche Ergebnisse wurden nie festgestellt.Beobachtungen früherer Autoren wie auch die hier vorgelegten Ergebnisse zeigen, daß der Gc‐Typ beim Kind bereits anläßlich der Geburt voll ausgeprägt ist. Eine änderung im künftigen Leben, sei es durch Krankheit oder andere Einflüsse, erscheint als höchst unwahrscheinlich. SummaryAnti‐bromelin antibodies prepared in rabbits were employed for immuno‐electrophoretic analysis and in serologic tests to define more clearly the property of bromelin responsible for agglutination of erythrocytes coated with incomplete antibody. The antiserum defined 9 antigenic components designatedathroughi. Immunoprecipitin bandihad no proteolytic or peroxidase activity but appeared to correspond to an electrophoretic component of bromelin that contains both proteolytic and serologic activity. Antiserum was used to demonstrate that antigenic components of bromelin become attached to the red cell and to show that the proteolytic and the sensitized‐erythrocyte‐agglutinating factor (SEAF) of bromelin were neutralized by the anti‐bromelin antibody.RésuméDes immuns‐sérums de lapin contenant des anticorps anti‐broméline ont été préparés; ces sérums ont été utilisés pour pratiquer des analyses immunoélectrophorétiques et des tests sérologiques de façon à déterminer d'une manière plus claire la raison pour laquelle la broméline agglutine les érythrocytes revêtus d'un anticorps incomplet. L'antisérum comportait neuf composants antigéniques désignés de a à i. La ligne de précipitation i n'avait pas d'activité protéolytique ou peroxydasique, mais semblait correspondre à un composant électrophorétique de la broméline contenant l'activité protéolytique sérologique. L'immun anti‐sérum a été utilisé pour démontrer que les composants antigéniques de la broméline pouvaient s'attacher aux érythrocytes et pour démontrer également que l'action protéolytique et le facteur agglutinant de l'érythrocyte sensibilisé (sensitized‐erythrocyte‐agglutinating factor [SEAF]) de la broméline était neutralisé par l'anticorps antibroméline.ZusammenfassungMittels Antibromelin‐Antikörpern vom Kaninchen wurden immunoelektrophoretische und serologische Untersuchungen durchgeführt, die daraufhinzielten, nähere Aufschlüsse über jene Bromelinkomponenten zu erhalten, die für die Agglutination von Erythrozyten, die mit inkompletten Antikörpern beladen sind, verantwortlich sind.
