Summary1A technique is described in detail [12] for detecting serological factors capable of agglutinating platelets at 37°C. It consists of a modification of the agglutination technique described byDausset and Malinvaud[9] in which an agitator was used. The modification consists of heating the platelets to 56°C. for several seconds after they have been washed. It is thought that heat destroys some clotting factors that are present in the area immediately surrounding each platelet.2The characteristics of these agglutinins are as follows: The optimum temperature of reaction is 37°C., and they react best at physiological pH and ionic concentration. They are thermostable. They are inhibited by a thermolabile factor which is probably complement (the same inhibitor as in the case of leucocyte antibodies); but traces of complement enhance the reaction. They are present in the gamma globulin fraction. They cause lysis, and also fixation of complement. The strongest can be also detected by the anti‐globulin consumption test, the mixed antiglobulin agglutination test, or by thromboelastography.3Platelet agglutinins active at 37°C. have been found in 87 out of 1633 pathological sera studied. The role of transfusion has been proved by the fact that the incidence of these antibodies increases from 3.63 % in patients who had received from one to three transfusions, to 47.36 % in those who have received 101 to 200 transfusions. Two patients who had chorio‐epitheliomata developed platelet agglutinins. Pregnancy also plays a part in the development of platelet antibodies. With one exception, platelet warm agglutinins were not found in sera from patients with idiopathic thrombocytopenic purpura.4They are probably immune iso‐antibodies. They do not react with the platelet of the host, and react only with certain batches of platelets. They are not related to any known red cell antibodies, cold platelet agglutinins, or anti‐platelet gamma‐globulin substance detectable by the antiglobulin consumption test in the serum of some cases of thrombocytopenic purpura, pancytopenia, or DLE. They are closely related to leuco‐agglutinins appearing in the same patients.5They may enable us to define platelet groups that are independent of the red cell groups. They may be the cause of the diminishing haemostatic effect of repeated platelet transfusions, of certain transfusion reactions, and perhaps of some cases of thrombocytopenic purpura of the newborn.Réumé1°Il est décrit une technique de recherche des substances sériques capables de provoquer à 37°C l'agglutination plaquettaire. Il s'agit d'une modification de la technique d'agglutination par agitation circulaire deDausset et Malinvaud[9]. La modification essentielle consiste à chauffer à 56°C la suspension de plaquettes lavées. Il est supposé que ce chauffage agit par destruction des facteurs de la coagulation présents dans l'atmosphère plaquettaire.2°Des thrombo‐agglutinines actives à 37°C ont été rencontrées dans 87 sérums provenant de l'étude de 1633 sérums pathologipues. Le rôle des transfusions est prouvé par le fait que la fréquence relative croît avec le nombre des transfusions, passant de 3,63 % chez les malades ayant reçu de un à trois transfusions à 47,36% chez ceux qui ont reçu de 101 à 200 transfusions. Deux malades ont développé des thrombo‐agglutinines au cours d'un chorio‐epitheliome. De même le rôle des grossesses est probable puisque on note une nette prédominance féminine (70,11%) chez les porteurs de thrombo‐agglutinines.3°Les caractères sérologiques de ces thrombo‐agglutinines sont les suivants: Elles ont un optimum d'action à 37°C, sont actives au voisinage de pH et de l'ionisation physiologique. Elles sont thermostables. Elles sont inhibées par un facteur sérique thermolabile qui; est probablement le complément (système inhibiteur identique à celui des leuco‐agglutinines). Elles sont présentes dans les gamma‐globulines. Elles peuvent provoquer également la lyse, la fixation du complément. Elles peuvent êitre décelées encore par la réaction de consommation de l'antiglobuline, par le test mixte d'agglutination par l'antiglobuline ou par la thromboélastographie. Elles n'agissent pas sur les propres plaquettes du donneur et agissent sur certaines variétés de plaquettes. Elles sont sans rapport avec les substances anti‐éry‐throcytaires, avec les thrombo‐agglutinines froides, avec les substances anti‐plaquettaires décelées dans les sérums de purpura thrombopénique non transfusés et sans antécédents de grossesse.Leur rapport avec les leuco‐agglutinines sont étroits: ils apparaissent le plus souvent chez les mêmes malades.4Ce sont vraisemblablement des iso‐anticorps immuns d'origine transfusionnelle (ou éventuellement d'origine gravidique). Ils permettront de définir des groupes plaquettaires qui sont indépendants des groupes érythrocytaires. Ils sont responsables de certains chocs transfusionnels et sans doute de certains purpura thrombopéniques du nouveau‐né.Zusammenfassung1Eine Technik zum Nachweis von Serumfaktoren, welche bei 37°C Plättchen agglutinieren, wird beschrieben. Es handelt sich um eine Modifikation der Agglutinationsmethode vonDausset und Malinvaud, bei welcher ein Schüttelapparat verwendet wird. Die Modifikation besteht darin, daß die gewaschenen Plättchen wahrend weniger Sekunden auf 56°C erwärmt werden. Es ist anzunehmen, daß durch diese Erwärmung an der Plättchenoberfläche haftende Gerinnungs‐faktoren zerstört werden.2Die Plättchenagglutinine zeigen folgende Eigenschaften: Ihr Wirkungsoptimum liegt bei 37°C, einem physiologischen pH und einer physiologischen Ionenkonzentration. Sie sind thermostabil. Sie werden durch einen thermolabilen Faktor, wahrscheinlich Komple‐ment, gehemmt. Derselbe Faktor hemmt auch die Leukozytenantikörper. Spuren von Komplement verstärken hingegen die Plättchen‐agglutination. Diese Agglutinine sind in der Gammaglobulinfraktion des Serums enthalten. Sie wirken lytisch und bewirken die Bindung von Komplement. Hochtitrige Plättchenagglutinine können auch mit dem Antiglobulinkonsumptionstest, der Antiglobulinmischzelagglutination sowie im Thrombelastogramm nachgewiesen werden.3Es gelang, in 87 von 1633 pathologischen Seren wärmeaktive Plättchenagglutinine nachzuweisen. Die Bedeutung der Bluttransfusionen für die Bildung dieser Antikörper ist daraus ersichtlich, daß diese nur bei 3,63% der Patienten, welche eine bis drei Transfusionen erhalten hatten, gefunden wurden, während anderseits 47,36% derjenigen Patienten, welche 101 bis 200 Transfusionen erhalten hatten, in ihrem Serum solche Antikörper aufwiesen. Zwei Patienten mit einem Chorioepitheliom entwickelten ebenfalls wärmeaktive Plättchenagglutinine. Auch Schwangerschaften spielen eine Rolle bei der Bildung von Plättchenagglutininen. Abgesehen von einer Ausnahme wurden wärmeaktive Plättchenagglutinine bei der idiopathischen thrombozytopenischen Purpura nicht gefunden.4Es handelt sich höchstwahrscheinlich urn Isoimmunantikörper. Sie reagieren nicht mit den «eigenen»d Plättchen; sie reagieren lediglich mit bestimmten, nicht aber mit allen Plättchenmustern. Sie haben keine Beziehung zu den heute bekannten antierythrozytären Antikärpern, zu den kälteaktiven Plättchenagglutininen und den durch den Antiglobulinkonsumptionstest nachweisbaren antithrombozytären Gammaglobulinen, welche bei gewissen Fällen von thrombopenischer Purpura, Pancytopenie oder Lupus erythematodes disseminatus vorkommen. Sie sind hingegen nahe verwandt mit den Leukoagglutininen, welche häufig bei denselben Patienten auftreten.5Es dürfte in Zukunft gelingen, mit Hilfe dieser Antikörper gruppenspezifische, von den Erythrozytenantigenen unabhängige, Plättcheneigenschaften darzustellen. Diese Plättchenagglutinine sind wahrscheinlich die Ursache für die Abnahme des hämostatische

Summary1Rabbits were successfully immunized with eluates prepared from human red blood cells.2The presence of demonstrable antibody activity in the eluate was not a prerequisite to provoking an immune response in rabbits.3Immune responses were obtained with eluates from red cells whose sensitization appeared to be non‐gamma globulin in type as well as from gamma globulin sensitized cells.4Weak but definite immune responses were obtained with eluates from normal red cells.5Broad cross‐reactivity was observed among the various anti‐eluate antisera when tested against a panel of cells whose sensitization covered the range of gamma globulin, mixed and non‐gamma globulin types.Résumé1°Des lapins ont été immunisés avec des éluats préparés à partir d'érythrocytes humains.2°Il n'est pas indispensable que l'activité de l'anticorps soit mise en évidence dans 1'éluat pour provoquer une réponse immunitaire chez le lapin.3°La réponse immunitaire a été obtenue à partir d'éluats d'érythrocytes traités, aussi bien par des substances sensibilisantes de type non gamma‐globuliniques que par des substances sensibilisantes de type gamma‐globuliniques.4°Les auteurs ont observé des réponses immunitaires faibles, mais certaines à partir d'éluats d'érythrocytes normaux non traités.5°Après avoir testé les différents anti‐séra anti‐éluat avec différentes catégories d'érythrocytes traités par des substances sensibilisantes de type gamma‐globulinique, mixtes et non gamma‐globuliniques, les auteurs ont observé de nombreuses reactions croisées entre les différents anti‐séra.Zusammenfassung1Es gelang mit Eluaten, die von menschlichen Erythrozyten gewonnen worden waren, Kaninchen mit Erfolg zu immunisieren.2Obschon in einigen Eluaten die Antikörperaktivität nicht nachweisbar war, gelang es trotzdem mit diesen Eluaten, Kaninchen zu immunisieren.3Die Antikörperbildung bei den Kaninchen wurde mit Eluaten beobachtet, die von Erythrozyten stammten, die mit Antikörpern vom Gammaglobulintypus und solchen vom «Nicht‐Gammaglobulin»‐Typus sensibilisiert worden waren.4Selbst mit Eluaten von nicht sensibilisierten Erythrozyten wurde eine schwache, aber deutliche Antikörperbildung beobachtet.5Die einzelnen Anti‐Eluat‐Kaninchenseren reagierten mit Erythrozyten, welche mit Antikörpern vom Gammaglobulintypus

Summary1Positive antiglobulin reactions were obtained with the red cells from 29 of 70 patients with acquired hemolytic anemia tested against serial dilutions of a conventional antiglobulin serum and three antieluate antisera. Of these, 17 were positive with 4 of 4 sera, 3 with 3 of 4 sera, 1 with 2 of 4 sera, and 8 with the conventional antiglobulin serum only.2Marked heterogeneity was observed among the 29 positive antiglobulin reactions. Five general patterns of reaction could be discerned, depending on which of the antisera reacted with the patients' cells and whether a prozone occurred. The patterns did not appear to depend on whether the red cell coating substance was gamma globulin or non‐gamma globulin in type.3Certain broad diagnostic categories (idiopathic acquired hemolytic anemia, chronic leukemia, lymphoma) tended to segregate predominantly into one or two of the major patterns of reaction, but the correlation was not absolute.Résumé1°Sur les 70 patients atteints d'anémie hémolytique acquise, les érythrocytes de 29 d'entre eux donnent des réactions positives à l'anti‐globuline. Les tests ont été pratiqués au moyen de dilutions progressives d'un sérum anti‐globuline ordinaire et de 3 séra anti‐éluat. Parmi les échantillons d'érythrocytes donnant des réactions positives, 17 d'entre eux réagissent avec les 4 séra; 3, avec 3 des 4 sera; 1, avec 2 des 4 séra et 8, avec le seul déum anti‐globuline ordinaire.2°Les 29 réactions positives à l'anti‐globuline ont démontré une forte hétérogénéité. D'une manière générale, les auteurs ont pu dé‐celer 5 types de réactions dépendant de l'anti‐sérum utilisé et de la présence ou de l'absence d'un phénomène de prozone. Le type de réaction ne semble pas dépendre du fait que les érythrocytes soient sensibilisés par des substances du type gamma‐globulinique ou non gamma‐globulinique.3°Certaines catégories d'affections (anémie hémolytique acquise, leucémie chronique, lymphome) ont une certaine prédominace à se classer dans le cadre d'un ou deux types de réactions; toutefois, la corrélation n'est pas absolue.Zusammenfassung129 von 70 Blutproben von Patienten mit erworbener hämolytischer Anämie zeigten im Ansatz mit einem gewöhnlichen Anti‐globulinserum und drei Anti‐Eluat‐Seren positive Reaktionen. Von diesen 29 Proben reagierten 17 positiv mit allen 4 Seren; 3 reagierten positiv mit 3 der 4 Seren; 1 reagierte positiv mit 2 der 4 Seren, und 8 reagierten lediglich mit dem gewöhnlichen Antiglobulinserum.2Die 29 positiv reagierenden Blutproben zeigten eine ausgesprochene Heterogenität ihres Reaktionstypus. Immerhin gelang es, auf Grund der verwendeten Seren und der vorhandenen bzw. fehlenden Prozone 5 Reaktionstypen auseinanderzuhalten. Tanned rabbit red blood cells coated with Leasubstance from human saliva or purified pseudomucinous ovarian cyst fluid produced agglutinating anti‐Leain rabbits. The results again indicate that soluble substance obtained from body fluids and other sources when coated on to tanned red cells can be used to stimulate antibody production.RésuméLes érythrocytes de lapin traités à l'acide tannique puis revêtus de substances Leaprovenant de salive humaine ou de liquide purifié de kyste ovarien pseudomuqueux produisent des anticorps anti‐Leachez le lapin.Ces résultats démontrent à nouveau que des érythrocytes préalablement traités par l'acide tannique et revêtus ensuite de substances solubles d'origine humorales ou autre peuvent être utilisés pour stimuler la formation d'anticorps.ZusammenfassungTanninvorbehandelte Erythrozyten, die mit Lea‐Substanz aus menschlichem Speichel oder aus pseudomuzinösen Ovarialzysten beladen waren, bewirkten bei Kaninchen die Bildung von Anti‐Lea‐Antikörpern.
