Using ethanolic phosphotungstic acid staining, we studied the development of synaptic junctions in the brains of mice subjected to total asphyxia. One‐day‐old mice, Std:ddY strain, were put into a chamber continuously flushed with 100% CO2‐gas for 35 min. The surviving mice (28.1%) were used as experimental models, while unasphyxiated littermates served as controls. The synaptic junctions in the frontal motor cortex and hippocampal cortex were studied at 20 and 60 days of age using E‐PTA staining, and the numbers of synaptic junctions in both areas were counted. In comparison with the control animals, the frontal motor cortex of the subject mice showed a smaller increase in the number of synaptic junctions, both at 20 and at 60 days of age. In the hippocampal cortex, the number of synaptic junctions seen in the experimental mice brains was similar to that in the controls at 20 days of age; however, the number of synaptic junctions in the treated mice brains showed a slower rate of increase than in the controls at 60 days

Ontogenic expression of somatostatin (SRIF)—messenger RNA (mRNA) in the gastrointestinal tract was examined in neonatal rats aged from 1 day preterm to 60 days postpartum in comparison with that in the hypothalamus. SRIF‐mRNA in the hypothalamus was already expressed in prenatal rats and its developmental change was relatively small. In contrast, a unique pattern of SRIF‐mRNA expression was seen in the different intestinal regions, gastric antrum, duodenum, jejunum and colon. In the duodenum, SRIF‐mRNA level was low at birth, markedly increased during the postnatal 3 days and declined to the previous level by day 21. Jejunal SRIF‐mRNA was found in neonates but progressively decreased in a similar way to duodenum. On the contrary, gastric SRIF‐mRNA level, which was low during early development, rose rapidly to a peak on day 21 and gradually declined to an adult level. In the colon age‐related change was not conspicuous, remaining at a low level. These results indicate that (1) expression of SRIF gene in the intestinal tract is regulated by local factor(s) as well as developmental stage, and (2) shift of SRIF‐mRNA pattern occurs during weaning from the duodenum‐dominant infantile pattern to the gastric‐dom

We analyzed the Interleukin‐2 receptor (IL‐2R) expression on fresh lymphocytes from three patients with rubella. Flow cytometric analysis demonstrated that lymphocytes from the patients expressed a considerable amount of IL‐2R/p75 recognized by Mik‐βl monoclonal antibody (MoAb), although they did not react with anti‐IL‐2R/pS5 (CD2S) MoAb. The expression of IL‐2R/p75 reached a peak at the end of the acute phase of illness. Fresh lymphocytes from rubella patients showed a marked proliferative response to IL‐2, which was completely inhibited by Mik‐βl, indicating that their IL‐2R is a functional receptor. The perinatal events of 40 term newborn infants who had severe birth asphyxia were reviewed and correlated with the neurological outcome at a mean age of 2½ years. Severe birth asphyxia was defined as an Apgar Score of 3 at 1 min. The majority (68.5%) of the babies had an abnormal fetal heart pattern before delivery. Persistent fetal bradycardia was most frequently associated with poor outcome. The neurological staging, the need for assisted ventilation for more than 24 hours and severe renal impairment were the best predictors of neonatal death or subsequent poor neurological outcome. The overall mortality was 10% and 15% of the surviving babies had varying degrees of neurological handicap

Progressive increase of the serum pH upon standing and storage in different conditions is documented. Bilirubin albumin titration studies were performed on serum of different pH by Sephadex gel filtration and horseradish peroxidase oxidation methods. Results showed that different pH produced different titration curves and that more reproducible and consistent results were obtained with the peroxidase oxidation titration method. Addition of HC1 and CO2to the serum also produced different bilirubin‐protein titration results. The urinary proteins of seven patients with asymptomatic low molecular weight proteinuria (ALMP) were compared with those of Lowe syndrome and Fanconi syndrome. A characteristic electrophoretic pattern was seen in ALMP by isoelectric focusing. Two dimensional electrophoresis revealed that the ALMP proteins were localized in a more negatively charged region of lower molecular weight and consisted of a smaller variety of proteins in comparison with those from Fanconi syndrome and Lowe syndrome. These results suggest that ALMP might be an isolated form of other types of congenital renal tubular diseases from the viewpoint of protein reabsorption and has a lesser functional defect on the proximal tubules.

The effect of intravenous recombinant human erythropoietin (rHuEPO) by two different dosage regimes was evaluated in 12 anemic children on continuous ambulatory peritoneal dialysis (CAPD). Eight patients (group A) were treated once a week at a mean dose of 89U/kg for 24 weeks. The other four patients (group B) were treated three times a week at a mean dose of 260 U/kg for 8 weeks, and then once a week at a mean dose of 88 U/kg for the next 16 weeks. In group A, the hematocrit (Ht) increased gradually and significantly from 19.9 ± 2.1% to 28.7 ± 1.5% at the 24th week, while in group B, it rapidly increased from 17.9 ± 2.5% to 30.2 ± 4.3% at the 8th week, and thereafter was maintained at a level of approximately 30%. However, there was no significant difference in the Ht between the two groups. In two of the four patients in group B, blood pressure increased from 136/87 mmHg to 168/102 mmHg and from 106/70 mmHg to 132/92 mmHg, at the 4th and the 8th weeks respectively, related to a rapid rise of the Ht. Neither antibody to rHuEPO nor other sides effect were evident. Gradual improvement of anemia by once‐a‐week intravenous administration of rHuEPO is advantageous for the quality of life in children

Twelve‐hour urinary sodium excretion, the number of sodium pump sites (Bmax) and the Na/K flux ratio in erythrocytes were measured in 13 previously diagnosed hypertensive subjects. At the time of the study, six subjects were still hypertensive and showed a significantly lower Na/K flux ratio in erythrocytes than the remaining seven subjects who were normotensive. Bmax was also lower in the hypertensive group compared to the normotensive group, although this was not statistically significant. DNA‐polymerase chain reaction(PCR)‐RFLP(restriction fragment length polymorphism) typing of the DQA1 gene was performed on 39 patients with type 1 diabetes and 30 controls. Analysis of the frequency of the subtypes of DQA1 alleles, using the DNA‐PCR‐RFLP typing technique, showed that the DQA1*0301 subtype was most strongly associated with the disease (97.4% vs 56.7%, R.R. = 19.8, pc<0.00005). These results indicated that DQA1*0301 may determine the disease susceptibility in the Japanese. In addition, we analyzed the frequency of the subtypes of DQA1 genes in the ten patients carrying the DRW8‐DQW8 haplotype, and found that at least eight of them (80%) were classified as having DQA1*0301. The long‐term treatment of a 5 month old boy with precocious puberty secondary to the production of chorionic gonadotropin (hCG) by a choriocarcinoma is described. Of 13 cases of infantile choriocarcinoma reported in the literature, none were successfully treated. The present study describes a partially successful outcome with embolization of a hepatic tumor, irradiation of lung and right submandibular tumors, chemotherapy with methotrexate (MTX), actinomycin D (ACD), cyclophosphamide (CPA) and etoposide (VP16), and splenectomy and hepatic lobectomy.
