ABSTRACTPro-opiomelanocortin (POMC) is the protein precursor to several physiologically distinct peptide hormones. The POMC gene is expressed in several distinct cell types and appears to be under differential regulation depending upon the cell location. The structure of the POMC gene in human, bovine, rat, and mouse is very similar, suggesting that this particular structure has functional significance. The gene is broken by intervening sequences into a 5′ noncoding region, a signal sequence-coding region, and a peptide hormone-coding region. Middle-repetitive DNA sequences are present in both introns of the genes as well as the 5′-flanking regions in all species studied; this too suggests that this structural element has functional significance. The peptide hormone-coding sequences are well conserved among species, whereas the nonhormonal "spacer" sequences are poorly conserved. Differential regulation of the POMC gene appears to occur by the differential recognition of effector molecules. ABSTRACTAmylase-coding sequences were detected in the population of cytoplasmic RNA molecules from cultured rat pituitary GH3cells. The amylase genes in these cells are in a micrococcal nuclease-sensitive, transcriptionally active chromatin conformation. The amount of cytoplasmic amylase mRNA appears to be controlled by small molecules provided by the fetal calf serum component of the culture medium.

ABSTRACTStarting with liver mRNA from cadmium chloride-treated rats, we constructed a cDNA library. A mouse metallothionein 1 (MT-1) cDNA restriction fragment (Durnamet al. 1980) was used as a hybridization probe to isolate from this library a cDNA (p2A10) which is a nearly complete copy of the rat MT-1 mRNA sequence. Southern analysis of rat DNA, using p2A10 as a probe, together with the screening of a rat genomic library, reveal that the rat MT-1 gene belongs to a family of related sequences. The rat MT-1 gene is inducible both by glucocorticoids and by metals.

ABSTRACTA cDNA for mouse hepatic metallothionein I has been cloned into pBR322 (Mbikayet al., 1981). Although this recombinant plasmid (M135) possesses the metallothionein sequence in the same reading frame as that of β-lactamase, it fails to direct the synthesis of a fused β-lactamase-metallothionein protein inEscherichia coli. Another plasmid (M244) was derived from M135 by deleting an internal 390-bp segment made of the 5′ noncoding region of metallothionein, the dG-dC tail, and some β-lactamase sequences. Bacteria harboring the new plasmid now contain in their periplasmic space a cysteine-rich, cadmium-binding protein of 12,000 daltons, as expected. ABSTRACTIn our attempt to make the aid of a computer for the molecular biology laboratory available on a low-cost basis, we have improved the programs that we published recently. We have experienced in our laboratory, and have learned from others, that the average lab has an increasing demand to use a time-sharing-independent computer for the bulk of data handling. A microcomputer can give this independence. At the same time, a microcomputer can serve as a terminal for the few computer jobs, such as data communication or extensive search programs, which have to be conducted on a time-sharing basis. In this paper, we describe the programs which can be executed on an Apple II microcomputer using the Apple Language System. This system gives the researcher the opportunity to learn to handle sequence data, and it also includes a word processing system useful in writing letters and manuscripts and storing text.

摘要:
ABSTRACTcDNAs prepared using poly(A)mRNA from pituitaries and containing the coding sequences for bovine and porcine growth hormones (bGH and pGH) were cloned in bacteria. The primary structures of the peptide hormones derived from the nucleotide sequences of the respective cDNAs show approximately 90% homology. The cloned cDNAs were modified using synthetic DNA to construct expression vectors for efficient bacterial production of the mature animal growth hormones.
