Two brothers, one with hepatic haemosiderosis and the other with hepatic haemochromatosis, died with clinical symptoms of acute abdominal pain and irreversible shock. In view of these deaths, 35 relatives were examined. The iron metabolism was studied by determination of the amount of stainable iron in the liver tissue, the fasting serum iron concentration, the iron saturation percentage of transferrin and the iron excretion in 24‐hours urine following intramuscular administration of 1000 mg desferrioxamine B.A defect in iron metabolism was found in three generations. The in vitro DNA and RNA synthesis of infectious mononucleosis (I.M.) cells have been studied in four patients. For evaluation, monocytoid, lymphocytoid and plasmocytoid I.M. cells were distinguished. Short‐term incubation with H3‐thymidine revealed that the DNA synthesis index was highest in plasmocytoid forms with 21 per cent labelling, followed by monocytoid I.M. cells with 7.5 per cent and lymphocytoid I.M. cells with 1.3 per cent uptake. In vitro RNA synthesis was also characterized by most active rates of incorporation of H3‐uridine and H3‐cytidine in plasmocytoid cell types and a similar drop in the intensity of uptake for the other two forms. The still enigmatic origin and function of these irritation cells of the peripheral blood are emphasized. The rather striking autoradiographic findings of cellular proliferation and active RNA synthesis in these cells reveals properties of immature cells. The previously suggested possibility of intracellular viral synthesis is considered to be rather unlikely from our findings. The present experiments seem to indicate that horse blood is deficient in both Factors VIII and IX, the concentration of Factor VIII varying from 30 to 60 per cent of the normal human levels, and the concentration of Factor IX varying from 50 to 80 per cent, the corresponding concentrations in 12 normal human males varying from 60 to 150 per cent and from 70 to 150 per cent, respectively.The finding that the recalcification time of horse plasma was considerably longer than that of normal human plasma is of interest, since it is known that only very low concentrations of Factor VIII or Factor IX, namely below 5 per cent, affect the results of the testFinally, by using the glass activation test of Margolis, it was shown that activated equine plasma and activated normal human plasma are equally effective in shortening the clotting time of intact human or equine plasma.

The results are reported of a study on the E.S.F. (erythropoietic‐stimulating‐factor) in serum from 47 patients with leucosis and 11 patients with other haematological disorders, and a survey is given of previous studies of the same natureThe analyses were performed over a period of 2 yearsFe59incorporation in the erythrocytes in starved rats was used as a criterion of the E.S.F. content in serumIn patients with chronic lymphatic leucosis and acute leucosis, a reciprocal relationship was found between the E.S.F. titre and the haemoglobin percentage, and a similar reciprocal relationship was found between the E.S.F. titre and the bone marrow erythropoiesis. This must be interpreted to mean that there are other factors which determine these patients' defective erythropoiesisIn patients with chronic myeloid leucosis, low values of E.S.F. were found in serum in spite of often reduced haemoglobin levels. Patients with high titres were observed, however, but in these cases the extramedullary erythropoiesis was inhibited, partly because of splenectomy, partly caused by roentgen treatment of the spleen. It seems reasonable to assume that the extramedullary, often ineffective splenic erythropoiesis requires E.S.F., and this might result in the low serum level and perhaps indirectly deprive the bone marrow of its natural stimula

Two anticoagulants, EDTA and ACD, were compared as to their influence on recovery and survival time of in vitro Cr51‐labelled blood plateletsWith EDTA, pronounced trapping of the platelets occurred during the first 24 hours after infusion, due to anin vitrotime‐consuming process. With ACD, the trapping was small, and maximum recovery occurred much earlier than with EDTA. Consequently, higher recoveries of infused platelets were obtained with ACD than with EDTA. The survical time was approximately the same with both anticoagulantsDuring progressive storage, survival time and maximum recovery both decreased more rapidly with EDTA than with ACDIt is concluded that ACD causes less damage to platelets than EDTA and is superior for platelet transfusion and labell

The possible use of desferrioxamine (DF) to measure body iron stores necessitated studies on the source of the iron excreted in the urine with the compound. DF did not take significant amounts of iron from transferrin because:(1) DF did not affect the plasma iron turnover(2) The excretion of a transferrin‐bound radioiron tracer was not increased by DF(3) Induced elevation of the plasma iron level was followed only by a slight increase in the iron excretion(4) No effect was observed on the incorporation into red cells of a transferrin‐bound radioiron tracer by DFOther studies indicated that iron, in the stage of being incorporated into red cell precursors, was not a probable source of the iron excreted with DFA significant delay in the excretion of iron with DF was regularly observed and found to be due to a delayed mobilisation of ironFurther studies are necessary to find the source of iron excreted after administration of

In 1961 Toohey&Barker reported findings suggesting that 5,6‐dimethylbenzimidazole‐coenzyme B12(DMB‐coenzyme B12) may represent the bulk of the B12content of the human liver. In investigations of the forms of B12in normal human plasma Lindstrand&Ståhlberg (1963) discovered a predominant, labile form of B12, which they provisionally called the “fourth factor”. The finding was followed up by studies on pig's liver, in which they found a form of B12showing the same lability and chromatographic properties as the fourth factor (Lindstrand&Ståhlberg 1963). The fourth factor in pig's liver is probably identical withmethylcobalamine(Lindstrand 1964), and much suggests that this also holds true for the fourth factor in human plasma (Ståhlberg 1964)The finding of the fourth factor in human plasma and pig's liver prompted investigation of the types of B12occurring in the human liver. 摘要:
The auto‐fluorescence of erythrocytes is very weak and erythrocytes are not stained by ordinarily used fluorochromes (ase.g.acridin‐orange). Within the haemoglobin molecule some of the conjugated double bonds of haematoporphyrin are bound to Fe‐atoms, whereby the ability of haematoporphyrin to fluoresce ceases. Heller showed (1916) that it was possible to split of the Fe‐atoms from haemoglobin by means of strong acids without destroying the porphyrin ring or the fluorescence. In this report is described how this method was used to get erythrocytes to fluoresceVenous blood was collected with acid dextrose citrate as anti‐coagulant and diluted by saline to get a 10 per cent suspension of erythrocytes. Smears prepared of this suspension were dried and fixed in aethanol for 10 min. One drop of 20 per cent HCl was placed on the glass slide and covered with a coverslip. The slides were examined using a Reichert Binolux fluorescence microscope (primary filter: Erreger filter BG 12/3 mm; secondary filter GG 9/1 mm + OG 1/1.5 mm; dark field condensor). After ultraviolet illumination in about 15 seconds all the erythrocytes showed a vivid red fluorescence while all the leucocytes and thrombocytes showed no fluorescence at allThe method is suitable to detect traces of haemoglobin and can be used to show the remaining quantity of haemoglobin by preparing erythrocyte ghostsThe method can also be used to show the existence of erythrocytes containing haemoglobin of an abnormal type, if haemoglobin‐A can be removed.
