The metabolic fate of 51Cr-β-glycerophosphate complex, a newly developed radiocompound that accumulates in tumor tissue, has been studied. Determinations of 51Cr uptake and phosphatase activity in the tissues indicate that in tumor the chromium accumulation is not due to simple phosphomonoesterase action. The current approach in managing effusions in the Cancer Research Institute Clinical Unit is as follows. Initial attempts are made to control the process with systemic chemotherapy, using diuretics as indicated. If the patient requires frequent taps, or is symptomatic from his effusions, an alkylating agent is instilled locally. Nitrogen mustard is generally used intrapleuraliy and Thio-TEPA intraabdominally because of ease of use, inexpensiveness, availability, and relative freedom from side effects, as well as their high degree of effectiveness. If two or three instillations fail to control the effusion, the patient is then referred either to the radioisotope or surgery service depending on the case.

1. In an experimental system of implant-induced supernumerary limb formation in the newt, two inhibitors of limb regeneration, X-rays and beryllium nitrate, were individually applied to either the frog kidney implants or to the host limb. 2. Kidney implants irradiated with 2,220 r or 3,000 r induced normal percentages of supernumerary limbs whereas normal implants placed into limbs previously irradiated with 2,000–4,000 r did not stimulate any supernumerary growth. 3. Implants soaked in N/7 beryllium nitrate for two minutes stimulated normal percentages of supernumerary growth, but, when normal implants were placed into beryllium-treated limbs, no regenerative response was noted. 4. Neither X-irradiation nor beryllium nitrate inactivate the ‘factor’ in frog kidney which induces supernumerary limb formation. When host limbs were treated with these inhibitors, dedifferentiation was blocked and a regenerative response did not occur despite the presence of an adequate stimulus in the form of the kidney implant. The author reports the mean DNA values of interphase nuclei in 25 human tumours of epithelial origin, not previously treated. These values are closely related to the chromosome numbers. The constancy of the observed values for each tumour makes it likely that each tumour represents a clone, derived perhaps from one single cell, in which the genetic changes first occurred. The analysis of tumour cell karyotypes indicates, in agreement with DNA estimations, that the mean chromosome size in tumour cells is larger than normal. The data confirm the view that most of the variations present in tumour cells are not only structural but also represent an increase of the active DNA content per chromosome. The practical implications of these findings are pointed out.

The behaviour of benzpyrene sarcoma, HeLa, Landschutz ascites, and Yoshida ascites sarcoma cells implanted into 16–18 h old primitive-streak stage chick embryos has been studied. Normal heterografts such as mammalian liver or pituitary were enveloped by abundant host mesoderm and thus practically isolated. This host mesodermal response (HMR) was not elicited to any significant degree by neoplastic cells. HeLa cells treated with chicken gamma globulin produced HMR with considerably enhanced frequency. Specificity was noticed in the interaction between the tumour cells and embryonic cells. The tumour cells showed mobility in the embryonic surroundings. HeLa and Land-schiitz cells appeared to direct pseudopodial activity specifically towards embryonic mesoderm. HeLa cells lost their mobility upon treatment with gamma globulin. Both normal and treated HeLa cells consistently formed an epithelium-like structure in the neighbourhood of the host epiblast. It has been suggested that such specificity of behaviour may be due to the amount of net negative charge present on the surfaces of the interacting cells. Yoshida ascites cells were found to be widely dispersed in the embryo, but they showed no specificity in their interaction except that they induced proliferation of the host epiblast cells in a small number of case

We inoculated 214 animals with a 20-Methylcholanthrene suspension in olive oil. The first reaction, 3 to 9 days after inoculation, is an intensive epithelial hyperplasia with cellular disorientation, anisocytosis and increased mitotic activity limited to the site of inoculation. Superimposed on this background of epithelial disorganization an inflammatory process is observed both in treated and control animals. Later a perforation of the basal membrane can occur with invasion of muscle and mesonephros or with pseudotumoral nodular formations. In other cases a papillomatous formation is observed. After 20 days a regressive phase of these processes sets in. The larvae of Bufo arenarum respond to 20-Methylcholanthrene treatment very promptly.
