Methods for purifying the human prostate specific antigen are described. The antigen was isolated from human prostate and seminal plasma. Purified antigen (ca. 1 mg/ml) from seminal plasma was immunologically identical and biochemically similar to that of prostatic tissue.

Well-preserved viable cancer cells, lymphocytes and mesothelial cells were prepared from various human malignant and non-malignant pleural effusions by a Ficoll-Hypaque discontinuous gradient. Using 125I-labelled protein A from Staphylococcus aureus as tracer, IgG was found on all cancer cells and also on apparently non-malignant mesothelial cells. Loss of cell surface IgG by overnight incubation, and fluctuation of surface IgG levels in the same patient occurred with cancer cells as well as with mesothelial cells. The IgG in vivo coat of cancer cells reflected the IgG levels of the sera rather than those of the pleural effusions. Binding of IgG from autologous and allogeneic sera and from supernatantsof acid and mild overnight elution of cancer cells and mesothelial cells did not indicate a cancer-specific uptake of IgG. Various unspecific modes of IgG uptake by cancer cells are discussed including the possibility of a non-immune mechanism. Modification and loss of antigens by metastatic cells have also been considered.

The effects of administration of phenformin and clofibrate to 32 breast cancer patients who underwent radical mastectomy and suffered from hormonal metabolic disturbances involving a decline in immunologic response were investigated. It was demonstrated that treatment with these drugs during 2–7 months results in an improvement in metabolic parameters and delayed hypersensitivity reaction to DNCB, tuberculin and candidin (75.5% of cases), an increase in T lymphocyte count (56.3%) and an improvement of the reaction of lymphocyte blast transformation (66.6%). The improvement in the immunologic status of the patients persisted for 6–8 weeks after the stoppage of phenformin administration; a gradual decline in immunologic response and return to the original level were recorded 4–6 months after stoppage of phenformin therapy. The effect of clofibrate on metabolic and immunologic parameters did not manifest itself as soon as 6–8 weeks after stoppage. Elimination of metabolic immunodepression, which gradually develops in the course of normal ageing and tumor process, should be the main objective of metabolic immunotherapy. To this end, therapeutic means, other than phenformin and clofibrate, may be used provided they exert the same effects on carbohydrate-fat metabolism. Circulating immune-complex (IC) levels of patients with breast cancer were measured before and after surgery using a complement-consumption technique. 16 of 26 patients showed raised pre-operative IC values. In 15 out of 16 patients the pathological IC levels decreased within 2–10 days following surgery. Double antibody radioimmunoassay (RIA), using radioiodinated melanoma-associated antigens (MAA), rabbit antiserum raised against 3 M KCl extract of human melanoma (AHMS) and goat antirabbit IgG antibody, was employed for the detection of MAA in tumors as well as in sera of melanoma patients. MAA were partially purified from crude KCl extract of melanoma tissue by affinity column chromatography using AHMS and concanavalin A. A high content of MAA was detected in all but one melanoma extract, while normal tissue and nonmelanoma tumor extracts contained MAA to a much lesser extent than did melanoma extracts. MAA were also found in melanoma patients’ sera (20/45) but not in sera of normal donors (0/10) and colon cancer patients (0/10). A 53-year-old patient with metastatic testicular choriocarcinoma died of renal failure after treatment with cis-platinum and antibiotics. Serum creatinine was first noted to be elevated on day 8 of cis-dichlorodiammineplatinum (DDP) therapy. However, a sharp rise in its level occurred soon after the start of antibiotic therapy with cephalothin, gentamicin and carbenicillin. Renal failure was progressive in nature and persisted until death occurred on day 24 of therapy and after 8 days of antibiotic treatment. The most likely explanation for the progressive worsening of renal function in this patient was renal injury incurred by DDP and antibiotics. The combination of cephalothin-gentamicin should be discouraged in the treatment of patients receiving platinum.

Mice were given either β-carotene or either of two carotenoids with no vitamin A activity – canthaxanthin or phytoene – or placebo. Skin tumors were induced in each group by each of three methods: (1) UV-B (290–320 nm); (2) dimethylbenz(a)anthracene (DMBA)/croton oil applications; (3) DMBA followed by low-dose UV-B. For tumors induced by UV-B alone, β-carotene-phytoene- and canthaxanthin-treated mice developed fewer tumors per mouse, with a delay in tumor appearance, than did control mice. The levels of copper, zinc, calcium, manganese and magnesium have been monitored in the sera of patients suffering from various types of cancer. Only serum copper appeared to be of any diagnostic significance, its levels being above the normal reported range in the breast cancer, leukaemia and Hodgkin’s lymphoma patients. In the case of breast cancer, serum copper is progressively elevated according to the stage of the disease. Serum calcium levels were also significantly lower in patients with tumours of the breast, gastrointestinal tract and cervix. Immunologic concepts have facilitated the recognition within the histiocytic lymphoma group of the Rappaport classification of five diagnostic categories of lymphoma (follicular center cell lymphoma large cleaved and large non-cleaved types, immunoblastic sarcoma B cell type, immunoblastic sarcoma T cell type and ‘true histiocytic lymphoma’). The first four of these ‘large cell lymphomas’ are closely related biologically and are difficult to distinguish histologically. This paper purports to examine the morphologic criteria employed in the differential diagnosis of these four large cell lymphomas in an objective manner, utilizing carefully prepared histologic sections from 24 cases of lymphoma that had been subjected to detailed B and T cell characterization by surface marker methods. 18 morphologic criteria were evaluated by four observers independently. The results show that reliable histologic distinctions can be made on histologic grounds in a proportion of cases only and that individual criteria show more overlap within diagnostic criteria than previously recognized. This is attributed in part to the close biologic relationship of these neoplasms.
