The surface morphology of herpes simplex virus transformed cells was examined by scanning electron microscopy in exponentially growing and density inhibited rat embryo fibroblast cultures. The cell surface of oncogenically-transformed cells became more villated and many microvilli showed branching. C-type virus particles budding from the cell surface were commonly observed. Virus induced cytopathic effects observed by scanning electron microscopy, included rounding up and detachment of degenerating cells from the substrate.

We have attempted to quantitate malignant invasion in vitro. Therefore we adapted the 51Cr-release cytotoxicity test to organotypical culture systems, that proved useful for morphological analysis of invasion. We counted 51Cr-release from labeled fragments of embryonic chick heart and mesonephros, cultured either on semisolid or in fluid medium, during their invasion by HeLa cells. Although demonstrated morphologically, malignant invasion did not bring about significant changes of 51Cr-release as compared to control cultures without HeLa cells. The failure of the 51Cr-release test in our experimental device might shed some light on the mechanisms of malignant invasion.

Peripheral blood lymphocytes obtained from controls and patients with lung cancer treated by radiotherapy were cultured with phytohemagglutinin (PHA). Patient lymphocyte response to PHA was depressed before therapy and even declined further after therapy. There was no prognostic significance associated with pre-therapy PHA responsiveness in this study.

Homogenates of human cancer tissues were assayed for chemotactic activity in modified Boyden chambers utilizing polycarbonate filters with an 8μ pore size. Peritoneal exudate cells (PEC) of guinea pigs were used as indicator cells in the assays and the chemotactic responses of both eosinophilic and neutrophilic leukocytes were recorded. The chemotactic responsiveness of leukocytes to the majority of homogenates was significantly less than that observed with the negative controls. It is suggested that the anti-chemotactic activity observed in vitro may operate in vivo to abrogate inflammatory responses and tumor rejection

Rat kidney cells isolated in vitro at intervals from one to 24 hours after the administration of a single, carcinogenic dose of dimethylnitrosamine (DMN) were characterized by the expression of morphological transformation and a prolonged life span. In cultures isolated four hours or more following the dose of DMN, morphological transformation was expressed at subculture five, but this effect was progressively delayed with earlier times of isolation. Relative to control kidney cell cultures, transformed cells exhibited enhanced proliferative properties evident from assays for plating efficiency and DNA synthesis. The capacity for increased cloning efficiency by the test cultures was acquired several subcultures after the expression of morphological transformation, while ability for colony formation in semi-solid media evolved later still. The results suggest that target cells permanently altered by the carcinogen are present one hour after the administration of DMN, but the effect is more significant by four hours.

Seven patients with multiple myeloma who developed a second neoplasm are presented. There were four patients with acute leukemia and three patients with non-hematologic neoplasms. The patients with acute leukemia were among the longest survivors (median duration ≈ 72 months) and the response to anti-leukemic therapy in these patients was generally poor. Of the three patients with non-hematologic neoplasms, one patient was observed with simultaneous renal cell carcinoma and the other two patients developed adenocarcinoma of the colon and lung subsequently. In addition, two patients with mammary carcinoma who subsequently developed multiple myeloma were included. Prednisone plus vincristine plus daunomycin induced complete remission in nine adults with acute lymphocytic leukemia. The median duration of complete remission will be between 14 and months, with three patients remaining in hematological remission at 10+, 11+, and 18+ months; The median survival will be between and 23 months, with all patients surviving at least one year. This remission induction regimen produced rapid complete remission in adult acute lymphocytic leukemia (ALL) with minimal toxicity, and could be administered predominantly on an outpatient basis.

Several parameters of the biology of cancer of the prostate have been reviewed with a continuing assessment of the possible etiologic role of virus. These aspects include epidemiology, clinical studies, morphology, pathology, enzymology, immunology, endocrinology, model animal studies, in vitro systems, and viral investigations. From available literature it is concluded that, to date, the association with several urogenital tissues of herpes-type viruses has been best documented. It is suggested that a fundamental barrier to more sophisticated virologic and biologic studies is the lack of long term cell cultures of normal and pathologic prostate epithelium from males of all ages.

Methyl-CCNU (NSC-95441) was administered to patients with malignant melanoma in a split dose schedule. Nausea and vomiting were eliminated as clinical problems during therapy. Response rate to the drug seemed somewhat lower than the average of three previously reported disease-oriented phase II trials, though the difference was not significant statistically. The time course of myelosuppression seemed comparable to that seen with a single dose schedule of administration.
