DNA and protein synthesis have been studied in placental villi from normal and pathological cases by in vitro incorporation of tritiated thymidine or tritiated proline and subsequent counting or autoradiography. It appeared that cytotrophoblastic DNA synthesis continued until term and that it was particularly important in preeclampsia cases and in cases of villous immaturity (rhesus sensitization). Protein synthesis was also increased in preeclampsia and seemed to be due to a very active cytotrophoblastic metabolism. The most interesting finding was that in preeclampsia cases, especially when intrauterine growth retardation was superimposed, villous capillary endothelial cell proliferation was as prominent as in cases where villous maturation was not achieved. Such results are highly suggestive of an important compensatory proliferative mechanism in the placentae of preeclamptic women.

The ultrastructural aspects of the appositional phase of interstitial blastocyst implantation in the guinea pig have been investigated using scanning and transmission electron microscopy. Blastocysts measured 28–34 μm in diameter and were encased in the zona pellucida. Trophoblast pseudopodia which penetrated the zona made contact with the underlying uterine epithelium and served, along with uterine epithelial contact with the zona, to hold the blastocyst over the subsequent site of implantation. Pseudopodia emanated from the embryonic pole of the blastocyst measured 2–4 μm in diameter and initiated uterine-blastocyst contact. Pseudopodia contained a cytoplasm rich in tubular mitochondria, rough endoplasmic reticulum, and Golgi apparatus and usually were found directed toward a sub-epithelial capillary plexus. These results suggest that the trophoblast pseudopodia are very active metabolically and may serve to align the blastocyst over stroma regions possessing a rich subepithelial vascular supply and high nutrient availability.

Pre-implantation blastocysts or trophoblast, obtained from ectoplacental cones, were transferred to the right oviducts of both pregnant and nonpregnant mice. Blastocysts which remained within the oviductal lumen did not show evidence of further differentiation into trophoblast although they readily implanted on various extratubal sites. Similarly, trophoblast from ectoplacental cones failed to show invasive properties when restricted to the lumen. In 3 cases where trophoblast giant cells were found within the tubal lumen, entry appeared to have been contingent on an existing breach in the epithelium. Invasive action on tubal epithelium by trophoblast was not seen.

The clearance curves of xenon-133 were registered with an external detector situated above the isolated, dually perfused guinea-pig placenta. The curves were analyzed with a computer program that calculated the parameters of a sum of exponentials. Fetal and maternal flow rates were varied systematically. It was attempted to relate subdivisions of the preparation and their flow rates to the results of washout analysis. No unequivocal correspondence between anatomical and calculated compartments could be detected, but mean calculated and real relative perfusion rates correlated reasonably well. It is concluded that the compartment analysis of xenon clearance curves is not advantageous to other methods.

Individual degrees of anxiety concerning fetal malformations were assessed in early, mid- and late pregnancy by means of self-rating on visual analogue scales. Two samples of women were compared, i.e., 699 women living in an area with a well-established serum alpha-fetoprotein screening program and 239 women living in an area where such a program had never been in operation. At the beginning of pregnancy the distributions of the individual degrees of anxiety were found to be similar in both samples of women. However, in mid- and late pregnancy the anxiety in women from the screening area was found to be lower than the anxiety in women from nonscreened areas.

By sodium chloride (0.66 M) extraction of cervical mucus on an inert chromatographic bed, soluble proteins can be conveniently recovered. Individual proteins such as albumin and IgG may then be quantitated by immunonephelometry. The procedure has been shown to improve reproducibility which is very poor in the presence of mucin. By concentrating the NaCl extract by centrifiltration, proteins present in low concentrations – such as complement C3 – can be measured. The concentrate has also been used for analyses by iso-lectric focusing or gradient gel electrophoresis on polyacrylamide gels.

In a multicentric study, the effect of the antiserotoninergic agent metergoline was evaluated in the management of patients with idiopathic normoprolactinemic secondary amenorrhea (NSA). The awareness that psychological factors might lead to a spontaneous reappearance of menses was also taken into account, and all the patients, after physical, gynecological and laboratory examinations, and the performance of the progesterone withdrawal bleeding test (100 mg i.m.) and the clomiphene citrate test (100 mg p.o./day for 5 days), were treated for 60 days with placebo; only patients showing no menses during placebo administration were later treated with metergoline. 108 patients entered the trial: of these, 48 experienced menses on admission or during placebo administration, and were withdrawn. Of the 60 patients not responding to placebo, 50 were treated for 90 days with metergoline (4 mg t.i.d.), and 23 had menses, ovulatory in 68.4% of cases. A new placebo treatment was accompanied, in the majority of cases, by recurrence of amenorrhea. These results indicate that many patients with NSA may experience a spontaneous disappearance of the disease: in cases more seriously affected metergoline might be a useful therapeutic agent.
