Paradoxical hypersecretion of human growth hormone (HGH) followed a glucose load in patients with endometrial cancer and atypical hyperplasia of the endometrium. All patients had normal fasting plasma glucose and serum HGH concentrations with a normal glucose tolerance curve. Their urinary estrogen concentrations were within the normal postmenopausal range. 6 months after abdominal total hysterectomy and bilateral oophorectomy, the glucose load was repeated. Following surgery all patients showed a normal suppression of HGH secretion during the hyperglycemic period. These findings suggest the existence of a HGH-releasing factor in the neoplastic and preneoplastic endometrial tissue. This factor might be related to the abnormal HGH responses observed.

Intravenous glucose tolerance tests (IVGTT) with simultaneous assessment of plasma insulin and analyses of the fatty acid composition of serum lecithin and cholesterol esters were performed in 11 women with previous gestational diabetes before and repeatedly during 6 months’ administration of a low-dose progestogen (lynestrenol = LYN). 8 of these women were also followed in an identical manner during 6 months of nonhormonal contraception (intrauterine device = IUD) and additionally 6 of these women were followed also during the use of a combined oral contraceptive (OC) (ethinyl estradiol + lynestrenol = EE + LYN). LYN did not alter the IVGTT or plasma insulin but decreased the proportion of polyunsaturated fatty acids (PUFA) in serum lecithin (p < 0.01) and cholesterol esters (p < 0.01) where oleic acid was reciprocally increased (p < 0.05). After 6 months’ use of IUD, on the other hand, thek value of IVGTT increased by 45% (p < 0.01) without significant changes in plasma insulin. In both lecithin and cholesterol ester PUFA increased (p < 0.05) and cholesterol ester oleate decreased (p < 0.01); i.e., virtually the reversal of the changes seen during LYN administration. The combined OC, EE + LYN, caused a decrease of the k value by 27% (p < 0.05) which was apparent even when compared to the effects of LYN alone. EE + LYN also increased (p < 0.05) lecithin palmitate and decreased stearate (p < 0.05) and had a concomitant tendency to lower PUFA and increase oleic acid in both lecithin and cholesterol esters. These results indicate that LYN has little influence on the glucose tolerance in women predisposed to diabetes but may provide poorer conditions for dietary treatment of subclinical diabetes than do nonhormonal IUDs. The combined OC, EE + LYN, on the other hand, promptly diminishes glucose tolerance and may also have an unfavorable influence on liver metabolism.

Human ovaries from adnexectomy were perfused via the ovarian artery with constant volume of Thyrode’s solution. Perfusion pressure was recorded while doses of PGF2α or PGE2 (2.5, 5 or 10 μg/min) were infused for 10 min. During the 10-min infusion period an increase in perfusion pressure (Δ mm Hg) was observed in proportion to the doses of PGF2α- PGE2 in 2.5- or 5-μg/min doses was ineffective, but its 10-μg/min dose increased the perfusion pressure. However, PGE2 (4 μg/min) infused continuously into the ovary inhibited the vasoconstriction induced by PGF2α, whereas the vasoconstrictor effect of noradrenaline remained unchanged.

We perfused in vitro the placentas of the mothers of 3 small for gestational age (SGA) and 3 adequate for gestational age (AGA) infants. After a systemic injection of 10 mg of dehydroepiandrosterone sulfate (DHEA-S), we measured the time course of the levels of DHEA-S, unconjugated DHEA, Δ4-androstenedione (Δ4-A), testosterone (T), estrone (E1) and estradiol (E2) in the perfusate. We observed a significant accumulation of Δ4-A + T and a concomitant low increase of E1 + E2 in the placental perfusion fluid of SGA infants. We conclude that the aromatase activity could be decreased in some cases of placental insufficiency. This could, at least in part, explain the accumulation of circulating androgens in the mothers of some SGA infants following DHEA-S loading test observed in vivo.

The influence of terbutaline on content of phospholipids and their fatty acid composition in fetal lung wash and lung homogenates was studied in 28-day-old rabbit fetuses. To obtain both the fatty acid composition and the quantity of lecithin and sphin-gomyelin gas-liquid chromatography (GLC) was used, which gave a linear relationship when compared to standard solutions of the phospholipids. The amount of lecithin was significantly increased in the lung wash in terbutaline-treated fetuses compared to controls, while the sphingomyelin content was unchanged. In the lung homogenates no quantitative differences were noted between the two groups. The fatty acid composition of lecithin and sphingomyelin did not alter neither in lung wash nor in lung homogenate after terbutaline administration. The present results indicate that beta-mimetics mainly act by favouring the release of preformed surfactant and not by stimulating the synthetic capacity of type II cells in the alveolar lining layer.
