The study deals with 9 women with recurrent abortions who do not possess the blocking factor which is present during normal pregnancy and which inhibits, nonspecifically, a cellular cytotoxicity reaction in vitro. Electrophoresis of the women’s serum revealed an abnormal glycoprotein, migrating in the α-globulin zone, which gives a precipitate with an anti-γ-globulin serum after mercaptoethanol treatment. These last two findings may indicate a possible abortion.

Prostaglandin E2 (PGE2), but not oxytocin, treatment to induce parturition brought about an increase (p < 0.001) in maternal serum levels of total estrogens (conjugated and nonconjugated estriol). There was no increase in nonconjugated estriol. To explore if the changes were related to an altered enterohepatic circulation, cholic acid assay in maternal serum was used. There were no changes in cholic acid which parallelled those in total estrogens. It is concluded that PGE2 does not appear to affect enterohepatic circulation of steroids, and it is suggested that the increase in total estrogens may be caused by changes in the kidney excretion of estriol.

We studied the effect of obstetric analgesia on maternal plasma levels of immunoreactive endorphin peptides (ir-EP) during labor and the postpartum period in three groups of parturients: group I (n = 22) had no analgesia, group II (n = 20) received pethidine intramuscularly, and grop III (n = 10) had continuous epidural analgesia. Initial levels of ir-EP were similar in all three groups. Patients without any medication and patients on pethidine showed a significant rise in ir-EP in late labor and at delivery. Epidural analgesia was characterized by constant levels of ir-EP during labor and an insignificant rise at delivery.

The myometrial response to prostaglandins (PGs) before labor has been studied earlier in this laboratory by the use of a superfusion technique. In the present investigation the same technique was used to explore the contractile effect of PGE2 and PGF2α on myometrial strips obtained during active, spontaneous labor. Both PGE2 and PGF2α had a marked stimulatory effect on strips from the upper uterine segment. The lower segment reacted to PGE2 by inhibition but not at all or slightly stimulatory to PGF2α Oxytocin constantly induced an excitatory response in both segments The results support the concept of a prostaglandin-mediated ‘fundal dominance’ during labor and indicate that PGs contribute to a relaxation of the lower uterine segment. Taken together with previously obtained data the present results provide evidence for a critical change in myometrial responsiveness to prostaglandins by the time of parturition.

The clinical efficacy of CU 32-085, a novel ergoline derivative, in puerperal lactation inhibition was investigated in 41 healthy postpartum women and compared with serum concentrations of prolaction (PRL) and CU 32-085. The women received 0.5 mg CU 32-085 twice daily for 14 days. In 40 women (i.e. 97%) treatment with CU 32-085 successfully inhibited lactation; slight to moderate breast activity appearing during the first few days receded spontaneously. 1 woman experienced full milk let-down on day 5 of treatment. No rebound lactational phenomena were recorded. 5 h after the initial dose, mean serum PRL concentrations decreased by 82% (from 169 to 30 ng/ml), and PRL values remained at that level during the course of treatment. The mean serum concentrations of CU 32-085 showed an inverse pattern compared to PRL. CU 32-085 was well tolerated, with transient slight to moderate dizziness, headache and nausea reported by 12 women (29%) following the initial dose. These disappeared after an average of 3 h. Routine safety chemistries remained within the normal range, and no statistically significant differences between the treatment group and the control group (25 normal nursing postpartum women) were encountered. It is concluded that CU 32-085, a potent dopamine agonist, is an effective and well-tolerated treatment in the indication of postpartum lactation inhibition.

Human plasma β-endorphin (β-EP) and β-lipotropin (β-LPH) were measured simultaneously by radioimmunoassay in 4 women with regular menstrual cycle (4 follicular phase and 4 luteal phase), 5 normal men and 10 term fetuses. Mean plasma β-EP level in the totalsamplesof 8 women was 30.7 ± 5.8 pg/ml( ± SE), significantly (p < 0.03) higher than the β-EP level in men (12.2 ± 2.0 pg/ml). There was no significant sex difference in fetal plasma β-EP level. These data suggest that the existence of sex differences for β-EP secretion and release, and opioid peptides secretion and release mechanism might change with sexual maturation, due to changing sex steroids secretion.

Semen for use in artificial insemination must be of good quality. To satisfy this demand we have to be able to predict the quality of semen from individual donors. Statistical analyses of data on 3,418 ejaculates from 87 donors demonstrated that each donor’s ejaculates could be characterized by a lognormal distribution of the sperm concentration, the concentration of motile spermatozoa before freezing and the concentration of motile spermatozoa after thawing. Furthermore, the ratio of the concentration of motile spermatozoa before freezing to the total sperm concentration and the fraction of motile spermatozoa surviving the freezing procedure were found to be constant for individual donors. Based on these observations a statistical quality control scheme for the selection of donors and for the continuous control of semen quality is presente

In 121 women attending the infertility clinic between 1978 and 1982, 127 cycles with luteal phases of 10 days or more were investigated by a complete basal body temperature (BBT) chart, three plasma progesterone assays taken between 11 and 4 days before menses, and a dated endometrial biopsy (EB) taken at the time of the last blood sample. All parameters were normal in 78 women (64.4%) categorized as group I ‘normal luteal phases (NLP)’. There were some discordances between the three parameters in the remaining 43 women: 8 had normal BBT but low progesterone levels and delayed EB. They form group II ‘inadequate luteal phases’. 29 women (23.9%) had delayed EB despite normal progesterone levels: forming group III ‘dysharmonic luteal phases (DLP)’. 6 women had advanced EB and normal progesterone levels: group IV. Apart from endometrial maturation, there was no difference between NLP and DLP, neither in clinical parameters (length of the follicular phase, date and duration of the thermal shift, length of the luteal phase), nor in progesterone levels. Fertilization occurred in four DLP biopsied cycles: pregnancies went normally to term, giving birth to normal babies. Therefore, the DLP syndrome, characterized by normal midluíeal progesterone levels and a delayed endometrial development, is not incompatible with normal nidation and gestation.

Wet weight of the vagina increased by more than two-fold following treatment of the ovariectomized rabbits daily for 7 days with 100 μg/kg estradiol- 17β or estriol. The concentration of estrogen receptors expressed on the basis of milligrams of protein in both the cytosolic and nuclear fractions decreased significantly after treatment with estradiol or estriol. The data reinforce the view that estriol given in low doses over a long period of time has as good an effect as estradiol on the vagina, and its use might be safer with respect to the effect on endometrial proliferation.
