In thenudemutant, a well-known model of athymia in mouse, abnormalities have been reported in the cerebellum, including a 45% reduction in cerebellar size and neuronal loss (Henderson et al. 1981). The present work was aimed at reappraising the claimed anomalies by using quantitative cytological techniques. We counted the Purkinje and the granule cells, on serial sagittal sections stained with thionin, in the cerebella ofnudemice (nu/nu) compared with their Balb/c controls (+/+). The latero-lateral extent of the cerebellar cortex was statistically different (mean±standard deviation) between thenude(7.05±0.45 mm, n = 3) and the control mice (8.05±0.35 mm, n = 3) (Student'st-test,p<0.05). The number of Purkinje cells was not statistically different (p= 0.98) between thenudemice (159,880±5,225, n = 3) and the control ones (160,060±12,300, n = 3). The number of granule cells (x 106) did not differ significantly (p= 0.40) innudemice (m = 12.1±1.8, n = 3) from the controls (m = 13.2±0.8, n = 3). Immunohistochemistry with a specific marker of Purkinje cells, calbindin protein, did not reveal any qualitative differences between thenudeand the control Purkinje cells. The only slight but significant difference concerned the mean cross-sectional area of the Purkinje cell somas: 188.2μm2(n = 149) in the control animals and 176.3μm2(n = 150) in thenudemice, (p<0.01). In conclusion, our data indicate only a mild effect, if any, of thenudemutation upon the cerebellar neuronal population.

We have shown that female-specific functions of the sex determination geneSex-lethal(Sxl) regulate sexual behavior and synthesis of the three major sex pheromones that have been identified in normal, sexually matureDrosophila melanogastermales and virgin females. Diplo-X flies, heterozygous intransfor two partial loss-of-functionSxlmutations, elicit less courtship than normal females and produce large quantities of the inhibitory pheromones that normal males synthesize. In addition, the mutant flies fail to synthesize the female-predominant aphrodisiac pheromone or make very small quantities of this compound.

RearingDrosophila melanogasterin constant darkness (DD) for multiple generations disrupts the circadian activity rhythm of adults. In order to determine under what conditions normal rhythms can be initiated, DD-rearedDrosophila(either the wild type or theperiodshort[perS] mutant) were exposed to light either as embryos, thirdinstar larvae, or adults. Exposing DD-reared flies to light as embryos or larvae had no effect, while exposing them as adults fully restored normal rhythms inperSand partially restored normal rhythms in the wild type. The percentage of adults with normal rhythms was not significantly different between animals given a 1-h pulse of light as adults and animals given two LD cycles as adults. LD-reared and DD-reared animals given 2 LD cycles were synchronous. In the latter, offset of activity followed the LD transition (CT 12) by 2–6 subjective hours inperSand 2–3 subjective hours inper+. Circadian rhythms did not exhibit phase coherence in the other treatments.

We found a new clock mutation,ritsu (rit), in an inbred strain separated from a natural population ofDrosophila melanogasterin Yamaguchi, Japan. This mutation lengthens the free-running period of the wild-type locomotor activity rhythm from ca. 24 hr to ca. 27 hr by lengthening both active and rest phases equally. To determine the loci involved, we established eight lines of chromosomal substituted flies between the inbred strain andCanton-S, and found that a major gene is located on the second chromosome. Furthermore, the result of recombination analysis suggested that the locus might be near or to the left ofcl(2-18.8).

In the neurological mutant mouseweaver(wv/wv), the majority of males are infertile due to hypospermatogenesis. Heterozygous weaver mice (wv/+) cease mating successfully when males reach an average age of 3.5 months. The contents of epididymal fluid were scored for the number of sperm and sperm motility inwv/wv,wv/+ and controls. Testes were examined in mice of the three genotypes at various ages using light and electron microscopy. Inwv/+ males, sperm counts were significantly lower than in controls and were significantly higher than inwv/wv.The seminiferous epithelium in weaver mice appears depleted soon after puberty and a wide range of degenerative changes was identified in both germ cells and Sertoli cells. Analogous cellular aberrations were detected in heterozygous males, but they appeared at an older age and were not as severe as inwv/wv.We hypothesize that in weaver homo- and heterozygosity the damage of Sertoli cells may induce degeneration of germinal cells and particularly affect the most advanced spermatogenic cells.
