ContentsIn vitro production of cattle embryos both from slaughterhouse material and live animals has progressed immensely during the past 5–10 years. It is now possible to achieve a blastocyst rate of 25% of the proportion of oocytes submitted to culture and when transferred these embryos have the same capability to develop into normal offspring as embryos produced in vivo. Improvement of the system is still needed, such as reduction of the oxygen tension and concommitant abolition of coculture, in order to minimize the risk for the formation of oxygen free radicals, which are known to cause damage to the early embryos. This would be of further advantage for avoiding transmission of contagious diseases and thus place less health hazard on the in vitro conceived embryos. Modifications of certain energy (glucose) – and protein substrates in the media may also improve the systems. The detrimental effect of freezing and thawing on in vitro produced embryos is of great concern, since it precludes a more widespread use of this technology. Although pregnanciees have been established also in our laboratory more research is needed in this essential part of the in vitro production procedure. Oocytes and in vitro produced embryos may be used for cloning by nuclear transfer, and although some progress has been made in this field the results are far from satisfactory. A simple calculation shows that considerable manpower must be involved just for production of a small number of cloned embryos per week and with the current freezing success they may not tolerated deep freezing and for recloning. In vitro produced embryos can be used for microinjection and as a mean of either substituting or supplementing superovulation e.g., in animals where this procedure has not been successful or in animals with sterility problems. The advance in ultrasound guided aspiration of oocytes from unstimulated animals has facilitated the use of in vitro embryo production as an alternative to normal embryo recovery. In the future cattle breeders may use these new technologies along with PCR on preimplantation embryos not only to determine their sex but also to select for other traits.InhaltIn vitro‐Techniken an Rinderembryonen und deren besondere Bedeutung für die RinderzuchtDie in vitro Produktion von Rinderembryonen bei denen die Eizellen sowohl von Schlachthofmaterial als auch aus lebenden Tieren gewonnen werden, haben in den vergangenen 5–10 Jahren stark zugenommen. Es ist möglich geworden, eine Blastozystenbildungsrate von 25% zu erreichen, bezogen auf die Eizellen, die kultiviert werden. Diese Embryonen besitzen die gleiche Entwicklungspotenz wie in vivo produzierte Embryonen. Trotzdem ist eine weitere Verbesserung des Kultursystems notwendig. Beispielsweise ist die Senkung der Sauerstoffpannung und der gleichzeitige Verzicht auf die Kokultur notwendig, um die Bildung freier Sauerstoffradikale zu verhindern, von denen bekannt ist, daß sie den frühen Embryo schädiyen können. Dabei würde gleichzeitig die Chance einer Krankheitsübertragung reduziert und dadurch weniger Gesundheitsrisiken für die in vitro erzeugten Embryonen bestehen. Änderungen bestimmter Energiequellen (Glukose) und Proteinsubstrate können evtl. eine weitere Verbesserung des in vitro Systems bewirken. Die schädigen den Auswirkungen von Tiefgefrierung und Auftauen auf in vitro erzeugte Embryonen ist von entscheidender Bedeutung, da sie eine weitere Verbreitung dieser Technik behindern. Obwohl wir einige Trächtigkeiten erzeugcn konnten, ist eine Bearbeitung dieser wichtigen Phase der in vitro Produktion erforderlich. Eizellen und in vitro produzierte Embryonen könnten für Klonierungen durch Kerntransfer benutzt werden. Eine einfache Kalkulation zeigt aber, daß ein erheblicher Aufwand betrieben werden muß, um nur eine kleine Zahl khnierter Embryonen pro Woche zu erzeugen. Für die z. Zt. übliclierweise verwendeten Gefrierverfahren und/oder für eine Reklonierung sind diese Embryonen dann noch nicht einmal geeignet.In vitro erzeugte Embryonen können für Mikroinjektionen benutzt werden, um beispielsweise als Ersatz oder Ergänzung für Superovulationsverfahren bei den Tieren zu dienen, bei denen diese Stimulationen leicht erfolgreich sind bzw. bei Tieren mit Sterililätsproblemen. Die Vorteile einer durch Ultraschall kontrollierten Eizellaspiration von nicht stimulierten Tieren hat die in vitro Produktion von Embryonen erleichtert und kann als Alternative zur normalen Embryogewinnung angesehen werden. Zukünftig werden die Züchter auf diese Techniken zurückgreifen, auch in Verbindung mit der PCR‐Methode, und nicht nur um das Geschlecht der Präimplantationsembryonen zu bestimmen, sondern auch um

ContentsThe aim of the present report is to describe the procedure followed for sexing embryos on the field using the french DNA probe (INRA patented).The procedure was first tested in conjunction with Rhone Merieux (G. Kohen), the biopsies were performed in farms and sent to the Central Laboratory of ours in Maisons Alfort for sexing. The second step which started in early 1991 is to sex the embryos in laboratories included in the AI Centers facilities, close to the farms, where the embryos are collected and biopsied. The results reported here however are essentially issued from the first step of the experiment. From 241 biopsied embryos, 88% had their sex determined in the laboratory, and 8% were questionable at that stage. The accuracy of unquestionable sex determined embryos was under these circumstances 100%. The sex ratio from the 212 sexed embryos was 48% females, 52% males. From the 61 biopsied embryos that were since then born, the calving rate was 54% (fresh embryos). In addition, some in vitro fertilized embryos (n = 53) were also sexed and 36% of them were females.InhaltGeschlechtsbestimmungen an Rinderembryonen unter Feldbedingungen mit Hilfe einer DNA‐SondeGegenstand des Berichtes ist die Beschreibung des Embryo‐Sexing unter Feldbedingungen mit Hilfe einer in Frankreich hergestellten DNA‐Sonde (INRA patentiert). Der Vorgang wurde zunächst in Verbindung mit Rhone Merieux (G. Kohen) getestet. Die Biopsien wurden auf dem Hof durchgeführt und die Proben in unserem Zentral‐labor in Maisons Alfort zur Geschlechtsbestimmung eingeschickt. In einem zweiten Schritt, der 1991 begann, wurden die Embryonen vor Ort in den Labors der Besamungsstationen, die nahe der Höfe liegen, gewonnen und biopsiert. Die Ergebnisse, die hier vorgestellt werden, beziehen sich aber vorwiegend auf den ersten Abschnitt. Von 241 biopsierten Embryonen wurde bei 88% das Geschlecht bestimmt, 8% waren zu dem Zeitpunktfraglich. Die Gesamtzahl der eindeutig bestimmten Embryonen betrug 10%. Das Geschlechtsverhaltnis von 212 gesexten Embryonen war 48% weiblich und 52% männlich. Bei 61 biopsierten Embryonen, die seitdem geboren wurden, betrug die Abkalberate 54% (frische Embryonen). ContentsThe use of ultrasonography in cases of embryo transfer in cattle provides valuable additional information. Indications are the examination of uterus and ovaries, control of superovulation, ultrasound‐guided follicle punction for oocyte collecting, pregnancy diagnosis from day 20 and monitoring of early pregnancy for occurrence of embryonic death. Especially the ultrasound‐guided follicular aspiration of oocytes seems to be a successful path for a avoiding superovulation.Inhalt: Die Sonographie in der gynäkologischen Diagnostik unter dem Aspekt des EmbryotransfersDurch die Sonographie können für Arbeiten in Zusammenhang mit dem Embryotransfer, wie am Beispiel des Rindes gezeigt wurde, wertvolle zusätzliche Informationen erhalten werden. Indikationen für den Einsatz von Ultraschall sind die Funktionsüber‐prüfung von Uterus und Ovarien, die Kontrolle der Superovulation, die Oozytengewinnung unter sonographisch überwachter Follikelpunktion mit transvaginalern Zugang, die Graviditätsdiagnose ab dem 20. Tag und die Überwachung der Frühgrauidität, wobei vor allem das frühzeitige Erkennen einer embryonalen Mortalität von Bedeutung ist. Besonders die transvaginale Follikelpunktion zur Oorytengewinnung scheint ein vielversprechender Weg zu sein, wenn eine Superovulation vermieden werden soll oder die betreffenden Tiere aufeine hormonale Stimulat
