Cyclosporine (CSA) is an effective immunosuppressive agent and is used in tissue transplantation. The present investigation was undertaken to determine the effect of topical delivery of CSA on allergic contact dermatitis in guinea pigs, Topical 15% CSA in an azone (l‐dadecychlohepatan‐2–one)containing vehicle blocked local elicitation in previously dintrochlorobenezene(DNCB) sensitized animals that received a single topical application just prior to elicitation. Elicitation was not blocked at a distant site, indicating a local effect of topical CSA. In contrast, topical CSA when applied twice daialy for a total of 5 applications during sensitization only, did not block subsequent elicitation. Various concentrations of NiCl. in a hydrogel were evaluated as a possible alternative to the standard patch test material of 5% NiSO4pet. Patch test responses were recorded on a total of 430 patients with known or suspected contact allergy. A Nicl2 concentration of 1% or less in the hydrogel failed to elicit a response in some patients who reacted to 5% NiSOi2 pet. The 2% NiCl2hydrogel produced a small increase in response frequency and may reduce false‐negative reactions. Along with the ability of the material to improve the topical bioavaitability of nickel ions, more irritant reactions were observed. Preparations of 2‐n‐octyl‐4–isothiazolin‐3‐one (Skane M‐8® at 1000 p.p.m. a.i. and 500 p.p.m. a.i. in pet. induced allergy in a cumulative insult human patch test. Challenge patch testing using 250 p.p.m. a.i. Skane M‐8 elicited allergy in 6 out of 6 sensitized individuals. Sensitized individuals did not cross react with Kathon® biocide, (Kathon CG). a mixture of 5–chloro‐2–methyl‐4–isiothiazolin‐J‐one and 1 methyl‐4–isothiazolin‐3–one. All pretrolatum preparations to which Tween 85® was added at 2.5% produced mild irritation in cumulative insult patch tests, but no irritation was observed with Tween 85 at 0.625%. Experimental sensitization of guinea pigs with 6 azo and anthraquinone dyes used frequently us disperse dyes for fabrics (stockings, panty hose) revealed that the blue dyes were moderate sensitizers while the red, yellow and orange dyes were only weak ones. This was found to be in contrast with the order based on the frequency of observed allergic sensitization. which showed the yellow and orange dyes at the top of the list and the blue dyes at the bottom.

Observation of an individual with immediate‐type reaction following exaggerated dermal exposure lo hypochlorite‐containing cleaning products prompted review of similar hypersensitivity reactions attributed to hypochlorite or other highly reactive chemicals. This review confirms isolated incidences of hypochlorite sensitivity of the delayed type (allergic contact dermatitis), as well as immediate‐type reactions from inhalation or topical challenge of sensitized individuals. We conclude that it is possible that excessive and prolonged exposure to hypochlorite may in some cases result in irritation and damage to the skin. Benzocc acid (BA) and the sodium salt of pyrrolidone carboxylic acid (NaPCA) were tested in 13 healthy persons to obtain information about, firstly, the irritant properties of NaPCA and, secondly. the reactivity of various skin sites. BA at 16, 8, and 4 mM pet., anil NaPCA at 50, 25, 12.5, and 6.25% in water, were applied to the forehead, check, neck and upper back Erythema reatt.imis were observed visually, and the changes in ihe skin blood How were monitored using a laser‐Floppier flowmeter. BA at 16 mM mcreused Wood flow on the cheek of 12 tesl subjects, and on the neck, forehead and upper back of 6 subjects, but S and 4 mM BA elicited reactions only on the cheek. 7 resin acids, 3 symbolically prepared derivatives and the neutral fraction of Chinese colophony were studied by experimental sensitization using a modified FCA method. 4 resin acids, laevopimaric. abietic. podocarpic and tetrahydroabietic. proved to be weak sensitizers Neoabietic, dehydroabietic, isopimarie acid and larixol remained negative. However, the derivative, methyl abietate and calcium abietate. were shown to be moderate sensitizers. and the maleic‐modified adduct to be a moderate to strong sensitizer. 4 strips of 5 Epquick™ patch tests (9 allergens plus (control) were applied symmetrically in duplicate on the left and right sides of the upper back in SOU consecutive patients 70 positive allergic patch test reactions were recorded on the left side and 67 on the right side. 67 positive reactions were concordat on both sides (64 with the same score of intensity and .1 with a different score). 3 positive reactions (in 2 patients) on the left side were negative on the right side (1 TMTD. I cobalt chloride and t wool alcohols). At rechallenge 6 weeks later, they were positive. No explanation could be found for negative results at the first challenge on one side and not on the other.
