摘要:
Since the first demonstration of skin antibodies in sera of pemphigus patients by IF (Beutner&Jordon 1964), the scope of skin diseases in which immunologic findings may be of interest has enormously enlarged and dermatology remains one of the leading clinical specialities applying IHC in both diagnosis and research. Immunohistological techniques and, to a lesser degree, immunocytological techniques, have been developed to include not only IF, but also immunoenzymatic methods. Even if not all immunological findings may be disease specific and diagnostic, immunohistological techniques have become a very useful tool for studying skin biopsies in connection with serological methods. Their importance could be increased with rapid development to a new field: the study of normal skin constituents; e.g. Ag, collagen, keratin, with the help of specific polyclonal and monoclonal antibodies.

摘要:
A prerequisite for using flow cytometry (FCM) is the availability of isolated single cells. Procedures for separation and isolation of keratinocytes from animals and man are available, and the resulting single cell suspensions have been subjected to FCM measurements. The major advantage of the method is the accuracy and speed with which a variety of cellular constituents can be quantified. FCM of keratinocytes has, hitherto, been mainly confined to measurements of nuclear DNA for estimation of cell‐cycle distributions and for ploidy studies.In mouse epidermis, cell‐cycle distributions were estimated from sequentially obtained DNA histograms and evaluated with other cell kinetic measurements, resulting in new information about epidermal cell‐cycle progression, not achievable by any of the methods alone. The best way, therefore, to increase our knowledge of keratinocyte proliferation, is the combined use of DNA FCM and other cell kinetic methods.DNA FCM has also been applied to healthy and diseased human epidermis, and may add valuable information to the classification of skin disease in selected cases. It is believed that further progress in the characterization of keratinocyte growth and development will depend on parameters other than DNA

Double layer immunocytochemical procedures were performed on fresh frozen serial sections of lichen planus (LP), using three monoclonal antibodies: OKT3 (T‐cells) and two anti Ia‐like antigens, monoclonal antibodies. Lymphocytes in dermal LP infiltrates were OKT3 positive and Ia‐like positive; lymphocytes in the paracortical area of human normal lymph‐node were OKT3 positive and Ia‐like negative. Unstimulated peripheral blood‐OKT3 positive, E‐rosette forming cells of normal donors were Ia‐like negative, while the same T‐cells, when mitogen‐stimulated, became Ia‐like positive. Therefore, in dermal infiltrates of LP, T‐lymphocytes are Ia‐like positive cells, representing an antigen‐stimulated T‐cell population. Three cases of trichilemmal keratosis (horn) were light microscopically examined and all showed numbers of U‐ or V‐shaped epidermal proliferations which keratinized in a fashion either identical or similar to trichilemmal keratinization. Electron microscopy revealed both uneven and linear borders between the keratinized and the keratinizing cells with a few keratohyalin droplets, remnants of desmosomes, no marginal band in the horny layer, perinuclear vacuolation, few spherical bodies in the intercellular spaces (ICS) of the upper epidermis, and widening of the ICS of the lower epidermis. A number of electron dense spherical particles. 40‐50 nm in diameter, were observed in nuclei of the upper epidermis. 126 basal cell carcinomas from 7 patients with nevoid basal cell carcinoma syndrome were examined histologically and ‐using a scoring system ‐ compared with 54 solitary basal cell carcinomas. The solid subtype formed 78.9% of all tumors in the head and neck region and 48.9% of those located elsewhere. A broader spectrum of histological subtypes was noticed in tumors from patients with the syndrome.
