The human hematopoietic progenitor cell antigen is known as CD34. This antigen is present on normal bone marrow progenitor cells and vaseular endothelial cells. We used the monoclonal antibody auti‐CD34 and immunoperoxidase staining techniques to evaluate the expression of CD34 in benign and malignant vascular and spindle cell tumors. All of the 42 vascular lesions, except two of three lesions of intravascular papillary endothelial hyperplasia, demonstrated diffuse membraneous staining of moderate lo strong intensity of their endothelial cells. Also, normal placentas (five) showed similar staining. All neurofibromas (12), three of five neuromas, and one of four neurilemmomas revealed moderate to strong, diffuse, membraneous staining. Five of eight piloleiomyomas, two of seven angiolciomyomas, and one of five uterine leiomyomas showed focal to dilluse, and weak to moderate, membraneous staining in the smooth muscle component. Six dermatofibrosarcoma protuberans were studied: generalized, strongly positive membraneous staining was present in four. All specimens showed staining of the normal endothelial cells and the cells surrounding the hair follicles (bulge area), sebaceous glands, and eccrine glands. No staining was demonstrated in any of the following fibrohistiocytic tumors: atypical fibroxanthomas (two), fibrous dermatofibromas (23), giant cell tumor of tendon sheath (one), and hemosiderotic dermatofibromas (18). Melanocytic tumors (Spitz nevi (three) and spindle cell superficial spreading malignant melanoma (one)], Merkel cell carcinomas (six), and spindle cell squamous cell carcinomas (two) did not stain with anti‐CD34. We examine the effects of the recombinant epidermal cytokines interleukin 1α (IL‐1α), interleukin 1β (IL‐1β), interleukin 3 (IL‐3), interleukin 6 (IL‐6), granulocyte‐macrophage colony stimulating factor (GM‐CSF), macrophage colony stimulating factor (M‐CSF), tumor necrosis factor alpha (TNF), and the effect of the lymphokine gamma interferon (γ‐IFN) on the expression of ICAM‐1 in short term organ cultures of newborn human foreskins. In normal skin, ICAM‐1 was detected immunohistochemically exclusively on endothelial cells. In addition to enhanced ICAM‐1 expression by endothelial cells (at 1 h) and keratinocytes (by 6 h) exposed to γ‐IFN, increased endothelial cell expression also resulted at 24 h after exposure to IL‐6 and GM‐CSF and at 48 h to M‐CSF. Dermal dendritic cell reactivity for ICAM‐1 was observed at 24 h after supplementation with IL‐1α, IL‐1β and IL‐6, and at 48 h with GM‐CSF and M‐CSF. Incubation with culture medium alone or with IL‐3 resulted in no change in baseline ICAM‐1 expression on any cell type, and incubation with TNF resulted in enhanced ICAM‐1 expression only by endothelial cells. Granulomatous reactions at sites of previous cutaneous herpes zosier lesions occur, but their etiology is not known. Three tissue specimens from 5 cases identified clinically and hislologically as post‐zosteric granulomatous reactions were studied for the presence of varicella‐zoster virus (VZV) deoxyribonucleic acid (DNA) by the polymerase chain reaction using specific primers For VZV. VZV DNA was detected in 1 of 3 cases where the granulomatous reaction occurred immediately in the wake of resolving vesicular herpes zoster lesions. Finding viral DNA in earlier reactions probably represents residue from the active herpetic process. VZV DNA was not identified in granulomatous reactions arising between 1 month and up to 4 years after resolved herpes zoster. The negative result in these cases supports the hypothesis that there is no association between persistence of VZV DNA and granuloma formation. Palisaded, encapsulated neuroma (PEN) has been described as a predominantly solitary, nodular tumor; plexiform or multinodular growth patterns have not been reported in detail. We reviewed 55 PEN in order to evaluate: 1) the predominant growth patterns: 2) features that may indicate previous trauma; and 3) association with acne‐like changes. Sixteen cases (29%) had growth patterns other than the solitary, nodular type. These patterns were: a) plexiform (7/16), b) multinodular (5/16), and c) fungating (4/16). Other unusual features included evidence of trauma, i.e., fibrosis, myxoid change, chronic inflammation (5/16), partially incomplete capsule (6/16), and association with acne‐like features (13/16). These results suggest that: 1) plexiform or multinodular growth patterns occur relatively often in PEN; 2) some changes indicate a possible histogenetic relation to traumatic neuroma, probably via secondary traumatization; 3) a high frequency of association with acne‐like changes which could imply, but does not prove, an acne‐induced histogenesis; and 4) the differential diagnosis of cutaneous plexiform lesions should incl

Nerve growth factor, S‐100 protein, CD44, and CD34 have unique expressions in or surrounding eccrine coil hut are not found in eccrine duct or apoerine gland. We studied 15 cases of cutaneous cylindroma to see if these antigens are found in this neoplasm. All were found in cylindroma to varying degrees. These results link the histogenesis of cylindroma to the eccrine coil. A unique feature of cylindroma is the large number of prominent dendritic cells most likely representing Langerhans cells that permeate the tumor aggregate

Clinical and hislological data from 8 trichilemmal carcinomas are reviewed. The tumors occurred mainly on sun‐exposed skin of the face of elderly people as small solitary, frequently ulcerated nodules. Histologically, they consisted of multilobulate, infiltrative growths, connected to the epidermis and piloscbaceous structures and showing features reminiscent of the outer root sheath of the hair follicle. The presence of lobules of clear, glycogen‐rich cells with peripheral palisading, hyaline basement membranes, trichilemmal keratinization, and abortive follicular sheaths and the absence of ductal or acinar differentiation allow distinction from other clear cell tumors of the skin. Prediction of long‐term survival for clinical stage I malignant melanoma and guidance of therapy has long relied upon assessment of tumor thickness. This parameter is a strong, but not infallible predictor of prognosis. Clark et al. have developed a prognostic model based upon the concept of tumor progression and the evaluation of six readily assessable clinical and histologic attributes. They report the most accurate prediction of long‐term survival of any prognostic method available for melanoma (100% accurate for radial‐growth‐phase and 84.1% accurate for vertical‐growth‐phase melanomas). This model was developed and validated via study of a patient population which largely resided in the northeastern portion of the United States. We report additional validation of this model using a data base of 55 patients from a different geographic location (North Carolina) and have observed virtual identity in the accuracy of predicting 8‐year survival for radial‐growth‐phase (100%) and vertical‐growth‐phase (85.1%) melanomas. The distribution of the lysosomal enzymes cathepsin B, lysozyme, ehymotrypsin, and neutrophil elastase was examined in eccrine, apocrine, and sebaceous glands using a postembedding immunogold labeling procedure. Various amounts of cathepsin B were detected in all glands. Lysozyme, however, was detected in apocrine glands only. The other two lysosomal enzymes Were not detectable immunologically. In apocrine and eccrine glands, anti‐cathepsin B antibody labeled all secretory granules. In sebaceous glands, only the peripheral layer of cells showed immunological activity for cathepsin B. In apocrine glands, granules containing remnants of cristae were more intensively labeled than those lacking cristae which supports the assumption that both granules are derived from mitochondria by acquiring lysosomal enzymes. The enzymes convert mitochondria to granules with cristae and finally to granules without cristae. Squamous syringometaplasia (SS) is defined as a mature squamous metaplasia of the eccrine ducts. We prospectively evaluated its frequency and clinical and pathological features in a series of 21 patients. SS was found in association with several diseases, chiefly chronic ulcers. The patients'ages ranged between 20 and 80 years and there was no sex predominance. The involved eccrine ducts were usually located in the superficial and mid dermis and displayed inner keratinization. The stimulus required for SS is unknown.
