Tumor promotion by regenerative epidermal hyperplasia in mouse skin |

Chemically induced epidermal carcinogenesis is usually divided into two stages: initiation, which involves the conversion of epidermal cells into latent neoplastic cells; and promotion, which allows the expression of this neoplastic change. One of the characteristics of promotion is the transformation of the normal epidermis into a hyperplastic epidermis.One of the unanswered questions about epidermal tumor promotion is whether the epidermal hyperplasia that characterizes promoted skin is simply a regenerative epidermal hyperplasia resulting from damage produced by the promoter. The opinion currently held is that the epidermal hyperplasia produced by tumor promoters is not simply a regenerative epidermal hyperplasia and possesses characteristics which a regenerative hyperplasia does not have, enabling it to evolve into an epidermal neoplasm.The purpose of this review is to present recent evidence which strongly suggests that promoter induced hyperplasia is a regenerative hyperplasia. Two principal lines of evidence arc reviewed. The first demonstrates that an epidermal regenerative hyperplasia repeatedly produced by wounding or abrasion can promote epidermal carcinogenesis in the initiated skin of mice. The second line of evidence demonstrates that the epidermal hyperplasia produced by the application of 12‐O‐tetradccanoyl‐phorbol‐13‐acetate (TPA), the most powerful and widely used promoter of skin carcinogenesis, is preceded by damage to the epidermis. This strongly suggests that the epidermal hyperplasia which ensues is a regenerative hyperplasia.The review concludes that during epidermal tumor promotion there is a chronic regenerative epidermal hyperplasia, and that there is no evidence for any other necessary factors during tumor promotion in appropriately initiated m

摘要:
A technique for primary plastic embedding for light microscopy was utilized in evaluating plaque lesions in four patients with mycosis fungoides and compared to tissues from the same patients embedded in paraffin. Standard 3 mm punch biopsies were utilized. Specimens were fixed in B‐5 and embedded in araldite epoxy resin. In specimens from all four patients, the cytological details of the mycosis cells were superior in the plastic embedded tissues. This practical and relatively inexpensive method may prove to be a sensitive indicator of mycosis cells in early lesions. 摘要:
The skin of a female Egyptian mummy, embalmed 100–300 A.D., was studied by transmission and scanning electron microscopy.After rehydration with cacodylate buffer containing sucrose, the ordinary technique for transmission electron microscopy was performed on skin of the right shoulder. In epidermal cells, mitochondria and their cristae were identified. Between epidermal cells, desmosomes, half the normal size, were found. In the dermis, collagen bundles and elastic fibres were observed. The collagen fibres had a diameter of 67–91 nm, and the band pattern showed 10–12 sub‐bands per one period of 55–61 nm length. The elastic fibres had less amorphous substance than normal and a fibrillar beaded pattern. The dermal components of the skin were well preserved in spite of the mummification.Throughout the dermis and the subcutaneous fatty tissue, a number of bacterial spores were found showing a characteristic appearance.Scanning electron microscopy of hairs showed a layer of the preserving material surrounding hairs, and under it a normal cuticle

摘要:
Murine keratinocytes, isolated by flotation trypsinization of skin, can be separated into five groups by centrifugation through Percoll, a colloidal silica gradient. Within each group a good correlation was found between density, plating efficiency, morphological appearance, DNA synthesis, and degree of keratinization/cornification. This method can be applied equally well to fetal, newborn, or adult keratinocytes and should be useful in a variety of studies including isolation of subpopulations of pathological cell types, work on chalones and hyperplastic diseases such as psoriasis, andin vitrotransformation studies.
