The frequency of spermatozoa bearing one or two fluorescent bodies (FB) has been studied in two series of semen, normal and abnormal (asthenozoospermic). The percentage of spermatozoa with one FB is lower in abnormal semen (41.19% vs 45.82%). On the other hand, there is no significant difference when spermatozoa bearing two FB are compared (1.95% vs 1.90%). Various correlations among semen characteristics were studied, including sperm count, mobility, vitality, percentage of normal forms. Two correlations were found to be significant: positive correlation between the frequency of one FB spermatozoa and mobility and a positive correlation between the frequency of normal forms and the frequency of one FB spermatozoa. The differences between normal and abnormal semen involve only one FB spermatozoa and not those with two FB.

A new index by which sperm longevity can be evaluated quantitatively is suggested for a routine semen analysis, and was calculated in seminal specimens of 50 donors. This index of longevity is defined according to each one of 2 different characteristics of sperm motility analyzed by the MEP method:ILM — the ratio between per cent of motile sperm measured four h and one h after ejaculation.ILV — the ratio between sperm velocity measured four and one h after ejaculation.The ratio clearly shows whether motility, testedin vitro, is stable or drops rapidly with time.ILM was found to range between 0.2 and 1.3 with average 0.8. ILV ranged from 0.5 to 1.8 and average 1.14. Neither ILM nor ILV showed any correlation with sperm concentration, abnormal forms, initial per cent of motility or initial sperm velocity and thus it was impossible to predict sperm longevity from any known sperm characteristics. However. a coefficient of correlation of 0.44 was found between ILM and ILV which means that to a certain extent, cases with well‐preserved percentage of motility may also reveal a well‐preserved sperm velocity.With the aid of this index it is possible to investigate if there is any correlation between sperm longevity and fertility rate, sperm survival in various media, alleged sperm stimulants or treatment given to t

A technique by which motile, non motile live (NML) and dead human sperm can be differentiated simultaneously is described. In this technique samples from human semen are mixed with aqueous solutions of eosin Y, and then colour slide photomicrographs with the aid of the MEP method are taken. By analyzing these projected slides, objective and accurate information about number and ratio between motile NML and dead sperms can be obtained as well as average and frequency distribution of sperm velocities. Data from experiments performed with this technique showed that: 1. There is no significant difference in results when using eosin concentrations between 0.5 and 2 per cent. 2. There is almost no difference results when incubation times were from one to five min. 3. Analyzing 16 specimens from normal and infertile men revealed that an average ratio of NML/dead sperms is about 2, which is much above figures obtained by other authors. Some suggested explanations for this difference are discussed.

Seminal fluid levels of hCG‐like material are determined by a radio‐immunoassay using an antibody against the β‐chain of hCG. In 267 unselected cases a significant positive correlation to sperm count and a negative correlation to the percentage of abnormal spermatozoa is found. An androgen binding protein (ABP) has been demonstrated in testis and epididymis (caput, corpus, cauda) supernatants of sheep aged 50, 120 and 200 days. This protein has a high affinity for 5α‐dihydrotestosterone (Ka = 0.184 ± 0.023 × 109M‐1), an electrophoretic mobility (RF) of 0.52–0.56 on PAGE and a molecular weight of about 70 000 daltons. In 50 day‐old animals (infantile) ABP is only present in the testis (392 fmoles/mg protein) with no dihydrotestosterone binding detectable in the epididymis. This is the first evidence of production of ABP by the testis some time before initiation of spermatogenesis. In the 120 day‐old animals (prepubertal), ABP is observed both in testis (142 fmoles/mg) and epididymis: caput (90 fmoles/mg), corpus (41 fmoles/mg) and cauda (165 fmoles mg). The distribution of [3H] carnitine in the epididymis has been studied by autoradiography 93 hours after injection of [3H]deoxycarnitine into intact, hemicastrated or hypophysectomized rats. In the intact animals, radioactivity is concentrated in the lumen of the caput and corpus with only a small amount observed in the caudal portion of the epididymis. In the hemicastrated rat, devoid of spermatozoa, the amount of radioactivity in the lumen of the caput and the cauda portions are lower than in the intact rats but still greater compared to the epididymal cells. In hypophysectomized rats the luminal labelling is much reduced. In contrast to the epididymis, the amount of radioactivity is low and evenly distributed in the heart from intact, hemicastrated and hypophysectomized rats and in the pituitary from intact and hemicastrated animals. The great reduction in luminal labelling after hypophysectomy emphasizes the hormonal dependence of the carnitine concentrating mechanism. These findings indicate that carnitine initially is concentrated in the caput and corpus portion of the epididymis. This intraluminal carnitine is subsequently transported into the caudal segment.The evidence strongly supports the existence of a hormonal dependent carnitine concentrating mechanism localized on the luminal side of the epithelium in epididymis.

The sensitivity (dose of hormone eliciting half maximal response) and the magnitude of the maximal response of Leydig cell suspensions prepared from immature intact and immature eight day hypophysectomized rats to hCG and dibutyryl‐cyclic‐3'5‐adenosine monophosphate (dBC) were assessedin vitroin the presence or absence of the phosphodiesterase inhibitor, methylisobutyl‐xanthine (MIX). In separate studies LH/hCG binding to dispersed Leydig cells from intact and hypophysectomized rats of the same age was evaluated by Scatchard analysis and autoradiography.Hypophysectomy was associated with a 3–4 fold increase in the sensitivity of response, but an 80 % reduction in the maximum steroidogenic response to both hCG and dBC (when expressed per million 3β‐hydroxy‐steroid‐dehydrogenase (3β‐HSD) positive cells. Addition of MIX resulted in a greater increase in the sensitivity of response from intact Leydig cells than from cells from hypophysectomized rats, suggesting that hypophysectomy is associated with a decrease in the activity of the phosphodiesterase. Scatchard analysis of [125I]hLH binding to Leydig cells indicated no major decrease in the average number of LH/hCG receptors per cell after 8 days of hypophysectomy. Testosterone and dihydrotestosterone were measured in spermatic venous blood of 37 azoospermic subjects. On the basis of testicular biopsy and vesiculodeferentography the patients were divided into three groups, i.e. subjects affected by azoospermia of obstructive origin with normal testicular histology, subjects with germinal cell arrest and those with Sertoli cell only syndrome (n = 6, n = 25 and n = 6 respectively).The mean values (± SE) of hormone concentrations in spermatic venous blood in the three groups were respectively 2312 ± 517 nM/l. 2292 ± 320 nM/l and 2006 ± 342 nM/l for testosterone and 28.8 ± 7.7 nM/l, 39.0 ± 5.9 nM/l and 41.9 ± 17.7 nM/l for dihydrotestosterone.
